Origins and functions of cells essential for periodontal repair: the role of fibroblasts in tissue homeostasis.

OBJECTIVE: A review is undertaken of rodent model systems and cell culture studies that address the role of periodontal fibroblasts in tissue homeostasis in both normal function and after wound healing. RATIONALE: Fibroblasts are the predominant cells of the periodontal ligament (PL) and of healthy gingiva and have important roles in the development, function and regeneration of the tooth support apparatus. REVIEW: In normally functioning periodontal tissues cell turnover involves generation of new cells by proliferation which in turn is balanced by apoptopic cell death. Consequently PL fibr

Osteocalcin, prostaglandin E2 and alkaline phosphatase in gingival crevicular fluid: their relations to periodontal status.

Prostaglandin E2 (PGE2) and alkaline phosphatase (ALP) have often been measured in gingival crevicular fluid (GCF) as possible indicators of gingival inflammation and bone metabolism. Osteocalcin (OC), a major component of bone matrix, is mainly produced by osteoblasts, and could also be considered as a marker of bone turnover. The aims of this preliminary study were to examine if OC was present in GCF and to assess the relationships of OC, PGE2 and ALP in GCF to periodontal conditions. GCF samples were collected with durapore strips from 34 healthy, 72 gingivitis and 118 periodontitis sites i

Aetiological aspects of mandibular tooth agenesis--focusing on the role of nerve, oral mucosa, and supporting tissues.

The purpose of the present investigation was to consider possible aetiological aspects of mandibular tooth agenesis by comparing the pattern of mandibular molar and premolar agenesis with radiological evidence of the mandibular canal. Orthopantomograms from 33 children affected by agenesis of at least four premolar and/or molar teeth in the mandible were investigated. Three children were affected by ectodermal dysplasia. Two anthropological mandibles with absence of mandibular canals were included in the study for comparison. According to the investigation, agenesis should be divided into thre

Studies related to reaction of supporting soft tissue to denture wear: the histological response of vervet monkey oral epithelium to a -80 mmHg vacuum.

The histological response of vervet monkey oral epithelium to a negative force similar to that experienced under dentures was investigated. Using impression trays with adherent impressions linked to a vacuum pump, the epithelia of the hard palate, attached gingiva and alveolar mucosa of 16 monkeys were subjected to a continuous vacuum of -80 mmHg, and then fixed by perfusion and immersion while in situ. After processing for light microscopy, sections were measured to obtain the rete peg length, supra-papillary width, epithelial width and basal, spinous and superficial cell density 700 microns-

In vivo findings associated with heat generation during thermomechanical compaction of gutta-percha. 2. Histological response to temperature elevation on the external surface of the root.

An in vivo investigation is described and a histological evaluation made of the effect of canal obturation by thermomechanical compaction of gutta-percha and sealer on the cementum on the lateral surface of the root and adjacent periodontal membrane and alveolar bone of the ferret canine after time intervals of 24 hours, 20 days and 40 days. These tissue reactions were compared with those in the roots of control teeth filled by lateral condensation of cold gutta-percha and sealer. Iatrogenic damage was apparent in a minority of the experimental specimens 20 and 40 days after obturation.

A comparison of alveolar bone in young and aged mice.

To study the relationship between aging and alveolar bone loss, the periodontia of young (12-week-old) and aged (94-week-old) C57B1/6 Nia mice were compared, giving special attention to the alveolar bone. The tibiae were also examined to distinguish local from systemic bone changes. The mean area of interproximal bone between the first and second mandibular molars was significantly less in the aged mice than in the young mice. While there was no difference in the distance from the cementoenamel junction (CEJ) to the alveolar crest, the width of the interproximal bone was narrower in the aged a

Post-operative dental pain and analgesic efficacy. Part II. Analgesic usage and efficacy after dental surgery.

The analgesics taken by patients after oral and periodontal surgery were noted over a three day observation period. Analgesic consumption matched closely the pain experience. The efficacy of self-prescribed analgesics was extrapolated from the pain scores obtained in the first 12 hours after surgery, and overall, the apparent efficacy appears poor. However, those patients who reported taking aspirin recorded significantly less pain than those who took either paracetamol or combination analgesics. Analgesic efficacy was not related to dose, although a significant correlation was noted between t

Association of periodontal condition with insulin sensitivity - results of the Oulu1935 cohort study.

OBJECTIVE: To investigate whether baseline periodontal condition predicts reduced insulin sensitivity (RIS) over a follow-up period of about … respectively. CONCLUSIONS: Poor periodontal condition and edentulousness predicted RIS. A healthy periodontium appears to support normal insulin function, but further studies are needed.

The oral-immune axis: neutrophil extracellular traps mediate the link between periodontitis and autoimmune diseases.

Epidemiological and clinical evidence supports a strong association between periodontitis and autoimmune diseases. This review systematically elucidates the pivotal role of the "oral-immune axis" in this relationship, focusing on neutrophil extracellular traps (NETs) as key mediators. Periodontal pathogens can compromise the gingival epithelial barrier, induce excessive NETs formation within the periodontium, facilitate systemic dissemination of pathogens, and promote their ectopic spread to organs such as the joints, pancreas, and kidneys. These processes, in turn, may activate neutrophils to…

Airway-Facially Generated Treatment Planning (AFGTP) in Complex Anterior Open-Bite Cases Integrating Airway Screening, FGTP, Aligner Therapy, and Skeletal Anchorage.

Complex anterior open-bite malocclusions require a holistic, risk-stratified approach that extends beyond dental alignment to address the airway, facial form, function, biomechanics, and periodontium. This article outlines a contemporary airway-facially generated treatment planning (AFGTP) protocol that begins with airway screening and dentofacial analysis, incorporates functional and biomechanical planning, and integrates orthodontic execution using clear aligners and skeletal anchorage (temporary anchorage devices [TADs]) to achieve posterior intrusion and mandibular counterclockwise autorot…

Mechanisms of mechanical force in periodontal homeostasis: a review.

Mechanical forces affect periodontal health through multiple mechanisms. Normally, mechanical forces can boost soft and hard tissue metabolism. However, excessive forces may damage the periodontium or result in irreversible inflammation, whereas absence of occlusion forces also leads to tissue atrophy and bone resorption. We systemically searched the PubMed and Web of Science databases and found certain mechanisms of mechanical forces on immune defence, extracellular matrix (ECM) metabolism, specific proteins, bone metabolism, characteristic periodontal ligament stem cells (PDLSCs) and non-cod…

Periodontopathogen-Related Cell Autophagy-A Double-Edged Sword.

The periodontium is a highly organized ecosystem, and the imbalance between oral microorganisms and host defense leads to periodontal diseases. The periodontal pathogens, mainly Gram-negative anaerobic bacteria, colonize the periodontal niches or enter the blood circulation, resulting in periodontal tissue destruction and distal organ damage. This phenomenon links periodontitis with various systemic conditions, including cardiovascular diseases, malignant tumors, steatohepatitis, and Alzheimer's disease. Autophagy is an evolutionarily conserved cellular self-degradation process essential for e…

However, due to the limited investigations and their inconsistent methodology, further well-designed prospective studies are necessary.
