Endogenous opiates mediate cardiac sympathetic inhibition in response to a pressor stimulus in rabbits.

Two different patterns of response to a pressor stimulus occurred in conscious rabbits. This difference was not apparent when a depressor stimulus was applied. At levels of mean arterial pressure exceeding 120 mmHg one group of animals exhibited a marked bradycardia which was due to sympathetic inhibition in addition to vagal activation while this sympathetic component appeared to be lacking in the second group of animals. Naloxone (0.1 mg/kg i.v.) markedly reduced the sympathetic inhibition elicited by phenylephrine but had no significant effect on the reflex vagal stimulation. Naloxone there

Analysis of carbamazepine actions in hippocampal slices of the rat.

Carbamazepine at therapeutic concentrations has a strong inhibitory effect on the spontaneous field bursts of the CA 1 region of rat hippocampal slices in low-Ca2+, high-Mg2+ solution. A reduction of excitability and synaptic transmission as well as no effect on posttetanic potentiation in the rat hippocampal slice by carbamazepine agrees with previous studies on spinal cord and peripheral nerve. Carbamazepine left synaptic inhibition and hyperpolarizing afterpotentials unaltered while these inhibitory processes were markedly enhanced by pentobarbital and adenosine, respectively. The spontaneo

Reflex adrenergic inhibition of gastric motility by nociceptive intestinal stimulation and peritoneal irritation in the cat.

The effects on gastric motility of nociceptive stimulation to jejunum and colon were studied in vagotomized anesthetized cats. Mechanical nociceptive stimulation and diathermy of the jejunum and proximal colon elicited reflex gastric inhibition that was significantly more pronounced than that obtained by stimulation of distal colon. Similarly differentiated reflex responses were induced by electric afferent stimulation of nerves from the respective intestinal segments. Strong nociceptive stimuli from the abdominal cavity, induced by peritonitis, completely blocked vagal excitatory influences o

An electrooculographic study of internuclear ophthalmoplegia.

The eye movements of 25 patients with internuclear ophthalmoplegia were recorded by electrooculography. The velocity of adducting saccades was markedly less than normal. The velocity of abducting saccades was within the normal range, but statistically there was a wider distribution. Recordings were made in 2 patients several months after the onset of internuclear ophthalmoplegia, at which time the adducting eye velocity was greater than the abducting eye velocity. A patient with a unilateral medial fasciculus lesion showed marked overshoot of the abducting eye on contralateral saccades and ove

Spike-wave rhythms in cat cortex induced by parenteral penicillin. II. Cellular features.

Epileptiform potentials, consisting of spontaneous, generalized bursts frequently assuming a 3/sec spike-wave form and tonic clonic electrographic seizures were produced in 32 lightly anesthetized cats by parenteral injections of penicillin. The activity of 83 identified pyramidal tract cells and 207 cortical non-pyramidal tract cells was correlated with the surface EEG. The majority of both cell types generated depolarizations and action potentials with the EEG spike. Hyperpolarizations, during which cells were inhibited, followed the depolarizations. The depolarizations responded to injected

Activation of the Gamma-Aminobutyric Acid (GABA)ergic Neural Circuit in Salicylate-Induced Tinnitus: the Inferior Colliculus to the Medial Geniculate Body.

OBJECTIVES: This study aimed to investigate the regulatory functions of gamma-aminobutyric acid (GABA)ergic neural circuits from the inferior colliculus (IC) to the medial geniculate body (MGB) in salicylate-induced tinnitus. METHODS: Mice were treated with salicylate to induce tinnitus, and tinnitus-like behaviors were evaluated via gap prepulse inhibition of acoustic startle. Using combined viral tracing methodologies, we identified and mapped the pathways and connections from the IC to the MGB. Furthermore, we employed Gq-coupled human M3 designer receptors exclusively activated by designe

Treacher Collins syndrome: unmasking the role of Tcof1/treacle.

