The separation of caffeine and its nine analogues by micellar electrokinetic capillary chromatography (MECC) using sodium dodecyl sulphate (SDS) as the micellar phase was investigated. This study was concerned with what stimulus information humans with amblyopia use to judge the shape of simple objects. We used a string of four Gabor patches to define a contour. A fifth, center patch served as the test pattern. The observers' task was to judge the location of the test pattern relative to the contour. A rapid and reliable procedure, based on a C8 bonded phase extraction and reversed-phase isocratic high-performance liquid chromatographic separation with internal standard quantitation, has been developed for the determination of the antipsychotic drug chlorpromazine and thirteen common metabolites. The method allows quantitation of these analytes at the ng/ml concentration range in human plasma. An evaluation of recovery, detection limits, and reproducibility is presented along with application of the method to patient samples.

The performance of a commonly used high-pressure liquid chromatographic stationary phase, octadecylsilane, has been evaluated for 30 compounds selected as representative of a wide variety of drug substances. Chromatographic behavior is found to be highly predictable on the basis of pKa and partition coefficient, and the stationary phase should be especially valuable for the separation of acidic and neutral drugs. For basic drugs, however, the column efficiency is poor, detracting from the overall usefulness.
