Induction of spermatogenesis in men with azoospermia or severe oligoteratoasthenospermia after antegrade internal spermatic vein sclerotherapy for the treatment of varicocele.

AIM: To evaluate the treatment outcome of antegrade internal spermatic vein sclerotherapy in men with non-obstructive azoospermia or severe oligoteratoasthenospermia (OTA) as a result of varicocele. METHODS: Between September 1995 and January 2004, 47 patients (mean age 33.8 +/- 6.3 years) underwent antegrade internal spermatic vein sclerotherapy for the treatment of varicocele with azoospermia (14 patients) or severe OTA (33 patients). Testicular core biopsy was also performed in complete azoospermic patients who provided informed consent. The outcome was assessed in terms of improvement in

Effects of paternal heat stress on the in vivo development of preimplantation embryos in the mouse.

The objective of this study was to examine the effect of paternal heat stress on the in vivo development of preimplantation embryos in the m … pididymis and germ cells in the testis are susceptible to damage by environmental heat stress, with spermatocytes being the most vulnerable. This study also demonstrates that subtle effects on the male such as a short exposure to elevated environmental temperatures can translate to quite profound paternal impacts on early embryo development.

The value of quantitative DNA flow cytometry of testicular fine-needle aspirates in assessment of spermatogenesis: a study of 137 previously maldescended human testes.

In order to assess the suitability of DNA flow cytometry of fine-needle aspirates for quantifying spermatogenesis, the results from DNA flow cytometry were compared to histological evaluation of testicular biopsies taken concomitantly from 171 previously maldescended testes. In 137 of 171 cases, sufficient material for flow cytometric as well as histological evaluation was obtained. Histological analysis of surgical biopsy specimens revealed spermatogenesis including the spermatid stage in 117 of the 137 gonads. In six of the 117 gonads no haploid cells were found using flow cytometry. On the

Changes in brain dopamine levels, oocyte growth and spermatogenesis in rainbow trout, Oncorhynchus mykiss, following sublethal cyanide exposure.

Whole brain dopamine (DA) and norepinephrine (NE) levels were measured in sexually maturing (2 years +) male and female rainbow trout, Oncorhynchus mykiss, following exposure to 0.01 mg/L hydrogen cyanide (HCN). Following a 12 day exposure period in July and August 1988, whole brain DA levels of HCN exposed fish were significantly higher (p less than 0.05), relative to control fish, as measured by high performance liquid chromatography (HPLC). Brain NE levels were unaffected by HCN exposure. Whole brain DA and NE levels showed a strong correlation in control fish (r = +0.81), but not in HCN ex

Antifertility effects of a potent LH-RH antagonist in male and female rats.

The antifertility effects of the antagonistic analogue N-Ac-D-p-Cl-Phe1,2,D-Trp3,D-Arg6,D-Ala10-LH-RH (ORG 30276) were investigated in male and female rats. Male rats were treated with ORG 30276 at doses of 10 micrograms/kg, 100 micrograms/kg, or 1000 micrograms/kg per day for 60 days. The control rats were injected with the vehicle only. The treatment with the highest dose of the antagonist caused a significant decrease in the weights of the anterior pituitaries, testes, seminal vesicles, and ventral prostates and reduced serum and pituitary LH levels. The intermediate dose of the antagonist

Ethylene glycol monomethyl ether altered rat sperm small RNAs with critical developmental roles.

Ethylene glycol monomethyl ether (EGME) is a testicular germ cell toxicant that selectively targets spermatocytes. In rats, male-only EGME exposure reduces mating success and can lead to an increase in resorbed fetuses. In a previous study, 5-d exposure to 50, 60, or 75 mg/kg/d EGME in male rats led to a decrease in sperm motility and increase in retained spermatid heads with a lowest observed adverse effect level (LOAEL) of 75 mg/kg/d. At 60 mg/kg/d, EGME exposure altered the proportion of sperm small RNA reads mapped to different small RNA categories and the distribution of read lengths. To …

Sequence characteristics, expression pattern, ovarian maintenance function and action mechanism of SIRT1 in Chlamys farreri.

SIRT1 (silent mating type information regulation 2 homolog 1), a histone deacetylase in Sirtuin family, regulates ovarian development and aging in higher mammals, but its role in invertebrates remains unexplored. This study investigated SIRT1 in the scallop Chlamys farreri. Its 2382 bp CDS (coding sequence) encodes 793 amino acids with the NTERM activation motif, the catalytic activity center and the ESA domain. QRT-PCR revealed that SIRT1 expression level in gonads was significantly higher than that in other tissues. In ovary, SIRT1 expression was the highest at growing stage, in the testes,

Proteomic analysis of the zone of degeneration at the mitosis-meiosis transition stage in wild-caught male catshark (Scyliorhinus canicula), correlated with an unusually high-water temperature in the English Channel.

In the context of current global change, variations in water temperature are one of the environmental conditions with serious consequences for marine life, including reproductive processes. In the small spotted catshark Scyliorhinus canicula, spermatogenesis occurs in spermatocysts composed of synchronously developing germ cells associated with Sertoli cells, forming a zonal arrangement of the spermatogenic wave. Male catsharks are known to show little to no seasonal variation in spermatogenesis, unlike other sharks that may have a seasonal break at the spermatogonia-primary spermatocyte trans

Mechanism of PP2A affecting ubiquitination pathway in spermatogenesis.

The heterotrimeric complex serine-threonine protein phosphatase 2 A (PP2A) is integral to the regulation of essential cellular processes. It is particularly crucial in spermatogenesis, where it is indispensable for meiosis, mitosis, sperm capacitation, and apoptosis. Previous research has concentrated on the knockdown of the catalytic subunit of PP2A, PPP2CA, in germ cells using Ddx4-Cre, resulting in male mouse sterility, disrupted meiotic recombination, and meiotic arrest of spermatocytes. To further elucidate the role of PP2A in spermatogenesis, we performed transcriptomic and proteomic seq…

Safety evaluation of ethanolic extract from aerial flowering part of spiny globe thistle (Echinops spinosus) in mice: Phytochemical screening and genotoxicity.

Echinops spinosus is widely used by the population due to its therapeutic potential; however, there is no evidence in the literature that substantiates its safety. Therefore, this study aimed to identify the chemical constituents of E. spinosus extract via GC/MS analysis and evaluate its cytotoxicity and genotoxicity. Male mice were orally given three doses of E. spinosus extract (250, 500, and 1000 mg/kg) for four weeks. Blood and tissue samples were collected after the end of treatment. GC-MS results revealed 73 compounds in the E. spinosus extract, including sugars, sugar alcohols, fatty ac

Loss of full-length DAZL isoform disrupts PABPC1-dependent translational regulation and meiosis.

Alternative splicing (AS) events are exceptionally active during spermatogenesis, enhancing the diversity of the testicular transcriptome and proteome. In mouse testes, the germ cell-specific RNA-binding protein DAZL undergoes alternative splicing to produce two isoforms: a full-length DAZL (DAZL_FL) and a short isoform lacking exon 8 (DAZL_Sh). While DAZL is a hallmark of germ cell development, the physiological roles of its alternative splicing, and the distinct functions of these two isoforms remain yet to be fully elucidated. To investigate this, we disrupted alternative splicing of Dazl b
