Caution on the assessment of intestinal parasitic load in studying parasite-mediated sexual selection: the case of Blackbirds coccidiosis.

The parasite-mediated sexual selection (PMSS) theory has led to an increasing number of experimental studies essentially focussed on blood parasites. Currently, more research is being carried out on intestinal parasites in relationship to this theory. Before testing the theory with gastrointestinal parasites, it is important: (i) to determine an optimal research methodology to obtain an accurate assessment of parasite burden and (ii) to have information about life-history traits of the parasite to interpret data appropriately. In this study, we present data on oocyst output of Isosporaturdi in…

5-ALA does not potentiate dihydroartemisinin against Plasmodium falciparum malaria parasites.

In this Correspondence, H. Behrens and colleagues show that the potentiating effect of the small molecule 5-aminolaevulinic acid (5-ALA) on the activity of artemisinin observed in cancer does not translate to malaria parasites. Thus, co-administration of artemisinin and 5-ALA does not seem to be a promising strategy to improve malaria treatment. [Image: see text]

The big picture : Parasites, People and the Path to Ending Schistosomiasis.

Affecting 110 million African children, schistosomiasis control demands an urgent shift from drug treatments to permanent transmission interruption. This requires pairing paediatric drug formulations and new interventions including any new vaccines with locally-relevant multisectoral policy, WASH infrastructure and community-led behaviour change as well as sustained domestic financing. Detection of intestinal parasites from feces of captive ophidians in Mexico.

Ophidians host a wide range of pathogens, including many parasite species, some of which may be of public health concern because they can infect other animal species and humans. Herpetariums house snakes in captivity as part of ex-situ conservation initiatives aimed at preserving threatened species. However, stress, immunosuppression, and limited space increase the risk of parasitic infections, including zoonotic diseases. Here, we detect the presence of intestinal parasites in ophidian species housed under conditions of captivity. Twenty specimens representing sixteen species were examined us…

COLLECTION AND EXAMINATION OF SMALL-BODIED, FRESHWATER FISH FOR HELMINTH PARASITES.

Small-bodied, freshwater fish are often used in parasitological studies because they host a variety of parasites. As a result, it is important to have an accessible resource for the collection, housing, and examination of helminth parasites. Fish may be collected using passive (e.g., minnow traps or fyke nets) or active (e.g., seine netting or electrofishing) sampling methods depending on the species and water body being sampled. It is best to keep fish alive (e.g., in aquaria) until immediately prior to euthanasia for dissection. Using an overdose of anesthetic is the preferred method of sacr…

A preliminary survey of feline filarial parasites in Kerala State, India.

Feline filarial parasites remain among the least investigated in India, despite the well-established endemicity of filariosis in canine and human populations. This study aimed to assess filarial infections in domestic cats in Kerala using both microscopy and molecular techniques, with a focus on evaluating their potential role as reservoir hosts. From September 2023 to February 2024, 300 owned cats presented to the University Veterinary Hospital, Mannuthy, were screened for circulating microfilariae using wet blood film examination. Four cats (1.33%) tested positive, of which three harbored sh…

Cross-species transmission of gastrointestinal parasites between wild and domestic canids.

The interface between wild and domestic canids can facilitate cross-transmission of gastrointestinal parasites, with implications for biodiversity conservation and public health. In this study, we evaluated the occurrence and diversity of these parasites in fecal samples from Cerdocyon thous (n = 7) collected within the Serra das Araras Ecological Station (EESA) and from domestic dogs (Canis lupus familiaris) (n = 10) sampled in surrounding rural properties in the Cerrado biome, Brazil. Four parasite morphotypes were identified in C. thous and six in dogs, as well as mite eggs in wild canids' …

Targeting Kinetoplastid Parasites with ProTide Prodrugs: A Proof-of-Concept Study.

Neglected tropical diseases (NTDs) remain a major global health challenge, particularly in low- and middle-income countries. Kinetoplastid parasites causing Chagas disease, leishmaniasis, and African trypanosomiasis rely on host purine salvage pathways, making nucleoside analogues attractive therapeutic candidates. However, their clinical utility is limited by poor cellular uptake and rapid metabolism. Herein, we report the application of the ProTide prodrug technology, a clinically validated approach that enhances the intracellular delivery of nucleoside monophosphates for the treatment of ki…

Current status of -omics research in platyhelminth parasites of teleost fish.

Parasitic platyhelminths (Trematoda, Monogenea, and Cestoda) are responsible for widespread disease in wild and cultured fish in both freshwater and marine environments globally, impacting both human health and the sustainability of aquaculture industries. Research into these parasites primarily aims to understand the impact of infections on host species, as well as developing management strategies to reduce or eliminate disease. -Omics-based research can assist in uncovering the biology of these parasites across a range of key areas, including understanding host-pathogen interactions, identif…

Nematomorph parasites potentially drive nutritional flow from terrestrial to aquatic ecosystems.

