The Role of Granulocyte Colony-Stimulating Factor in Endometrial Preparation for Embryo Implantation in In Vitro Fertilization.

Granulocyte colony-stimulating factor (G-CSF) has been suggested as a supplementary approach for endometrial preparation in IVF. Clinical results continue to be inconsistent. This narrative review synthesises molecular and clinical information to elucidate the function of G-CSF in modifying endometrial receptivity and to identify patient categories most likely to benefit. A thorough assessment was conducted on published research on G-CSF administration in women with treatment-resistant thin endometrium, recurrent implantation failure, and unselected IVF populations. The research demonstrates t

The Role of Immature Granulocyte Count and Percentage as Novel Indicators of Systemic Inflammation in Plaque Psoriasis.

Background/Objective: Psoriasis is a chronic immune-mediated inflammatory disease with systemic involvement. Immature granulocytes (IGs) are potential inflammatory markers, but their role in psoriasis is unclear. We evaluated immature granulocyte count (IG#) and percentage (IG%) in plaque psoriasis, their associations with disease severity and treatment response, and their ability to distinguish patients from healthy controls. Methods: This retrospective study included 85 adults with plaque psoriasis and 85 healthy controls. Severity was assessed by the Psoriasis Area and Severity Index (PASI)…

Protective effect of granulocyte colony stimulating factor on cyclophosphamide-induced prostate injury in rats.

Cyclophosphamide (CPA) is a commonly used chemotherapeutic agent whose metabolites readily accumulate in male reproductive organs such as the testes and epididymis, exhibiting reproductive toxicity. Granulocyte colony-stimulating factor (G-CSF) possesses anti-inflammatory and antioxidant effects, but its protective mechanism against CPA-induced prostate injury remains unclear. This study aimed to investigate the protective effect of G-CSF against CPA-induced prostate injury in rats. Thirty SPF male Sprague-Dawley (SD) rats were randomly divided into a control group (C group), a CPA model group

Semaglutide-associated agranulocytosis requiring hospitalisation and granulocyte colony-stimulating factor: a case report.

Semaglutide, a glucagon-like peptide-1 receptor agonist (GLP-1RA), is widely used in obesity and type 2 diabetes. Although generally well tolerated, severe haematological adverse reactions remain poorly characterised. A 48-year-old woman treated with subcutaneous semaglutide for obesity developed febrile agranulocytosis with an absolute neutrophil count of 0.0×10⁹/L after several months of therapy. Baseline blood counts had been normal prior to treatment. Bone marrow aspirate and biopsy showed preserved cellularity with granulocytic maturation arrest at the promyelocyte/myelocyte stage. Semagl

Granulocyte apheresis in donors: A comparative study of spectra optia and cobe spectra apheresis systems.

BACKGROUND: Granulocyte transfusion (GTX) is a significant adjunctive treatment for patients with refractory infections and severe neutropenia. High-dose granulocyte concentrates are best collected by optimized mobilization and apheresis techniques. Among the most popular platforms, COBE Spectra and Spectra Optia differ in terms of technology and operational characteristics; however, comparative data from Indian settings are limited. METHODS: This retrospective study evaluated 316 granulocyte apheresis sessions conducted between 2013 and 2022 in a tertiary care centre in Kerala, India. G-CSF

Granulocytic sarcoma as an initial manifestation of acute promyelocytic leukemia: A case report with literature review.

RATIONALE: Granulocytic sarcoma (GS) is a rare tumor consisting of myeloid blasts with or without maturation and occurs in sites other than the bone marrow. Due to its low prevalence, clinical cases and pathogenesis need to be studied. Therefore, we present a rare case of humerus GS occurring simultaneously with acute promyelocytic leukemia (APL) and studied a retrospective analysis of clinical characteristics and related treatment strategies, hoping that it could help to standardize the early diagnosis and treatment of APL/GS. PATIENT CONCERNS: We present a case of humerus GS complicated wit

Granulocyte colony-stimulating factor (G-CSF)-induced aortitis in a woman with breast cancer.

Granulocyte colony-stimulating factor (G-CSF) is used alongside cytotoxic chemotherapy regimens to reduce the severity and duration of neutropenia and decrease infection risk. Here, we add to the growing body of evidence that aortitis can be considered a recognised side effect of G-CSF therapy.A woman in her 60s with breast carcinoma treated with G-CSF cover alongside adjuvant chemotherapy presented with intermittent fever and epigastric pain. There were no localising signs of infection. Results demonstrated raised inflammatory markers. The patient did not respond to antibiotics, and imaging s

Neuroprotective effects of granulocyte colony-stimulating factor against tramadol-induced cerebellar neurotoxicity.

