The effects of pH on the sodium-withdrawal contractures evoked from frog atrial trabeculae.

Contractures can be evoked from frog atrial trabeculae by raising extracellular pH in Na-free fluid after the spontaneous relaxatin of the tension evoked by removing extracellular Na+. These alkalinity contractures are unaffected by local anaesthetics but are absent following a brief perfusion with Ca-free fluid but are partially inhibited by local anaesthetics. Perfusion by solution containing a high Pco2 fails to induce contractures. The relationship between [Na]0 and tension is altered by changing extracellular pH; acidic fluids below pH 6.0 reduce tension and alkaline fluids increase tensi

Acute meningoencephalitis after withdrawal of antibiotics in Whipple's disease.

A man with Whipple's disease was treated with oral penicillin (500 mg twice a day) for 2 years with eradication of bacillary organisms from the jejunum and a return of jejunal histologic findings to normal. While he was on this regimen, however, intermittent vertigo and tinnitus and decreased auditory acuity developed. Two days after penicillin was withdrawn, the patient developed acute meningoencephalitis that responded to parenteral penicillin and chloramphenicol therapy. Subsequently, central nervous system signs and symptoms and cerebrospinal fluid pleocytosis have been controlled with chr

[Mechanism of action of Novocain administered intravenously to animals].

Tests conducted with 48 dogs and 100 rabbits demonstrated that intravenous administration of novocain has a well-marked positive influence in extreme conditions, such as massive blood losses and traumatic shocks. It is shown that in the mechanism of the drug's action particularly important is its capability to increase the cardiac performance, to eliminate spasms of resistive vessels and to contract the blood-carrying vessels of the microcirculatory stream. The final effect, however, is in a great measure dependent upon the initial functional state of the organism (the period of hemorrhagic or

Mode of action of trichloroethylene on squid axon membranes.

The mode of action of trichloroethylene on electrical properties of squid giant axons has been studied by means of voltage clamp techniques. Trichloroethylene decreased the resting membrane potential in a manner dependent upon the concentration, the depolarization by 50% saturated trichloroethylene attaining 28.4 and 32.7% of the initial value at 20 and 10 degrees C, respectively. Leakage conductance was decreased to 34.6% of the control by 30% saturated trichloroethylene at 10-12 degrees C. It appears that the trichloroethylene-induced depolarization is at least in part due to a decrease in r

Skeletal muscle necrosis following membrane-active drugs plus serotonin.

Administration of imipramine plus serotonin (5-HT) to rats has been proposed as an animal model of Duchenne muscular dystrophy. We studied t … rs-labilizers". Other drugs which are "membrane stabilizers-labilizers" such as trihexyphenidyl and procaine also promoted skeletal muscle necrosis when given prior to 5-HT. It is proposed that the effects of imipramine plus 5-HT on skeletal muscle are not due to the blockade of neuronal uptake of 5-HT and subsequent vascular-induced ischemia, but reflect …

Role of the vegetation in experimental Streptococcus viridans endocarditis.

This study examines the role of the vegetation in catheter-induced experimental endocarditis in predisposing to bacterial colonization of cardiac valves and in influencing the course of the disease and response to penicillin therapy. Platelet-fibrin vegetations developed at areas of valvular trauma and were colonized when Streptococcus viridans were injected intravenously. Pretreatment with warfarin prevented vegetation formation, but animals still developed endocarditis at the same rate after injection of 10(6)S. viridans. The course of the disease in anticoagulated animals was more explosive

The actions of some cholinomimetic drugs on the isolated taenia of the guinea-pig caecum.

1. The actions on the taenia of 4-(m-chlorophenylcarbamoyloxy)-2-butynyl-trimethylammonium chloride (McN-A-343), N-benzyl-3-pyrrolidyl acetate methobromide (AHR-602), tetramethylammonium (TMA) and choline phenyl ether have been examined and compared with the actions of acetylcholine, nicotine and 1,1-dimethyl-4-phenylpiperazinium (DMPP).2. Responses of the taenia to these agonists are quantitatively and often qualitatively dependent on the tone of the preparation. A method is described which makes allowance for the effect of tone on heights of contractions.3. Acetylcholine, McN-A-343 and AHR-6
