Movement disorders associated with chronic GM2 gangliosidosis. Case report and review of the literature.

A 24-year-old man presented with dystonia, dementia, amyotrophy, choreoathetosis, and ataxia. Partial hexosaminidase A deficiency was documented in serum and leukocytes and confirmed by rectal biopsy with ganglion cells containing membranous cytoplasmic bodies. A brief review of the literature reveals that tremor, dystonia and choreoathetosis are common but neglected symptoms associated with chronic GM2 gangliosidosis.

[Experience with using amedin in children with movement disorders of extrapyramidal and labyrinth origin].

Using previously obtained data concerning the effectivity of amedin in motor and speech disorders due to muscular dystonia the drug was used for the treatment of 50 children from 3.5 months to 18 years, with motor disorders of an extrapyramidal and labyrinth nature. A high therapeutical effectivity of amedin in an increased pathological activity of tonic cervical and labyrinth reflexes in infants and in torsion spasm of school age children is demonstrated.

Cryoprobe to current practice: neurosurgical legacies of Dr. Irving S. Cooper in movement disorders.

Before the advent of levodopa therapy for Parkinson's disease, surgical lesioning of the motor thalamus or globus pallidus represented the primary therapeutic intervention for Parkinson's disease and essential tremor at a time when effective medical alternatives were severely limited. In this context, Irving S. Cooper directed one of the most prolific and controversial lesioning practices of the mid-20th century, performing thousands of procedures and documenting outcomes. Through chemopallidectomy, chemothalamectomy, and later cryosurgical techniques, Cooper investigated the relationship betw

Aquaporin-4 in Narcolepsy Type 1: Investigation of Perivascular Fluid Movement in Sleep Disorders.

Narcolepsy type 1 (NT1) is caused by the loss of hypocretin-1 leading to excessive daytime sleepiness and cataplexy. Additionally, disrupted nighttime sleep has become an increasingly recognized feature of NT1. As the glymphatic fluid movement has been linked to sleep architecture, we investigated cerebrospinal fluid (CSF) Aquaporin-4 (AQP4) as potential biomarker for perivascular fluid exchange processes in 133 NT1 patients, 145 patients with other sleeping disorders, and 62 controls. NT1 patients showed lower CSF AQP4 concentrations compared to both control groups (p < 0.0001 and p = 0.01).

Magnetic Resonance-Guided Focused Ultrasound (MRgFUS) for Movement Disorders: A Practical Guide for Neurologists.

Magnetic resonance-guided focused ultrasound (MRgFUS) is a novel, noninvasive therapeutic approach that shows potential in treating movement disorders, such as essential tremor, Parkinson's disease, and dystonia. This technique combines high-intensity focused ultrasound with real-time magnetic resonance imaging, allowing for accurate targeting and thermal ablation of deep brain areas without incisions or exposure to ionizing radiation. MRgFUS provides precise lesioning with intraoperative feedback, enabling immediate assessment of symptoms. Clinical trials and initial clinical applications hav…

Research progress in the role of tubal ciliary movement in female infertility-related disorders.

Tubal ciliary movement is one of the essential transport mechanisms for female fertility, playing a key role in facilitating oocyte pickup and transporting the fertilized ovum. This movement is mediated by multiciliated cells and regulated by specific proteins and hormones that modulate ciliary number, length, polarity, beat frequency, and amplitude to ensure proper function. Genetic mutations, inflammatory stimuli, and hormonal fluctuations can impair ciliary activity or induce ciliary apoptosis, leading to ciliary dysfunction. Disorders of tubal ciliary movement are frequently observed in pr

Vagus Nerve Stimulation in Movement Disorders, from Principles to a Systematic Review of Evidence.

BACKGROUND: The vagus nerve (VN), the principal component of the parasympathetic branch of the autonomic nervous system (ANS), mediates bidirec communication between the central nervous system (CNS) and peripheral organs. Vagus nerve stimulation (VNS), delivered through invasive (iVNS) or non-invasive (transcutaneous cervical [tcVNS] and transcutaneous auricular [taVNS]), exerts multimodal effects on multiple circuits and neurotransmitters, and is currently under investigation in movement disorders. OBJECTIVES: This systematic review evaluates the therapeutic potential of VNS in movement diso…

Using Multinodal Deep Brain Stimulation for Treatment of Complex Movement Disorders: Initial Case Series.

BACKGROUND AND OBJECTIVES: Mixed and complex movement disorders represent significant challenges for surgical treatment. These disease states are likely the result of combined or complex network pathologies affecting multiple pathways. Until recently, attempts to treat complex movement disorders with deep brain stimulation (DBS) have primarily focused on stimulating a single deep target to address patients' most severe symptoms. However, a multinodal targeting approach for DBS by simultaneously stimulating more than 1 pair of homologous nuclei could be a more effective treatment strategy. In t…

Neurodegenerative and neurodevelopmental roles for bulk lipid transporters VPS13A and BLTP2 in movement disorders.

BACKGROUND: Bridge-like lipid transfer proteins (BLTPs) mediate bulk lipid transport at membrane contact sites. Mutations in BLTPs are linked to both early-onset neurodevelopmental and later-onset neurodegenerative diseases, including movement disorders. The tissue specificity and temporal requirements of BLTPs in disease pathogenesis remain poorly understood. OBJECTIVES: To determine the age-of-onset and tissue-specific roles of VPS13A and BLTP2 in movement disorder pathogenesis using Drosophila models. METHODS: We generated tissue-specific knockdowns of the VPS13A ortholog ( Vps13 ) and th…

The spectrum of movement disorders in young children with ARX-related epilepsy-dyskinesia syndrome.

Children with developmental and epileptic encephalopathies often present with co-occurring dyskinesias. Pathogenic variants in ARX cause a pleomorphic syndrome that includes infantile epilepsy with a variety of movement disorders ranging from focal hand dystonia to generalized dystonia with frequent status dystonicus. In this report, we present three patients with severe movement disorders as part of ARX-associated epilepsy-dyskinesia syndrome, including a patient with a novel pathogenic missense variant (p.R371G). These cases illustrate diagnostic and management challenges of ARX-related diso…

Movement disorders associated with substance use in adults living in precarious housing or homelessness.

OBJECTIVE: Many individuals living in precarious housing or homelessness have multimorbid illnesses, including substance use, psychiatric, and neurological disorders. Movement disorders (MDs) associated substance use are amongst the poorly studied subtopics of drug-induced MDs. The aim of the present study was, therefore, to determine the proportion affected and severity of different signs of MDs, as well as their associations with substance use in a community-based sample of precariously housed and homeless individuals. METHODS: Participants were recruited from an impoverished urban neighbor…
