High-Pressure Freezing EM Tomography of Entire Ribbon Synapses in the Retina.

In the retina, presynaptic active zones in photoreceptors and bipolar cells are distinguished by a plate-like "ribbon" linked to the plasma membrane (PM) and surrounded by dozens of synaptic vesicles (SVs) tethered to it. SVs at the base of the ribbon, closest to the PM, are thought to constitute the readily releasable vesicle pool (RRP), i.e., SVs primed to be released 1-2 ms following stimulation. The number of SVs in the RRP is a critical synaptic parameter that influences synaptic strength and varies with light levels to enable ribbon synapses to compute visual information. Physiological R…

Decoding cell-class specific roles of non-coding variants in human retina.

Non-coding variants in cis-regulatory elements such as promoters and enhancers contribute to inherited retinal diseases (IRDs), however, characterizing the functional impact of most regulatory variants remains challenging. To improve identification of variants of interest, we implemented machine learning using a gapped k-mer support vector machine approach trained on single nucleus ATAC-seq data from specific cell classes of the adult and developing human retina. We developed 18 distinct ML models to predict the impact of non-coding variants on 39,437 cell-class-specific regulatory elements. T…

Mechanisms Underlying Myopia Progression from Visual Signaling to Metabolic Remodeling in Retina.

Myopia has emerged as a major global health concern due to its rising prevalence and the associated risk of irreversible visual impairment. Although scleral and choroidal changes have traditionally received emphasis, recent studies highlight the retina as a key contributor to the onset and progression of myopia. This review incorporates recent advances across three interrelated domains: visual pathway modulation, functional impairment, and metabolic remodeling. Abnormal ON pathway signaling-particularly under mesopic lighting-disrupts emmetropization feedback and promotes ocular elongation. As…

Short-Chain Fatty Acids and the Gut-Retina Connection: A Systematic Review.

The interplay between gut microbiota and retinal health, known as the gut--retina axis, has gained increasing attention in recent years. Short-chain fatty acids (SCFAs), metabolites produced by gut microbiota, have been identified as key mediators of gut-retina communication. This systematic review explores the role of SCFAs in retinal health and their potential impact on the development and progression of retinal diseases, such as diabetic retinopathy (DR), age-related macular degeneration (AMD), and glaucoma. A literature search was conducted across multiple databases, including PubMed, Goog…

Recent advances of novel nanoformulations combatting blue light hazards in the retina.

Exposure to blue light significantly threatens retinal health, affecting approximately 73 % of the global population. It impairs vision, lowers the quality of life, and adds to public health challenges. Its treatment is getting difficult due to chronicity, limited drug efficacy, side effects, and ocular barriers that hinder conventional drug delivery. Nanoformulations (NFs) have gained attention as adaptable drug delivery systems (DDSs), offering controlled release and customizable physicochemical characteristics. Their application in ocular therapy has shown promise for targeting both anterio…

Horizontal cell connectivity in the anchovy retina-a 3D electron microscopic study.

BACKGROUND: Block-face scanning electron microscopy has opened a new era of connectomics research, in which it is possible to make dense reconstructions of all cells in a clipping of a neuronal network, such as the retina, resolving synaptic contacts. Anchovies, exceptionally abundant marine teleosts, have retinae with regions for triple cone-based color vision and a region with specialized cone photoreceptors, so-called polycones, made of long and short cones with axially oriented outer segment lamellae for polarization contrast vision. This modality, discovered in the 1970s, is unique in ver…

Virtual human retina: Simulating neural signalling, degeneration, and responses to electrical stimulation.

INTRODUCTION: Current brain-based visual prostheses pose significant challenges impeding adoption such as the necessarily complex surgeries and occurrence of more substantial side effects due to the sensitivity of the brain. This has led to much effort toward vision restoration being focused on the more approachable part of the brain - the retina. Here we introduce a novel, parameterised simulation platform that enables study of human retinal degeneration and optimization of stimulation strategies. The platform bears immense potential for patient-specific tailoring and serves to enhance artifi…

Protective effects of empagliflozin against NMDA-induced excitotoxicity in the rat retina.

