The intrinsic pathogenic role of autoantibodies to aquaporin 4 mediating spinal cord disease in a rat passive-transfer model.

Neuromyelitis optica (NMO) is causally linked to autoantibodies (ABs) against aquaporin 4 (AQP4). Here, we focused on the pathogenic effects exclusively mediated by human ABs to AQP4 in vivo. We performed cell-free intrathecal (i.th.) passive transfer experiments in Lewis rats using purified patient NMO immunoglobulin G (IgG) and various recombinant human anti-AQP4 IgG-ABs via implanted i.th. catheters. Repetitive application of patient NMO IgG fractions and of recombinant human anti-AQP4 ABs induced signs of spinal cord disease. Magnetic resonance imaging (MRI) revealed longitudinal spinal co

A strategy for designing inhibitors of alpha-synuclein aggregation and toxicity as a novel treatment for Parkinson's disease and related disorders.

Convergent biochemical and genetic evidence suggests that the formation of alpha-synuclein (alpha-syn) protein deposits is an important and, probably, seminal step in the development of Parkinson's disease (PD), dementia with Lewy bodies (DLB) and multiple system atrophy (MSA). It has been reported that transgenic animals overexpressing human alpha-syn develop lesions similar to those found in the brain in PD, together with a progressive loss of dopaminergic cells and associated abnormalities of motor function. Inhibiting and/or reversing alpha-syn self-aggregation could, therefore, provide a

Physiological significance of 2-buten-4-olide (2-B4O), an endogenous satiety substance increased in the fasted state.

A sugar acid, 2-B4O, has been found to increase from 3.5 to 13 microM in rat serum at 36 h after food deprivation. Injections of 2-B4O (2.5 microM) into the rat III cerebral ventricle (III ICV) suppress food intake and single neuronal activity in the lateral hypothalamic area (LHA). 2-B4O is effective even in 72 h food-deprived rats. 2-B4O hyperpolarizes glucose-sensitive neurons in the LHA via Na+-K+ pump activation, but depolarizes glucoreceptor neurons in the ventromedial nucleus (VMH) via closure of ATP-sensitive K channels. The plasma levels of glucose, corticosterone, and catecholamines,

Plasma Markers of Astrocytic and Axonal Integrity in Idiopathic/Isolated REM Sleep Behavior Disorder (iRBD) as Predictors of Dementia with Lewy Bodies.

BACKGROUND: Plasma biomarkers of neurodegeneration, astrogliosis, and neuroinflammation have been studied as potential biomarkers in neurodegenerative diseases. This study investigated whether these markers may predict phenoconversion to Parkinson's disease or dementia with Lewy bodies (DLB) in idiopathic/isolated REM sleep behavior disorder (iRBD). METHODS: In this longitudinal, single-center, iRBD cohort study (enrolled 2004-2022), plasma glial fibrillary acidic protein (GFAP), interleukin-6 (IL-6), neurofilament light chain (NfL), snare-associated protein 25 (SNAP25), soluble triggering re

Intracellular screening of a peptide library to derive a potent peptide inhibitor of α-synuclein aggregation.

Aggregation of α-synuclein (α-syn) into toxic fibrils is a pathogenic hallmark of Parkinson disease (PD). Studies have focused largely on residues 71-82, yet most early-onset mutations are located between residues 46 and 53. A semirationally designed 209,952-member library based entirely on this region was constructed, containing all wild-type residues and changes associated with early-onset PD. Intracellular cell survival screening and growth competition isolated a 10-residue peptide antagonist that potently inhibits α-syn aggregation and associated toxicity at a 1:1 stoichiometry. This was v

Vesicle permeabilization by protofibrillar alpha-synuclein: implications for the pathogenesis and treatment of Parkinson's disease.

Fibrillar alpha-synuclein is a component of the Lewy body, the characteristic neuronal inclusion of the Parkinson's disease (PD) brain. Both alpha-synuclein mutations linked to autosomal dominant early-onset forms of PD promote the in vitro conversion of the natively unfolded protein into ordered prefibrillar oligomers, suggesting that these protofibrils, rather than the fibril itself, may induce cell death. We report here that protofibrils differ markedly from fibrils with respect to their interactions with synthetic membranes. Protofibrillar alpha-synuclein, in contrast to the monomeric and

Structural diversity and specific distribution of O-glycans in normal human mucins along the intestinal tract.

