Mutagenicity, unscheduled DNA synthesis, and metabolism of 1-nitropyrene in the human hepatoma cell line HepG2.

The cell line HepG2 is derived from a well differentiated human hepatoblastoma, which retains many of the morphological characteristics of liver parenchymal cells. These cells at passages greater than 95 were found to metabolically activate carcinogens to genotoxic metabolites. The addition of 6.8 microM 1-methyl-3-nitro-1-nitrosoguanidine, 5.3 microM 4-nitroquinoline-N-oxide, and 4-20.3 microM 1-nitropyrene resulted in the induction of mutations at the HGPRT locus as determined by 6-thioguanine resistance. This is the first description of the induction of mutations in these cells. Additionall

Chaihu-Guizhi Decoction Attenuates Airway Remodelling in COPD Mice via the TGF-β1/Smads Signalling Pathway: An Integrated Proteomic and Metabolomic Analysis.

Chronic obstructive pulmonary disease (COPD) is characterised by persistent airway injury and remodelling, and the mechanisms by which Chai-Hu-Gui-Zhi Decoction (CGD) acts in COPD remain unclear. This study investigated whether CGD attenuates airway remodelling through the TGF-β1/Smads-EMT-ECM signalling axis. A mouse COPD model was established by cigarette-smoke exposure plus intranasal lipopolysaccharide. After model induction, mice received dexamethasone acetate or low-, medium- or high-dose CGD for 28 days. UHPLC-MS/MS, network pharmacology, molecular docking, molecular dynamics, proteomic

Ginsenoside Rg1 suppresses urate-induced renal tubular ferroptosis through PPARG-dependent antioxidant signaling.

Hyperuricemic nephropathy (HN) is characterized by renal tubular injury, oxidative stress and interstitial remodeling, but the signaling mechanisms linking urate stress to ferroptotic tubular damage remain incompletely defined. Here, we investigated whether Ginsenoside Rg1 protects against hyperuricemia-associated renal injury by modulating PPARG-dependent ferroptosis signaling. In mice challenged with potassium oxonate and hypoxanthine, Rg1 improved renal histopathology and reduced serum uric acid, creatinine, blood urea nitrogen and xanthine oxidase activity. Rg1 also reversed ferroptosis-as

Antisense Dipeptide Repeat Proteins Drive Widescale Purine Metabolism Aberration in C9orf72 Amyotrophic Lateral Sclerosis via ADA.

Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disease characterised by the death of motor neurons leading to paralysis and death, generally 3-5 years post-symptom onset. The most frequent genetic cause of ALS is a hexanucleotide repeat expansion (HRE) in the chromosome 9 open reading frame 72 (C9orf72) gene, that has three major hypothesised pathological mechanisms including the production of dipeptide repeat proteins (DPRs). Our laboratory has previously identified purine metabolism dysfunction in induced neural progenitor cell-derived astrocytes (iAstrocytes) from C9orf72 ALS (C

Systematic evaluation of pre-analytical variables on synovial fluid metabolomic profiles using GC-ToF-MS and UHPLC-MS.

Synovial fluid is a clinically valuable biofluid for studying joint diseases through metabolomic analysis. However, given the viscoelastic nature of this biofluid its analysis is challenging. Indeed, the lack of standardised pre-analytical processing protocols for synovial fluid metabolomics potentially introduces significant variability that can compromise data reliability and hinder the application/interpretation of results. This study systematically assesses how common sample handling variables including sample dilution, freeze-thaw cycling, blood staining and viscosity reduction (via hyalu

Brassica rapa L. (Turnip) Ethanol Extract Alleviates Hyperuricemia by Modulating Purine Metabolism Enzymes and Renal Urate Transporters.

Hyperuricemia (HUA) is a metabolic disorder caused by excessive uric acid (UA) production and/or insufficient UA excretion. Brassica rapa L. (turnip) is a traditional medicinal and edible plant in northwestern China. In this study, the urate-lowering effects of B. rapa L. ethanol extract (BrEE) and associated underlying mechanisms were investigated using a hypoxanthine/potassium oxonate-induced HUA mouse model and UA-stimulated HK-2 cells. BrEE was prepared and characterized; quercetin-3-glucuronide was the most abundant compound in BrEE, with a relative abundance of 22.04%. BrEE significantly

Sodium cyclamate aggravates DSS-induced colitis in mice with associated microbial, metabolic, and epithelial alterations.

