Effect of combined oral contraceptive use on verbal memory function in healthy women.

PURPOSE: Female sex hormones as well as the synthetic hormones contained within combined oral contraceptives (COCs) may influence emotional and cognitive functioning including learning and memory; however, findings are inconsistent. We here present the largest study to date investigating the effect of COC use on verbal memory in healthy women. METHODS: COC use and verbal memory scores were available from the CIMBI database for 205 healthy women in the reproductive age. We assessed if verbal memory and affective bias differed between COC users and non-users. In a subgroup of natural cycling wo

The association of memory disorders and chronic HIV disease in the antiretroviral therapy era: a systematic literature review.

OBJECTIVES: Despite recent pharmacological progress, memory impairment is still frequently reported in people living with HIV. We aimed to conduct a systematic literature review investigating the presence of impairment of (sub)components of memory function in patients prescribed highly active antiretroviral therapy (ART). METHODS: We adopted a cognitive neuropsychological model of memory function as the theoretical framework, distinguishing between a short-term working memory component and a long-term component of memory, along with their specific (sub)components. We systematically searched f

Apelin-13 prevents hippocampal synaptic plasticity impairment in Parkinsonism rats.

The hippocampus is involved in learning and memory for novel information and implicated within the cognitive dysfunction in Parkinson's disease. Long-term potentiation (LTP), the most type of synaptic plasticity, is the base of learning and memory. We evaluated the consequences of apelin-13 on early long-term potentiation (E-LTP) in the Cornu Ammonis (CA1) area of the hippocampus and synaptic hippocampal protein expression of postsynaptic density protein 95 (PSD-95) and dopaminergic receptor (DR1) of the rat model of Parkinsonism. 6-hydroxydopamine (6-OHDA) was infused within the right substan

Evolutionary conserved role of neural cell adhesion molecule-1 in memory.

The neural cell adhesion molecule 1 (NCAM-1) has been implicated in several brain-related biological processes, including neuronal migration, axonal branching, fasciculation, and synaptogenesis, with a pivotal role in synaptic plasticity. Here, we investigated the evolutionary conserved role of NCAM-1 in learning and memory. First, we investigated sustained changes in ncam-1 expression following aversive olfactory conditioning in C. elegans using molecular genetic methods. Furthermore, we examined the link between epigenetic signatures of the NCAM1 gene and memory in two human samples of healt

Comparative effects of zopiclone, triazolam and placebo on memory and psychomotor performance in healthy volunteers.

This study evaluated the effects of acute doses of zopiclone (7.5 mg), triazolam (0.25 mg) and placebo on memory and psychomotor performance of 12 normal volunteers. The subjects received both drugs in a repeated measure, double-blind Latin square design. The tests (CFF, CRT, DSST, memory assessments) were performed before and 2 and 6 hr after treatment. Zopiclone and triazolam induced an anterograde amnesia affecting short-term and long-term memory which lasted less than 6 hr. No retrograde amnesia was observed. Two hr after drug intake of both hypnotics psychomotor performances were signific

Cholecystokinin B receptor agonists alleviates anterograde amnesia in cholecystokinin-deficient and aged Alzheimer's disease mice.

BACKGROUND: As one major symptom of Alzheimer's disease (AD), anterograde amnesia describes patients with an inability in new memory formation. The crucial role of the entorhinal cortex in forming new memories has been well established, and the neuropeptide cholecystokinin (CCK) is reported to be released from the entorhinal cortex to enable neocortical associated memory and long-term potentiation. Though several studies reveal that the entorhinal cortex and CCK are related to AD, it is less well studied. It is unclear whether CCK is a good biomarker or further a great drug candidate for AD.

Contextual fear memories formed in the absence of the dorsal hippocampus decay across time.

Mammals suffering damage to the hippocampus display a dramatic loss of explicit, recently formed memories (retrograde amnesia). In contrast, deficits in the ability to form new memories following hippocampal damage (anterograde amnesia) can be overcome with sufficient training. By combining contextual fear conditioning with lesions of the dorsal hippocampus in rats, we discovered that while animals can form long-term contextual fear memories in the absence of the hippocampus, these memories decay with time, lacking the permanence that is a hallmark characteristic of normal fear memories. These

Induction of memory-like dendritic cell responses in vivo.