Long-chain polyunsaturated fatty acids (LC-PUFA), particularly eicosapentaenoic acid (EPA; 20:5n-3) are synthesized primarily by algae, such as diatoms and cryptophytes, in aquatic ecosystems. Although LC-PUFA flux has commonly been considered unidirectional from aquatic to terrestrial ecosystems, recent evidence suggests a potential reciprocal flow of EPA from terrestrial to aquatic systems via amphibious insects and terrestrial arthropods. Here, we report the possibility that nematomorph parasites drive the flow of EPA from terrestrial to aquatic ecosystems by inducing EPA-bearing terrestria…

Neutrophils and purinergic signaling: Partners in the crime against Leishmania parasites?

The parasite of the genus Leishmania is the causative agent of diseases that affect humans called leishmaniasis. These diseases affect millions of people worldwide and the currently existing drugs are either very toxic or the parasites acquire resistance. Therefore, new elimination mechanisms need to be elucidated so that new therapeutic strategies can be developed. Much has already been discussed about the role of neutrophils in Leishmania infection, and their participation is still controversial. A recent study showed that receptors present in the neutrophil membrane, the purinergic receptor…

Genetically attenuated parasites show promise as a next-generation malaria vaccine.

Metabolically active, genetically attenuated Plasmodium falciparum parasite lines are promising second-generation malaria vaccine candidates. Lamers et al. and Roozen et al. demonstrated in recent Phase 1/2a trials that GA2 parasites, designed to arrest late during liver-stage development and transmitted via mosquito bites, can induce substantial protection against sporozoite challenge infection.

Host-Derived Geranylgeraniol Shields Intraerythrocytic Stages of Malaria Parasites from Fosmidomycin.

Fosmidomycin was proposed as an antimalarial drug but failed in clinical trials due to recrudescence, a phenomenon whose causes remain poorly understood. The mechanism of action of fosmidomycin is the inhibition of the methylerythritol 4-phosphate (MEP) pathway, essential for producing isoprenoids in Plasmodium parasites. The key isoprenoids produced by the MEP pathway are farnesyl and geranylgeranyl pyrophosphates (FPP and GGPP), vital for protein isoprenylation, ubiquinone, and dolichol biosynthesis. In vitro studies have demonstrated that prenols, like farnesol (FOH) and geranylgeraniol (GG…

An Essential Adaptor for Apicoplast Fission and Inheritance in Malaria Parasites.

Blood-stage Plasmodium falciparum parasites rely on a non-photosynthetic plastid, the apicoplast, for survival, making it an attractive target for antimalarial intervention. Like the mitochondrion, the apicoplast cannot be generated de novo and must be inherited by daughter parasites during cell division. This inheritance relies on coordinated apicoplast positioning and fission, but the molecular mechanisms controlling these processes remain poorly understood. Here, we identify a previously uncharacterized P. falciparum protein (Pf3D7_0613600), which we name PfAnchor, as a key regulator of api…

Understanding the traits underlying vaccine-driven virulence evolution in malaria parasites.

BACKGROUND: Vaccine-driven evolution can erode the beneficial effects of vaccination and is a concern, especially for newly introduced vacci … malaria infection dynamics to experimental time series data from infections in mice inoculated with parasites that had evolved in either vaccinated mice or sham-vaccinated (control) mice. We compared parameter estimates across treatments to understand which parasite traits could plausibly explain differences in infection dynamics and virulence. RESULTS: Va …

Within-Host "Infraecology": Asymmetric Interactions Between Two Co-infecting Eye Parasites.

Parasites within the host body can occupy similar ecological niches and, therefore, compete for resources. Similarly to macroecological environments, within-host habitats often provide unequal access to resources for different parasitic species. Parasites that live closer to the source of nutrients likely "intercept" resources, thus inhibiting the growth of "downstream" competitors. The eyes of vertebrates are an example of such asymmetric habitats, as lenses have priority access to nutrients over the vitreous humor. However, parasitic interactions in asymmetric environments have been rarely p…

Advances, challenges, and applications of laboratory culture of digenean trematode parasites.

The development of parasite cultures has long been pivotal in advancing parasitology, with broad applications in medicine, veterinary science, and biology. Laboratory cultures are invaluable tools for studying parasite biology, host-parasite interactions, and the development of treatments and vaccines. However, cultures of digenean trematodes under laboratory conditions remain a challenging yet critical endeavour in parasitology. These parasites hold significant importance to both human health and ecological systems. Nevertheless, trematodes exhibit a complex life cycle involving multiple host…
