Toll-like receptor 2 mediates early inflammation by leptospiral outer membrane proteins in proximal tubule cells.

Tubulointerstitial nephritis is a cardinal renal manifestation in leptospirosis and LipL32, the major lipoprotein component of leptospiral outer membrane proteins (OMPs), induces a robust inflammatory response in cultured renal proximal tubule cells through a nuclear factor-kappaB-related pathway. Here, we investigated whether Toll-like receptor (TLR), known to play a pivotal role in innate immunity, could mediate the inflammatory response induced by leptospiral OMPs in renal proximal tubule cells. TLR expression was analyzed by flow cytometry and indirect immunofluorescence in cultured mouse

Exposure to continuous darkness ameliorates gastric and colonic inflammation in the rat: both receptor and non-receptor-mediated processes.

BACKGROUND AND AIM: Melatonin is a hormone involved in the transduction of photoperiodic information, and appears to modulate a variety of neural and endocrine functions. The present study was designed to determine the impact of continuous darkness (CD) on acute gastric and colonic inflammation and the involvement of melatonin receptors in the darkness-related alterations in oxidant gut injury. METHODS: Rats were housed either in CD or in standardized light/dark (12/12 h) cycles for 15 days before the induction of colitis or gastric ulcer. Luzindole (MT(2) receptor antagonist) was given at a

Runx3 regulates mouse TGF-beta-mediated dendritic cell function and its absence results in airway inflammation.

Runx3 transcription factor regulates cell lineage decisions in thymopoiesis and neurogenesis. Here we report that Runx3 knockout (KO) mice develop spontaneous eosinophilic lung inflammation associated with airway remodeling and mucus hypersecretion. Runx3 is specifically expressed in mature dendritic cells (DC) and mediates their response to TGF-beta. In the absence of Runx3, DC become insensitive to TGF-beta-induced maturation inhibition, and TGF-beta-dependent Langerhans cell development is impaired. Maturation of Runx3 KO DC is accelerated and accompanied by increased efficacy to stimulate

Transcription factor Nrf2 regulates inflammation by mediating the effect of 15-deoxy-Delta(12,14)-prostaglandin j(2).

Activated macrophages express high levels of Nrf2, a transcription factor that positively regulates the gene expression of antioxidant and detoxication enzymes. In this study, we examined how Nrf2 contributes to the anti-inflammatory process. As a model system of acute inflammation, we administered carrageenan to induce pleurisy and found that in Nrf2-deficient mice, tissue invasion by neutrophils persisted during inflammation and the recruitment of macrophages was delayed. Using an antibody against 15-deoxy-Delta(12,14)-prostaglandin J(2) (15d-PGJ(2)), it was observed that macrophages from pl

Role of CD4(+) CD25(+) regulatory T cells in T helper 2 cell-mediated allergic inflammation in the airways.

It has recently been shown that CD4(+) CD25(+) T cells are immunoregulatory T cells that prevent CD4(+) T cell-mediated organ-specific autoimmune diseases. To determine whether CD4(+) CD25(+) T cells downregulate Th2 cell-mediated allergic inflammation in the airways, we studied antigen-induced eosinophil recruitment in the airways in BALB/c Rag-2(-)(/-) mice transferred with CD4(+) CD25(+) T cell-depleted or unfractionated T cells from ovalbumin-specific TCR transgenic mice. Antigen-induced eosinophil recruitment into the airways was significantly decreased in the mice transferred with CD4(+)

Environmental particulate-mediated cytokine production in lung epithelial cells (A549): role of preexisting inflammation and oxidant stress.

Epidemiologic data show that air pollution particulates cause adverse pulmonary health effects, especially in individuals with preexisting lung disease. We sought to model in vitro preexisting lung inflammation in order to investigate the hypothesis that "primed" lung epithelial cells will exhibit enhanced phlogistic responses [e.g., interleukin-8 (IL-8) production] to particulate air pollution. Exposure of tumor necrosis factor alpha (TNF-alpha) primed or control A549 cells to the air pollution particulates, residual oil fly ash (ROFA), and the known pathogenic dust alpha-quartz, but not iner

E-series resolvin metabolome, biosynthesis and critical role of stereochemistry of specialized pro-resolving mediators (SPMs) in inflammation-resolution: Preparing SPMs for long COVID-19, human clinical trials, and targeted precision nutrition.