Dendritic cells (DCs), a vital component of the innate immune system, are considered to lack antigen specificity and be devoid of immunological memory. Strategies that can induce memory-like responses from innate cells can be utilized to elicit protective immunity in immune deficient persons. Here we utilize an experimental immunization strategy to modulate DC inflammatory and memory-like responses against an opportunistic fungal pathogen that causes significant disease in immunocompromised individuals. Our results show that DCs isolated from protectively immunized mice exhibit enhanced transc

Short-Long Heart Rate Variation Increases Dispersion of Action Potential Duration in Long QT Type 2 Transgenic Rabbit Model.

The initiation of polymorphic ventricular tachycardia in long QT syndrome type 2 (LQT2) has been associated with a characteristic ECG pattern of short-long RR intervals. We hypothesize that this characteristic pattern increases APD dispersion in LQT2, thereby promoting arrhythmia. We investigated APD dispersion and its dependence on two previous cycle lengths (CLs) in transgenic rabbit models of LQT2, LQT1, and their littermate controls (LMC) using random stimulation protocols. The results show that the short-long RR pattern was associated with a larger APD dispersion in LQT2 but not in LQT1 r

Negative attributes of mixed-valence memories strengthen over long retention intervals and the degree of enhancement is predicted by individual differences in state anxiety.

Memories are multifaceted and can simultaneously contain positive and negative attributes. Here, we report that negative attributes of a mixed-valence memory dominate long-term recall. To induce a mixed-valence memory, running responses were randomly reinforced with either food (∼83% of trials) or footshock (∼17% of trials), or a noise conditioned stimulus (CS) was followed randomly with either food (∼80% of trials) or footshock (∼20% of trials). Control animals were consistently reinforced with only food. Mixed-valence training promoted unstable behavior (e.g., erratic approach and withdrawal

In vivo assessment of mechanisms underlying the neurovascular basis of postictal amnesia.

Long-lasting confusion and memory difficulties during the postictal state remain a major unmet problem in epilepsy that lacks pathophysiological explanation and treatment. We previously identified that long-lasting periods of severe postictal hypoperfusion/hypoxia, not seizures per se, are associated with memory impairment after temporal lobe seizures. While this observation suggests a key pathophysiological role for insufficient energy delivery, it is unclear how the networks that underlie episodic memory respond to vascular constraints that ultimately give rise to amnesia. Here, we focused o

Central Nervous System Vasculitis Mimicking Glioma in a Patient With Ulcerative Colitis: A Case Report and Literature Review.

A patient with ulcerative colitis presented with progressive memory impairment and alexia, with brain imaging showing a mass-like lesion mimicking glioma. Histopathology confirmed central nervous system (CNS) vasculitis. The patient's memory and reading ability improved and finally recovered following corticosteroid treatment, with no recurrence during the long-term follow-up. The case report and literature review highlight that clinicians should consider CNS vasculitis in patients with ulcerative colitis and tumor-like brain lesions.

A Novel NLRP3 Inhibitor AMS-17 Rescues Deficits in Long-Term Potentiation Following Mild Traumatic Brain Injury in Adult C57Bl/6 Mice.

Traumatic brain injury (TBI) is a leading cause of long-term disability, with limited effective treatment options. A key factor of TBI pathophysiology is neuroinflammation, which can involve the activation of the nucleotide-binding domain leucine-rich repeat protein 3 (NLRP3) inflammasome. Aberrant inflammation following injury has the ability to reduce the capacity to induce long-term changes in synaptic plasticity, a leading mechanism for the development of learning and memory deficits following injury. This study investigated the potential of a novel NLRP3 inhibitor, AMS-17, to mitigate syn

Memory Rehabilitation for people with multiple sclerosis.

BACKGROUND: Impairments in cognitive function, particularly memory, are common in patients with Multiple Sclerosis (MS) and these can potentially affect their ability to complete functional activities. There is evidence from single-case or small group studies that memory rehabilitation can be beneficial for patients with MS but findings from randomised controlled trials (RCTs) and systematic reviews have been inconclusive. OBJECTIVES: To determine the effectiveness of memory rehabilitation for patients with MS who have memory problems, and the effect of such interventions on functional abilit

Impairment in social interaction and hippocampal long-term potentiation at perforant pathway-dentate gyrus synapses in a prenatal valproic acid-induced rat model of autism.

