Two trials were conducted to compare the efficacy of parvaquone and buparvaquone for the treatment of naturally acquired East Coast fever (ECF, Theileria parva infection) which, if untreated, is almost invariably fatal in European breeds of cattle. In the first trial 28 naive cattle were exposed in a paddock infested with ticks carrying a virulent form of the disease. Twelve were treated with each drug when they developed clinical ECF. All 24 cattle were cured. In the second study, 100 cases of ECF occurring naturally on farms in Kenya were treated, 50 with each drug. Parvaquone cured 44 (88%)

Insecticide resistance is a major problem that continues to plague efforts to control pests of animals and crops. An important mechanism by which insects become resistant to insecticides is via increased detoxification mediated by the cytochrome P450 microsomal monooxygenases (monooxygenases). One of the fundamental gaps in our knowledge about this resistance mechanism is an understanding of how insects express high levels of the specific cytochrome P450(s) responsible for resistance. One such P450, CYP6D1, causes resistance to pyrethroid in the house fly and is expressed at 9-fold higher leve

From May 1989 to June 1990 an experimental study has been undertaken in two village's malarial hyperendemy of the Malian savannah. This study's objective was to test the effectiveness, on paludometric parameters and the openness of the population to the impregnated curtains and blankets, of permethrin as strategy of controlling malaria. The experimental village (Tiénéguébougou) and the control one (Kambila) are located in the same ecosystem and as at least 3 km any from each other. The impregnated curtains were installed in all the experimental village's houses and all the 216 residents have r

The Onchocerciasis Control Programme of the World Health Organization uses larvicides to fight against the aquatic stages of the vector Simulium damnosum s.l., and thereby interrupt transmission of the disease. Since the appearance of resistance to Abate and chlorphoxim in certain cytotypes of the vector, the efficacy of many possible replacement insecticides has been tested and the impact of the best of them (permethrin, cyphenothrin, pyraclofos, and carbosulfan) on the aquatic fauna evaluated.

Effects of local anesthetics and anticonvulsants on the pyrethroid-modified sodium current in cultured mouse neuroblastoma cells have been investigated using the suction pipette voltage clamp technique. In the presence of 10 microM of the pyrethroid deltamethrin the sodium current consists of an enhanced peak current during membrane depolarization and a slowly decaying, deltamethrin-induced tail current remaining after repolarization. At the onset of block the local anesthetics tetracaine, lidocaine and QX 314 reduced the deltamethrin-induced tail current more effectively than the peak current

During an outbreak of Rhodesian sleeping sickness in the Lambwe Valley in 1980 initial tsetse control measures consisted of applications of dieldrin to the periphery of the Ruma National Park. This activity had a marked effect on the prevalence of sleeping sickness. Concern about the use of dieldrin caused the cessation of this programme and justified an aerial spray programme using endosulfan. Although the Lambwe Valley did not appear to be a good candidate for aerial spray, the endosulfan had a marked effect on tsetse fly levels and on the prevalence of sleeping sickness. Sleeping sickness c

Cypermethrin (CYP), a neurotoxic pesticide, is released into the environment uncontrollably, posing a particular threat to aquatic species. Amphora coffeaeformis was demonstrated to have a protective effect against hazardous CYP exposure by markedly reducing neurotoxicity in Oreochromis niloticus. A total of 240 O. niloticus were divided equally into four groups in triplicate as follows: Control, AMP (5% A. coffeaeformis/kg diet), CYP (1.25 µg/L), and AMP + CYP (5% A. coffeaeformis/kg diet + 1.25 µg/L). Our data showed that CYP exposure for 30 days induced a significant decrease in γ-aminobuty

BACKGROUND: Canine leishmaniosis (CanL) is endemic in Italy and is an important veterinary and public health concern. Integrated preventive strategies combining vaccination and vector control are recommended in endemic areas, and field data supporting the safety and effectiveness of this concurrent use are limited.
METHODS: A longitudinal field study conducted on Favignana Island (Sicily, Italy) evaluated the concurrent use of a vaccine (LETIFEND®, Leti) and sandfly repellent collar (SCALIBOR®, MSD Animal Health). Client-owned dogs (154) were prescreened by clinical examination, serology, qua

BACKGROUND: Monitoring insecticide levels and physical integrity over time is essential for assessing the durability of insecticide-treated nets (ITNs), which largely depends on the net handling habits of users. This study determined the insecticide content and effectiveness of ITNs (Yorkool and PermaNet 2.0) at 6, 12, 24, and 36 months after a mass distribution campaign in Mali.
METHODS: At 6 months (May, 2018), 12 months (December, 2018), 24 months (November-December, 2019), and 36 months (November-December, 2020), 30 nets were randomly collected from households in the districts of Kenieba

Over-reliance on pyrethroid insecticides in insecticide-treated nets (ITNs) has imposed significant selection pressure for the evolution of insecticide resistance among major malaria vector species. In the Democratic Republic of Congo (DRC), the country with the second highest malaria burden globally, pyrethroid resistance is pervasive, but there is a paucity of information regarding the molecular mechanisms driving resistance. A clear understanding of the specificity of resistance mechanisms to individual insecticides and the likelihood of selecting for cross-resistance mechanisms is crucial