BACKGROUND: Tramadol (TRM) is a centrally acting synthetic opioid and serotonin/norepinephrine reuptake inhibitor. Despite being a potent painkiller, long-term use can induce permanent neurotoxicity. Granulocyte colony-stimulating factor (G-CSF) is a cytokine that helps to mobilize stem cells and facilitate their integration over injured neurons. AIM: This work aims to study the histopathological, biochemical, and molecular alterations in the cerebellar cortex induced by TRM in comparison to the postulated protective effect of G-CSF versus TRM withdrawal. METHODS: 32 adult male albino rats w

Granulocyte Colony-Stimulating-Factor-Producing Bladder Carcinoma: A Case Report and Literature Review.

Granulocyte colony-stimulating factor (G-CSF)-producing bladder carcinoma has an aggressive clinical course. We report a case of G-CSF-producing bladder carcinoma in a 78-year-old Japanese man who had a bladder tumor with a diameter of 38 mm. Transurethral resection of the bladder tumor was performed. Pathological examination revealed a high-grade muscle-invasive urothelial carcinoma (pT2). The patient had three courses of neoadjuvant chemotherapy with a combination of gemcitabine and carboplatin and thereafter underwent robot-assisted radical cystectomy. The surgically resected bladder specim

Distinct osteoclastogenic potential of granulocyte-macrophage colony-stimulating factor-induced monocyte subsets.

BACKGROUND: Rheumatoid arthritis (RA) is a chronic autoimmune disease characterized by synovial inflammation and progressive bone destruction. Although osteoclasts mediate bone resorption in RA, recent evidence suggests that inflammatory osteoclasts differ from physiological osteoclasts in various aspects, including their progenitor origins. This study aimed to compare the osteoclastogenic potential of monocyte-derived dendritic cells (moDCs) and other granulocyte-macrophage colony-stimulating factor (GM-CSF)-induced cells to identify potential progenitor populations involved in inflammatory b

ATRA-induced NEAT1 upregulation promotes autophagy during APL cell granulocytic differentiation.

AIMS: Acute promyelocytic leukemia (APL) progresses quickly and often leads to early hemorrhagic death. Treatment with all-trans retinoic acid (ATRA) promotes differentiation of APL cells and clinical remission, making APL a potentially curable malignancy. Understanding how ATRA works may lead to new treatments for other types of leukemia. Long non-coding RNA NEAT1 has been implicated in the differentiation of APL cells. This study aims to elucidate the specific role of NEAT1 in the granulocytic differentiation of APL. METHODS: The influence of NEAT1 on autophagy and PML/RARα degradation was

GLUT1 mediates the release of HMGB1 from airway epithelial cells in mixed granulocytic asthma.

Asthma is quite heterogenous and can be categorized as eosinophilic, mixed granulocytic (presence of both eosinophils and neutrophils in the airways) and neutrophilic. Clinically, mixed granulocytic asthma (MGA) often tends to be severe and requires large doses of corticosteroids. High mobility group box 1 (HMGB1) is one of the epithelium-derived alarmins that contributes to type 2 inflammation and asthma. This study was aimed to investigate the role of glucose transporter 1 (GLUT1) in modulation of airway epithelial HMGB1 production in MGA. Induced sputum and bronchial biopsy specimens were o

Long COVID syndrome in children: neutrophilic granulocyte dysfunction and its correlation with disease severity.

BACKGROUND: Many children suffer from lingering symptoms after COVID-19, known as long COVID syndrome (LCS), otherwise called Post COVID-19 Condition (PCC). Despite extensive research, the prevalence of symptoms, its impact on quality of life, and underlying mechanisms still need to be fully understood. As neutrophilic granulocytes play an essential role in COVID-19, and their prolonged disruption was found to cause immunological diseases, we hypothesized their ongoing disturbed functionality in LCS. METHODS: We studied 129 children with LCS, 32 convalescent children (CG+), and 8 uninfected c…

Mitochondrial CLPB is a pro-survival factor at the onset of granulocytic differentiation of mouse myeloblastic cells.

Loss-of-function mutations in the CLPB gene lead to congenital neutropenia due to impaired neutrophil differentiation. CLPB, a member of the AAA+ family of proteins, resides in the intermembrane space of mitochondria. The mechanism by which a loss of CLPB elicits defects in the differentiation program of neutrophil precursor cells is not understood. Here, we used 32D clone 3 (32Dcl3) cells, an interleukin-3 (IL-3)-dependent mouse myeloblastic cell line model, to investigate the effects of CLPB knockout on myeloblast-to-neutrophil differentiation in vitro. We found that CLPB-deficient 32Dcl3 ce