Empagliflozin (EMPA), a sodium-glucose cotransporter 2 inhibitor used in patients with type 2 diabetes, exerts various beneficial effects, including anti-inflammatory and antioxidant properties, in addition to its glucose-lowering effect. In this study, we examined whether EMPA protects against N-methyl-d-aspartic acid (NMDA)-induced retinal excitotoxicity and the mechanisms underlying its protective effects. Male Sprague-Dawley rats (7-8 weeks old) were used in this study. The number of cells in the ganglion cell layer (GCL) decreased 7 days after intravitreal injection of NMDA (50 nmol). Sim…

Brain and retina in Alzheimer's disease: Pathological intersections and estimates from imaging.

Recent studies have highlighted retinal optical coherence tomography (OCT) imaging as a promising biomarker for the early detection of Alzheimer's disease (AD). This review connects AD brain pathology - particularly amyloid beta (Aβ), tau, and vascular changes - with corresponding retinal changes. Evidence suggests that abnormal Aβ and tau deposits in the retina may reflect brain pathology, though their formation mechanisms remain unclear. Retinal vascular changes may also mirror brain pathology, with recent data emerging on other co-pathologies. Retinal thickness changes, especially in acetyl…

Identification of Chlamydia pneumoniae and NLRP3 inflammasome activation in Alzheimer's disease retina.

Chlamydia pneumoniae (Cp), an obligate intracellular bacterium, has been implicated in Alzheimer's disease (AD), yet its role in retinal pathology remains unexplored. We analyzed postmortem tissues from 95 human donors and found 2.9-4.1-fold increases in Cp inclusions in AD retinas and brains, with no significant elevation in mild cognitive impairment (MCI). Proteomics revealed dysregulation of retinal and brain bacterial infection-related proteins and NLRP3 inflammasome pathways. NLRP3 expression was markedly elevated in MCI and AD retinas, and its activation was evident by increased N-termin…

Identification of Chlamydia pneumoniae and NLRP3 inflammasome activation in Alzheimer's disease retina.

Emerging evidence implicates bacterial infections, including Chlamydia pneumoniae (Cp), a gram-negative obligate intracellular bacterium responsible for community-acquired pneumonia, in Alzheimer's disease (AD) pathogenesis. However, the involvement of Cp in early and advanced AD in the retina is unknown. Here, we identified the existence and distribution of intracellular Cp inclusions and related NLRP3 inflammasome activation and neurodegeneration in postmortem retinas and brains from 95 human donors. Histological analysis in neuropathologically-confirmed MCI and AD patients compared with cog…

Selective Assembly of TRPC Channels in the Rat Retina during Photoreceptor Degeneration.

Transient receptor potential canonical (TRPC) channels are calcium channels with diverse expression profiles and physiological implications in the retina. Neurons and glial cells of rat retinas with photoreceptor degeneration caused by retinitis pigmentosa (RP) exhibit basal calcium levels that are above those detected in healthy retinas. Inner retinal cells are the last to degenerate and are responsible for maintaining the activity of the visual cortex, even after complete loss of photoreceptors. We considered the possibility that TRPC1 and TRPC5 channels might be associated with both the hig…

Altered Functional Responses of the Retina in B6 Albino Tyrc/c Mice.

PURPOSE: Mammals with albinism present low visual discrimination ability and different proportions of certain retinal cell subtypes. As the spatial resolution of the retina depends on the visual field sampling by retinal ganglion cells (RGCs) based on the convergence of upstream cell inputs, it could be affected in albinism and thus modify the RGC function. METHODS: We used the Tyrc/c line, a mouse model of oculocutaneous albinism type 1 (OCA1), carrying a tyrosinase mutation, and previously characterized by a total absence of pigment and severe visual deficits. To assess their retinal functi…

Measuring the Release of Lactate from Wild-Type and rd1 Mouse Retina.