It is well established that prenatal valproic acid exposure in rats leads to autism-like behaviours and social deficits. Long-term potentiation changes in the brain have been proposed as a potential mechanism in the development of autistic behaviour. However, there are controversies regarding the effect of in utero valproic acid exposure on long-term potentiation. This study examined the social interaction and long-term potentiation induction in perforant pathway-dentate gyrus synapses in male offspring of a rat model of autism induced by prenatal exposure to valproic acid. On Embryonic Day 12

The 5-Hydroxytryptamine 2C Receptor Agonist Improves Sensorimotor Gating and Cognitive Deficits by Reversing Excitation/Inhibition Imbalance in the Medial Prefrontal Cortex.

BACKGROUND AND HYPOTHESIS: Although the 5-hydroxytr-yptamine 2C receptor (5-HT2C) receptor agonists have demonstrated efficacy in mitigating sensorimotor gating deficits in the schizophrenic animal model of glutamatergic dysfunction, the precise neural mechanisms underlying these effects remain incompletely understood. This study investigates how 5-HT2C receptor activation improves sensorimotor gating and cognitive function by modulating Excitation/Inhibition balance in the medial prefrontal cortex (mPFC). STUDY DESIGN: We first employed chemogenetic approach-es to establish that selective ac

Alprazolam induces anterograde amnesia for contextual fear memory and alters dorsoventral hippocampal neuronal ensembles in female mice.

Benzodiazepines (BZDs) are commonly prescribed anxiolytic drugs that act on GABAa receptors, and can result in anterograde amnesia, or the inability to form new memories. While BZDs have been used for decades, the brain regions and neuronal mechanisms responsible for this detrimental side effect are largely unknown at the systems neuroscience level. To analyze the effects of BZDs on long-term memory, activity-dependent ArcCreERT2 x eYFP mice were injected with Alprazolam 30 min prior to a 3-shock contextual fear conditioning (CFC) procedure and encoding ensembles were tagged with eYFP. Mice we

Selective decline of intact HIV reservoirs during the first decade of ART followed by stabilization in memory T cell subsets.

OBJECTIVES: To investigate the short- and long-term dynamics of intact and defective proviral HIV DNA during ART. DESIGN: We evaluated viral reservoir dynamics in a cohort of nine individuals with chronic HIV-1 subtype B who initiated first-line ART and were followed for 20 years while continuing ART. METHODS: PBMCs were obtained before ART ( n  = 5), during the first year, and after 8.5 and 20 years of treatment. T cell subsets (naive, central-memory, transitional-memory and effector-memory) were sorted at 8.5 and 20 years. DNA was isolated and analyzed using the intact proviral DNA assay (

Dietary n-3 PUFAs Deficiency Increases Vulnerability to Inflammation-Induced Spatial Memory Impairment.

Dietary n-3 polyunsaturated fatty acids (PUFAs) are critical components of inflammatory response and memory impairment. However, the mechanisms underlying the sensitizing effects of low n-3 PUFAs in the brain for the development of memory impairment following inflammation are still poorly understood. In this study, we examined how a 2-month n-3 PUFAs deficiency from pre-puberty to adulthood could increase vulnerability to the effect of inflammatory event on spatial memory in mice. Mice were given diets balanced or deficient in n-3 PUFAs for a 2-month period starting at post-natal day 21, follo

Animals lacking endothelin-converting enzyme-2 are deficient in learning and memory.

Endothelin-converting enzyme (ECE)-2 is a metalloprotease that possesses many properties consistent with it being a neuropeptide-processing enzyme. This protease is found primarily in neural tissues, with high levels of expression in midbrain, cerebellum, hypothalamus, frontal cortex and spinal cord and moderate levels in hippocampus and striatum. To evaluate its role in neural function, mice have been generated lacking this enzyme. Physical appearance, autonomic reflexes, motor co-ordination, balance, locomotor activity and spontaneous emotional responses appear normal in these knockout (KO)