The COVID-19 pandemic has raised international awareness of the importance of rigorous scientific evidence and the havoc caused by uncontrolled excessive inflammation. Here we consider the evidence on whether the specialized pro-resolving mediators (SPMs) are ready to meet this challenge as well as targeted metabololipidomics of the resolution-inflammation metabolomes. Specific stereochemical mechanisms in the biosynthesis of SPMs from omega-3 essential fatty acids give rise to unique local-acting lipid mediators. SPMs possess stereochemically defined potent bioactive structures that are high-

Nitrogen-containing bisphosphonates and lipopolysaccharide mutually augment inflammation via adenosine triphosphate (ATP)-mediated and interleukin 1β (IL-1β)-mediated production of neutrophil extracellular traps (NETs).

Among the bisphosphonates (BPs), nitrogen-containing BPs (N-BPs) have much stronger anti-bone-resorptive actions than non-N-BPs. However, N-BPs have various side effects such as acute influenza-like reactions after their initial administration and osteonecrosis of the jawbones after repeated administration. The mechanisms underlying such effects remain unclear. To overcome these problems, it is important to profile the inflammatory nature of N-BPs. Here, we analyzed the inflammatory reactions induced in mouse ear pinnae by the N-BPs alendronate (Ale) and zoledronate (Zol). We found the followi

Upregulation of HTRA1 mediated by the lncRNA NEAT1/miR-141-3p axis contributes to endometriosis development through activating NLRP3 inflammasome-mediated pyroptotic cell death and cellular inflammation.

The present study identified a novel upstream long chain non-coding (lncRNA) NEAT1/miR-141-3p/HTRA1 axis that regulated the activation of NLR family pyrin domain containing 3 (NLRP3) inflammasome to modulate endometriosis (EM) development. Specifically, clinical data suggested that the expression of NLRP3 and apoptosis-associated speck-like protein containing CARD (ASC), the cleavage of caspase-1 and gasdermin D (GSDMD), and the production of inflammatory cytokines (interleukin (IL)-1β, IL-6, tumor necrosis factor (TNF)-α, and IL-18) were all significantly increased in the ectopic endometrium

Systemic Inflammation Mediates the Association between Thyroid Hormone Sensitivity and Depressive Symptoms in Euthyroid Population: Findings from NHANES.

AIM: The relationship between thyroid function and depression remains unclear in euthyroid individuals. This study investigated the association between central thyroid hormone sensitivity (CTHS) and depression, and explored whether systemic inflammation mediated this relationship. METHODS: This cross-sectional analysis included 6,728 adults from National Health and Nutrition Examination Survey 2007-2012. Depression was defined as Patient Health Questionnaire score ≥10. CTHS was assessed using four indices: thyroid-stimulating hormone index (TSHI), thyrotropin thyroxine resistance index (TT4RI

ZDHHC5-mediated S-palmitoylation of NOD2 drives neutrophil extracellular trap formation via mitophagy to exacerbate inflammation in COPD.

Chronic obstructive pulmonary disease (COPD) is characterized by persistent neutrophilic inflammation, but its upstream molecular regulators remain incompletely defined. Here, NOD2 and the palmitoyl-acyltransferase ZDHHC5 were increased in COPD lung tissue and detected in pulmonary neutrophils. In CS-exposed mice, NOD2 deficiency attenuated lung inflammation, collagen deposition, EMT-associated changes, and neutrophil extracellular trap (NET) formation. Anti-Ly6G-mediated neutrophil depletion reproduced much of this protection and markedly reduced the differences between wild-type and NOD2-def

Evolocumab alleviates atherogenesis by inhibiting inflammation-mediated endothelial cell activation via the PI3K/AKT/NF-κB pathway.