Metabolic resistance to pyrethroids driven by cytochrome P450s is threatening malaria control interventions and may provide cross-resistance to insecticides with unrelated modes of action. Here, we show that cytochrome P450 genes CYP6P9a/b associated with pyrethroid resistance in Anopheles funestus also confer cross-resistance to a novel mitochondrial complex I inhibitor insecticide, code-named Sherlock. Using standard bioassays (CDC bottle bioassays, WHO cone bioassays, and WHO tunnel tests), the Sherlock and pyrethroid insecticides were tested against pyrethroid-resistant An. gambiae s.s. (N

INTRODUCTION: Luangwa District has one of the longest running legacy datasets in Zambia regarding reliable monitoring of confirmed malaria cases through the national Health Management Information System (HMIS). It was also one of the first districts to achieve sustained coverage with long-lasting insecticidal nets (LLINs) and indoor residual spraying (IRS) of insecticides.
METHODS: HMIS data from2009 to 2021 were analysed using generalized linear mixed models, to assess the effects of LLINs and IRS on rates of inpatient admissions with severe malaria and total confirmed malaria cases. IRS tre

BACKGROUND: Development of insecticide resistance (IR) in sand fly populations is an issue of public health concern, threatening leishmaniasis mitigation efforts by insecticide-based vector control. There is a major knowledge gap in the IR status of wild populations worldwide, possibly attributed to the unavailability of specialized tools, such as bioassay protocols, species baseline susceptibility to insecticides and molecular markers, to monitor such phenomena in sand flies.
METHODOLOGY/PRINCIPAL FINDINGS: Sand fly populations from (semi-)rural regions of Greece, Turkey and Iran were sample

BACKGROUND: Organophosphates and pyrethroids are two major groups of insecticides used for crop protection worldwide. They are neurotoxicants and exposure during vulnerable windows of brain development may have long-term impact on human neurodevelopment. Only few longitudinal studies have investigated associations between prenatal exposure to these substances and intelligence quotient (IQ) at school age in populations with low, mainly dietary, exposure.
OBJECTIVE: To investigate associations between maternal urinary concentrations of insecticide metabolites at gestational week 28 and IQ in of

Beta-cypermethrin (β-CYP) is a highly effective broad-spectrum insecticide that can potentially affect female reproduction. However, little is known about the effect of β-CYP on uterine decidualisation, which is a vital process by which the uterus provides a suitable microenvironment for pregnancy maintenance. Therefore, we focused on the effect and mechanism of β-CYP on endometrial decidualisation during early pregnancy in mice. The results indicated that the expression levels of HOXA10, BMP2, and IGFBP1 was significantly downregulated in the decidual tissue and primary endometrial stromal ce

Thymus alternans and Teucrium montanum subsp. jailae are medicinal and aromatic plants, typical of Slovakian flora, producing bioactive essential oils. In the present study, we evaluated the insecticidal potential of the essential oils, obtained by hydrodistillation from the plant aerial parts and analysed by GC-MS, as insecticidal agents. For the purpose, they were assayed against three insect species acting as agricultural pests or vectors of medical relevance, such as the common housefly, Musca domestica L., the lymphatic filariasis vector, Culex quinquefasciatus and the Egyptian cotton lea

In 2005, the zoonotic acute sleeping sickness was spreading rapidly from the endemic areas of southeastern Uganda with potential for merger into areas affected by the chronic form of the disease in northwest Uganda. Movement of cattle reservoirs due to restocking was blamed for the rapid spread. To stop the spread of the zoonotic sleeping sickness, cattle in the disease endemic areas had to be treated with trypanocidal drugs and sprayed with deltamethrin to promote the live bait technology that helps suppress the tsetse vector. The initiative that started in five high-risk districts in 2006 wi

BACKGROUND: Head lice infestation (Pediculosis) is one of the most important health challenges particularly in primary school-aged children. It is often present among 6-11-year-old students in various tropical and temperate regions of the world. The aim of this study was to examine epidemiologic indices and comparative analysis of two pyrethroid-based and one non-chemical pediculicide products on head lice treatment of primary school girls in a rural setting of Fars province, south Iran, as part of a randomized controlled assessor blind trial.
METHODS: Before treatment, infested students were

Fusarium dieback (FD) is a new vascular disease of hardwood trees caused by Fusarium spp. and other associated fungal species which are vectored by two recently introduced and highly invasive species of ambrosia beetle (Euwallacea spp. nr. fornicatus). One of these ambrosia beetles is known as the polyphagous shot hole borer (PSHB) and the other as the Kuroshio shot hole borer (KSHB). Together with the fungi that they vector, this pest-disease complex is known as the shot hole borer-Fusarium dieback (SHB-FD) complex. Mitigation of this pest-disease complex currently relies on tree removal; how