Treacher Collins syndrome (TCS) is a rare congenital birth disorder characterized by severe craniofacial defects. The syndrome is associated with mutations in the TCOF1 gene which encodes a putative nucleolar phosphoprotein known as treacle. An animal model of the severe form of TCS, generated through mutation of the mouse homologue Tcof1 has recently revealed significant insights into the etiology and pathogenesis of TCS (Dixon and Dixon, 2004; Dixon et al., 2006; Jones et al 2008). During early embryogenesis in a TCS individual, an excessive degree of neuroepithelial apoptosis diminishes the

Synaptic Inhibition in Avian Interaural Level Difference Sound Localizing Neurons.

Synaptic inhibition plays a fundamental role in the neural computation of the interaural level difference (ILD), an important cue for the localization of high-frequency sound. Here, we studied the inhibitory synaptic currents in the chicken posterior portion of the dorsal nucleus of the lateral lemniscus (LLDp), the first binaural level difference encoder of the avian auditory pathway. Using whole-cell recordings in brain slices, we provide the first evidence confirming a monosynaptic inhibition driven by direct electrical and chemical stimulation of the contralateral LLDp, establishing the re

Korsakoff syndrome and altered pain perception: a search of underlying neural mechanisms.

OBJECTIVES: Patients with Korsakoff syndrome (KS) may have a diminished pain perception. Information on KS and pain is scarce and limited to case descriptions. The present study is the first to investigate the underlying neural mechanisms of altered pain perception in patients with KS more systematically. METHODS: We conducted a literature search on neural correlates of pain perception in other neurocognitive disorders in which extensive research was done. RESULTS: The brain areas that are affected in KS showed considerable overlap with the neural correlates of pain perception in other neuro

3D organoids containing endothelial and neural cells generation by serial inductions of differentiation on human iPSC-derived embryoid bodies.

3D brain organoids have been widely used as a tool to study human brain development and disorders. Although endothelial cells play important roles in the brain development and pathogenesis in neurological disorders, most 3D brain organoids lack inherent endothelial cells and need either the addition of endothelial cells or to be transplanted to animals to reconstitute such vascular structures, likely missing the developmental interactions of endothelial cells and other cells in the human brain. In order to reconstitute a 3D organoid mimicking the in vivo neural and endothelial cells developmen

Reticulospinal neurons inactivated by warming of the preoptic area and anterior hypothalamus of rabbits.

To identify the premotor neurons for vasoconstrictors of the skin, activities of reticulospinal neurons in the rostroventral medulla, the ear sympathetic nerve (ESNA) and the renal sympathetic nerve (RSNA) were recorded in anesthetized and immobilized Japanese White or New Zealand White rabbits. Two groups of neurons were identified according to their responses to thermal stimulation of the preoptic area and the anterior hypothalamus (POAH) and to electrical stimulation of baroreceptor afferents, the aortic nerve (AN). Neurons (Type I neurons, n = 21) whose activity was inhibited by warm stimu

The mesodermal and myogenic specification of hESCs depend on ZEB1 and are inhibited by ZEB2.

Human embryonic stem cells (hESCs) can differentiate into any cell lineage. Here, we report that ZEB1 and ZEB2 promote and inhibit mesodermal-to-myogenic specification of hESCs, respectively. Knockdown and/or overexpression experiments of ZEB1, ZEB2, or PAX7 in hESCs indicate that ZEB1 is required for hESC Nodal/Activin-mediated mesodermal specification and PAX7+ human myogenic progenitor (hMuP) generation, while ZEB2 inhibits these processes. ZEB1 downregulation induces neural markers, while ZEB2 downregulation induces mesodermal/myogenic markers. Mechanistically, ZEB1 binds to and transcript

Developmental Disruption of GABAAR-Meditated Inhibition in Cntnap2 KO Mice.