The retina has the highest energy consumption of any tissue in the human body. Remarkably, to satisfy its energy demand, the retina appears to rely mostly on aerobic glycolysis, which results in the production and release of large amounts of lactate. In the present study, we compared two different methods to assess lactate release from in vitro organotypic retinal explants cultured under entirely controlled, serum-free conditions. We used a standard lactate assay kit and 1H-nuclear magnetic resonance (NMR) spectroscopy-based analysis. We found that during the culturing of retinal explants deri…

Hyperreflective dots in the avascular outer retina in relapsing-remitting multiple sclerosis.

BACKGROUND: Hyperreflective granular elements with a transient presence in the retina can be detected non-invasively by optical coherence tomography (OCT). Such foci or dots may represent aggregates of activated microglia. However, in multiple sclerosis an increased number of hyperreflective foci has so far not been demonstrated in the intrinsically hyporeflective and avascular outer nuclear layer of the retina where there are no fixed elements in healthy eyes. Therefore, the present study intended to investigate the presence of hyperreflective foci in the outer nuclear layer in patients with …

Mechanism of low-frequency, low-intensity ultrasound modulation of the mouse retina.

Objective.Ultrasound has been shown to modulate the activity of retinal ganglion cells (RGCs) in mice, but the mechanism remains poorly understood. This study aims to address this question.Approach.Multi-electrode recordings together with pharmacological methods were used to investigate the possible cellular/circuitry mechanism(s) underlying the neuronal modulation induced by low-frequency (1 MHz), low-intensity (ISPTA0.5 W cm-2) ultrasound stimulation.Main results. We found that ultrasound activated mechanosensitive channels (transient receptor potential vanilloid 4 (TRPV4) channels are involv…

Mechanical Deformation of Peripapillary Retina in Response to Acute Intraocular Pressure Elevation.

Elevated intraocular pressure (IOP) may cause mechanical injuries to the optic nerve head (ONH) and the peripapillary tissues in glaucoma. Previous studies have reported the mechanical deformation of the ONH and the peripapillary sclera (PPS) at elevated IOP. The deformation of the peripapillary retina (PPR) has not been well-characterized. Here we applied high-frequency ultrasound elastography to map and quantify PPR deformation, and compared PPR, PPS and ONH deformation in the same eye. Whole globe inflation was performed in ten human donor eyes. High-frequency ultrasound scans of the poster…

The Mlc1 Promoter Directs Müller Cell-specific Gene Expression in the Retina.

PURPOSE: Because the importance of glia in regulating brain functions has been demonstrated, genetic technologies that manipulate glial cell-specific gene expression in the brain have become essential and have made great progress. However, it is unknown whether the same strategy that is used in the brain can be applied to the retina because retinal glia differs from glia in the brain. Here, we aimed to find a method for selective gene expression in Müller cells (characteristic glial cells in the retina) and identified Mlc1 as a specific promoter of Müller cells. METHODS: Mlc1-tTA::Yellow-Came…

Widespread retina and optic nerve neuroinflammation in enucleated eyes from glaucoma patients.

Neuroinflammation is recognized as a key component of neurodegenerative disease. In glaucoma, a common neurodegenerative disease and the leading cause of irreversible blindness, the evidence for neuroinflammation in patients is lacking. Animal models have demonstrated significant pro-inflammatory activation of resident glia in the retina, as well as influx of blood-derived monocytes and pro-inflammatory factors. Confirmation of this in human donor tissue has been challenging due to a lack of well-preserved and well-characterized post-mortem tissue. To address this we utilize archived, wax embe…

Multimodal imaging including optical coherence tomography angiography of benign familial fleck retina.

A 44-year-old woman presented with complaints of pain in the right eye (RE). Fundus examination revealed disc edema in the RE along with retinal flecks sparing the macula in both eyes (BE). Fundus autofluorescence demonstrated a symmetrical pattern of white flecks in BE. Spectral-domain optical coherence tomography (SD-OCT) revealed the lesions at the level of retinal pigment epithelium with impingement onto the outer retina. SD-OCT angiography through the flecks revealed hyperreflective lesions at the level of avascular retina. RE B-scan revealed a T-sign. Based on these findings, she was dia…