Purified human mucins from different parts of the intestinal tract (ileum, cecum, transverse and sigmoid colon and rectum) were isolated from two individuals with blood group ALe(b) (A-Lewis(b)). After alkaline borohydride treatment the released oligosaccharides were structurally characterized by nano-ESI Q-TOF MS/MS (electrospray ionization quadrupole time-of-flight tandem MS) without prior fractionation or derivatization. More than 100 different oligosaccharides, with up to ten monosaccharide residues, were identified using this technique. Oligosaccharides based on core 3 structures, GlcNAc(

Clinical relevance of the Helicobacter pylori gene for blood-group antigen-binding adhesin.

Infection with Helicobacter pylori is associated with different human gastric diseases. Biochemical studies, in vitro adherence assays, and in vivo animal models revealed that epithelial attachment of H. pylori can be mediated by the blood-group antigen-binding adhesin (BabA) targeting human Lewis(b) surface epitopes. Studies with transgenic mice expressing the Lewis(b) epitope have shown that such attachment can alter disease outcome. In the current study, the presence of the babA2 gene encoding the adhesin was investigated in clinical isolates from a German population by using PCR and revers

Cyclic (alkyl)(amino)carbenes (CAACs): stable carbenes on the rise.

CONSPECTUS: Carbenes are compounds that feature a divalent carbon atom with only six electrons in its valence shell. In the singlet state, they possess a lone pair of electrons and a vacant orbital and therefore exhibit Lewis acidic and Lewis basic properties, which explains their very high reactivity. Following the preparation by our group in 1988 of the first representative, a variety of stable carbenes are now available, the most popular being the cyclic diaminocarbenes. In this Account, we discuss another class of stable cyclic carbenes, namely, cyclic (alkyl)(amino)carbenes (CAACs), in wh

Characterization of the immune response to a secondary encephalitogenic epitope of basic protein in Lewis rats. II. Biased T cell receptor V beta expression predominates in spinal cord infiltrating T cells.

The immune response of Lewis rat lymph node T cells to guinea pig myelin basic protein (GP-BP) in experimental allergic encephalomyelitis is directed primarily against a region of basic protein encompassed by residues 72-89. T cells that respond to this epitope are restricted by the RT1.B class II molecule of the MHC and use V beta 8.2 exclusively in their TCR. A second region of GP-BP, residues 87-99, also induces experimental allergic encephalomyelitis in Lewis rats but this response is restricted primarily by RT1.D. Elsewhere we describe the biologic characteristics of T cell clones respond

Operando Chlorination Engineering in PEMWE for Direct Brine Electrolysis to Produce Green Hydrogen.

Direct seawater electrolysis in proton exchange membrane water electrolyzers is severely bottlenecked by competitive chlorine evolution and catalyst corrosion. Here, we report an operando chlorination strategy that harnesses corrosive ions (Cl-, SO4 2 -) to self-assemble a renewable catalytic layer on a defective RuIrAg pre-catalyst for highly selective water oxidation. In acidic brine (0.5 M H2SO4 + 3.0 M KCl), it achieves a remarkably low oxygen evolution reaction (OER) overpotential of 108 mV at 10 mA cm- 2, with a full-cell energy consumption of 41.01 kWh kg- 1 H2 at 1 A cm- 2. Exploring the Catalytic Activity of Ge(II) and Sn(II) Cationic Crown Ether Complexes: Hydrosilylation.

Lewis acid-catalyzed hydrosilylation of carbonyl compounds was explored using cationic germanium(II) and tin(II) crown ether complexes as catalysts. Only the germanium(II) crown ether complexes were found to reduce aldehydes efficiently and selectively to one product, the symmetric ether. The scope of the reaction was explored using [Ge([12]crown-4)2][OTf]2 as the catalyst revealing tolerance for a variety of functional groups, excluding those containing nitrogen. Lower conversions and less selectivity were observed in ketone hydrosilylations. The mechanism of aldehyde reduction catalyzed by [

Synergistic Ni-MoO3 Redox Catalysis in MIL-101(Cr) for Efficient H2O2 Activation and Ultra-Deep Desulfurization.