GABA released from presynaptic sites induces short-lived phasic inhibition mediated by synaptic GABAA receptors (GABAARs) and longer-duration tonic inhibition mediated by extrasynaptic GABAA or GABAB receptors (GABABRs). A number of studies have found that contactin-associated protein 2 (Cntnap2) knockout (KO) mice, a well-established mouse model of autism, exhibit reduced interneuron numbers and aberrant phasic inhibition. However, little is known about whether tonic inhibition is disrupted in Cntnap2 KO mice and when the disruption of inhibition begins to occur during postnatal development.

Cognitive maps and schizophrenia.

Structured internal representations ('cognitive maps') shape cognition, from imagining the future and counterfactual past, to transferring knowledge to new settings. Our understanding of how such representations are formed and maintained in biological and artificial neural networks has grown enormously. The cognitive mapping hypothesis of schizophrenia extends this enquiry to psychiatry, proposing that diverse symptoms - from delusions to conceptual disorganization - stem from abnormalities in how the brain forms structured representations. These abnormalities may arise from a confluence of ne

Towards a neural network based therapy for hallucinatory disorders.

Pattern completion in a neural network model of the thalamus and a biologically plausible model of synaptic plasticity are the key concepts used in this paper for analyzing some cognitive disorders that involve hallucinations of several kinds: visual hallucinations in the Charles Bonnet syndrome and psychedelic drugs consumption, somatic hallucination in phantom limbs, cognitive hallucinations in schizophrenia and even in multiple personality disorders. All these types of hallucinations are proposed to be the result of a pattern completion dynamics performed in thalamic deafferented areas. Eff

Modulation of human primary motor cortex plasticity during proactive and retroactive interference between motor practice and paired associative stimulation.

Proactive and retroactive interference have been widely demonstrated behaviorally, but their neurophysiological effects have rarely been compared within the same experimental framework. This study compared the primary motor cortex (M1) plasticity responses when motor practice (MP) was performed before or after paired associative stimulation (PAS25) and explored whether these responses were accompanied by changes in intracortical inhibition. A within-subject design was employed. A right-hand precision grip task with real-time electromyographic feedback was used to examine practice-related refin

Direct evidence of upper motor neuron excitability changes in a patient with ALS.

A key feature of amyotrophic lateral sclerosis (ALS) pathophysiology is motor neuron hyperexcitability. However, the mechanisms of hyperexcitability are not well understood. Prior studies have used transcranial magnetic stimulation (TMS) to demonstrate increased motor cortex excitability and reduced intracortical inhibition in human ALS. Yet, interpretation of these findings is limited because measurement of muscle responses cannot disentangle the specific contribution of upper and lower motor neurons and of cortical interneurons to excitability changes. We had the rare opportunity to record d

Cerebellar Brain Inhibition and Its Association with Motor Inhibition and Reaction Time In Younger and Older Adults.

Motor performance declines with age, particularly affecting reaction time and proactive response inhibition. While cortical influences on age-related motor decline are well-documented, the cerebellum's role remains unclear. Cerebellar Brain Inhibition (CBI), which can be measured through dual-site transcranial magnetic stimulation (TMS), may provide insights into age-related changes in motor control. We aimed to (1) compare resting-state CBI between young and older adults, (2) investigate the relationship between CBI and upper limb motor performance, and (3) examine whether this relationship d

Necl-4/CADM4 regulates GABAergic synaptic strength on GABAergic inhibitory neurons via ErbB4 activation and prevents neuronal impairments.

Neural networks comprise excitatory and inhibitory neurons, linked through excitatory and inhibitory synapses. Synaptic excitation/inhibition balance is controlled for brain development and functions, and its dysregulations are implicated in aging-dependent neuronal impairments. Here, we found that Necl-4/CADM4, an immunoglobulin superfamily cell adhesion molecule, is expressed in γ-aminobutyric acidergic (GABAergic) inhibitory neurons and localizes at GABAergic synapses on inhibitory neurons in cultured hippocampal neurons and the mouse hippocampus. Necl-4 genetic ablation induced neuronal lo