Sodium cyclamate is widely used as a non-nutritive sweetener, but its effects during intestinal inflammation remain poorly defined. We investigated whether chronic cyclamate exposure aggravated dextran sulfate sodium (DSS)-induced colitis in mice using disease phenotyping, 16S rRNA gene sequencing, targeted short-chain fatty acid and bile acid measurements, metabolomics, lipidomics, discovery proteomics, and independent western blot validation. Relative to DSS alone, DSS + Cyclamate mice exhibited greater disease severity and higher abundances of Bacteroides and Klebsiella; Lactobacillus and M

The Current Applications of Metabolomics in Understanding Endometriosis: A Systematic Review.

Endometriosis is a common gynecological disease that affects approximately 10-15% of reproductive-aged women worldwide. This debilitating disease has a negative impact on the quality of life of those affected. Despite this condition being very common, the pathogenesis is not well understood. Metabolomics is the study of the array of low-weight metabolites in a given sample. This emerging field of omics-based science has proved to be effective at furthering the understanding of endometriosis. In this systematic review, we seek to provide an overview of the application of metabolomics in endomet

Mechanistic insights into metformin's anti-hyperuricemic effect: Targeting PPP/DNPB/XOD-mediated purine pathway, purinosome assembly, and gut microbiota homostasis in rats.

Hyperuricemia has become a public threat to human health, and conventional medical treatment only aims to inhibit xanthine oxidase (XOD). Endogenous purine biosynthesis and purinosome formation are neglected in research of medical mechanism. In this study, the therapeutic effect and mechanism of metformin was explored in chronic high-fructose-induced hyperuricemic rats. Results indicated that four weeks of metformin administration effectively reduced uric acid (UA), creatinine, and urea levels, ameliorated renal and hepatic injuries, and promoted glycogen synthesis in hyperuricemic rats. Prolonged Fructose Overconsumption Activates Original Biosynthetic and Metabolic Pathways of Endogenous Purine in Rats.

Fructose is a nutrient used widely and indiscriminately as a sweetener in various types of foods and beverages. Accumulating evidence suggests that dietary fructose has a close relationship with metabolic diseases like hyperuricemia (HUA). However, the role and metabolic fate of fructose intake remain elusively obscure. The present work made a pioneering endeavor to unravel the potential mechanistic link between prolonged fructose overconsumption and the incidence of HUA. High-fructose (30%) free drinking was provided to rats for 17 weeks. Results indicated that long-term high-fructose intake

Xanthine Oxidase-Dependent Activation of NLPR3 Inflammasome in Epithelial Cells Sustains Inflammation in Inflammatory Bowel Disease.

IMPORTANCE AND OBJECTIVE: Xanthine oxidase (XO) plays a key role in purine metabolism, catalyzing the oxidation of hypoxanthine to xanthine, and xanthine to uric acid (UA) with the production of superoxide anions. The accumulation of UA has been shown to initiate the inflammatory process through NLRP3 inflammasome, and the production of reactive oxygen species (ROS) contributes to inflammation-related tissue damage in different organs. This study aimed to investigate the role of XO in the pathogenesis of Inflammatory Bowel Disease (IBD) and the anti-inflammatory potential of XO inhibition. DE

Effects of WN1703 on Cardiovascular Function in Chronic Hyperuricemia Rats and Myocardial Injury Mechanism Exploration in H9C2 Cells.

Hyperuricemia, a prevalent condition, is typically preceded by disturbances in purine metabolism and is frequently associated with hyperlipidemia and other dysfunctions of metabolism. WN1703 demonstrated an inhibitory activity against xanthine oxidoreductase (XOR) that was comparable to febuxostat in our prior investigation. In this study, we assessed the cardiovascular safety of WN1703 in a chronic hyperuricemia rat model induced by potassium oxonate in combination with hypoxanthine. We investigated the changes in cardiovascular biomarkers in chronic hyperuricemia rats treated with febuxostat