Atherosclerosis, a chronic inflammatory disease, is the most relevant cause of ischaemic stroke or myocardial infarction. Vascular endothelial cells (ECs) play a significant role in the development of atherosclerosis. In this chronic inflammatory environment, we aimed to investigate whether a Evolocumab (Evb) could mitigate atherosclerosis progression by inhibiting EC activation via in vivo and in vitro assays. In vivo, we investigated the ability of Evb to prevent atherosclerotic lesion formation in ApoE-/- mice fed a Western diet and subjected them to carotid artery ligation. The results sho

Associations of estimated glucose disposal rate and overactive bladder and the potential mediating role of systemic inflammation: NHANES 2005-2018.

BACKGROUND: Insulin resistance (IR) is linked to overactive bladder (OAB), with chronic inflammation serving as a common pathophysiological mechanism in both conditions. As a robust marker of IR, estimated glucose disposal rate (eGDR) was selected as the exposure variable to examine its relationship with OAB, with a specific focus on the mediating role of inflammation. METHODS: We conducted a cross-sectional study using data from the 2005-2018 National Health and Nutrition Examination Survey (NHANES). We utilized weighted logistic regression to assess the association of eGDR with OAB, while r

Association of dietary fiber with all-cause mortality in U.S. adults with diabetes: Mediating role of the systemic immune-inflammation index.

Dietary fiber is recommended in diabetes management for its potential anti-inflammatory effects, but currently, the association between fiber intake, systemic inflammation, and long-term survival in adults with diabetes remains uncertain. This cohort study analyzed data from 3894 U.S. adults with diabetes across 10 National Health and Nutrition Examination Survey cycles (1999-2020). Fiber intake levels (grams of fiber per kilogram of body weight, g/kg) was assessed via dietary interviews, and mortality outcomes were linked to National Death Index records through 2019. The systemic immune-infla

Microglial serine racemase knockout alleviates Alzheimer-like neuropathology and behavioral deficit via lactylation-mediated anti-inflammation.

Serine racemase (SR) dysregulation associates with brain aging and Alzheimer's disease (AD), as both a deficiency and an excess of D-serine can impact synaptic neurotransmission and the integrity of synapses. Neuronal SR decreases with aging, while glial SR is upregulated in AD. However, the role of SR in microglia involved in AD remains elusive. Here, Srr knockdown/knockout in microglia enhanced whereas overexpression of SR inhibited phagocytosis. Lipopolysaccharide-treated Srr-/- microglia upregulated anti-inflammatory factors-an effect blocked by histone lactylation inhibition. Conditional

Associations of dietary inflammatory index with respiratory tract infections among US adults and the mediating roles of systemic inflammation markers.

Respiratory tract infections (RTIs) remain a major public health burden among adults, with substantial interindividual heterogeneity in susceptibility. Chronic low-grade systemic inflammation has been proposed as an important host-related determinant of infection risk, and the dietary inflammatory index (DII) provides an integrative measure of the inflammatory potential of habitual diet. However, evidence linking DII to RTI risk and the underlying immune-inflammatory mechanisms remains limited. We analyzed data from 9 cycles of the National Health and Nutrition Examination Survey, including 43

From isolation to inflammation-behavioral mediation explains the social origins of depression in obesity: insights from NHANES 2005-2023.

BACKGROUND: Depression and obesity frequently co-occur and share overlapping biological and psychosocial mechanisms. Yet, how social, dietary, and systemic inflammatory processes jointly shape depression risk in obesity remains unclear. METHODS: Using nationally representative data from the U.S. National Health and Nutrition Examination Survey (NHANES 2005-2023), we analyzed obese adults (n = 11,608). A multidimensional Social Isolation Index (SII) was developed to capture structural and socioeconomic isolation, alongside the Dietary Inflammatory Index (DII) and serum C-reactive protein (CRP)