In this study, a novel Ni-MoO3@MIL-101(Cr) composite catalyst was synthesized via a one-step hydrothermal method for the oxidative desulfurization (ODS) of dibenzothiophene (DBT). The catalyst combines the high specific surface area of MIL-101(Cr) with the redox properties of Ni-MoO3, providing an effective synergy between adsorption and catalytic oxidation. The incorporation of Ni as a promoter significantly enhances catalytic activity, enabling complete sulfur removal (∼100%) under mild conditions (50 °C) within 60 min. The Catalytic Enantioselective Sommelet-Hauser Rearrangement.

The Sommelet-Hauser rearrangement of ammonium ylides incorporates a dearomatizing [2,3]-sigmatropic rearrangement of a (hetero)aromatic substituent, followed by tautomerization (rearomatization), to generate (hetero)benzylic amine derivatives. Despite widespread interest in this process, asymmetric variants have previously relied upon stoichiometric chiral auxiliary or chirality transfer strategies to induce stereocontrol. In this manuscript, the organocatalytic enantioselective Sommelet-Hauser rearrangement of ammonium ylides bearing heteroaromatic substituents is demonstrated using a Lewis b

Lysosome-Acidifying Nanoparticles Rescue A30P α-Synuclein Induced Neuronal Death in Cellular and Drosophila Models of Parkinson's Disease.

Parkinson's disease (PD) is the second most common neurodegenerative disorder, affecting over 10 million people worldwide. It is characterized by the progressive loss of dopaminergic neurons in the substantia nigra and the accumulation of misfolded α-synuclein (αSyn) in intracellular inclusions known as Lewy bodies. Emerging evidence links αSyn accumulation to impaired lysosomal acidification and defective autophagy-lysosomal degradation, which are central to disease progression. To address this lysosomal dysfunction, we engineered a novel type of lysosome-targeted acidic nanoparticles (AcNPs)

Lipid Acyl Chain-Driven α-Synuclein Fibril Polymorphisms and Neuronal Pathologies.

Conformational variations in α-syn fibrils are thought to underlie the distinct clinical features of synucleinopathies, including Lewy body dementia (LBD), Parkinsons's disease (PD), and multiple system atrophy (MSA), suggesting that distinct fibril structures act as molecular fingerprints linked to disease phenotype. While the origins of these conformational variations remain unclear, increasing evidence points to membranes as key modulators of fibrils conformations. In this study, we investigated how age-related alterations in membrane composition and fluidity influence α-syn fibril formatio

Regioselective Aldol Reactions Directed by Charge and Orbital Effects in Cyclic Germyl Dienolates: Synthesis of Three Distinct Regio- and Stereoisomers.

Regioselective aldol reactions of dienolates represent a valuable method for the construction of complex molecular scaffolds; however, controlling the reaction site of the dienolate remains a significant challenge. This study presents a method for regioselectivity switching in the aldol reactions of cyclic Ge-dienolates, which are generated from the (4 + 1) cycloaddition of allenoates with Ge(II) salts. Through systematic tuning of the charge and orbital distributions of the Ge-dienolates and their aldehyde reaction partners, regio- and stereoselective access to three distinct aldol adducts (s

Multi-site Molecular Regulation Enables Efficient Low-Dimensional Metal Halide/Organic Nano-Interdigitated Heterojunction Solar Cells.

Low-dimensional metal halides offer considerable environmental stability and structural tunability; however, their power conversion efficiencies (PCEs) still lag far behind those of their three-dimensional counterparts. While ordered p-n interdigitated heterojunctions present a highly promising architecture to maximize interfacial charge extraction, implementing this strategy in low-dimensional frameworks is severely impeded by disordered grain growth, high trap densities, and phase heterogeneity. Herein, we introduce bis(carboxymethyl) trithiocarbonate (BMTTC) as a multifunctional bulk additi

Anion-Doped Manganese Oxides Enabling Relay Catalysis for Aerobic Alcohol Ammoxidation.

The development of noble-metal-free heterogeneous catalysts for room-temperature aerobic ammoxidation of alcohols to nitriles represents an enduring challenge, primarily hampered by unselective lattice-oxygen over-oxidation via the classical Mars-van Krevelen (MvK) pathway and strong site-blocking effects. Herein, we report a dual-anion (C/N) doped MnOx (C/N-MnOx) synthesized via a green, solvent-free crystallization strategy, which exhibits excellent catalytic performance (95.3% benzyl alcohol conversion, >99% benzonitrile selectivity) and broad substrate versatility (35 diverse alcohol subst
