Association between TLR4 polymorphism and periodontitis susceptibility: a meta-analysis.

TLR4 has been implicated in periodontal disease, but the association between the TLR4 Asp299Gly and Thr399Ile polymorphisms and the risk of periodontal disease remains unclear. Therefore, the aim of this study was to investigate the association between the TLR4 Asp299Gly and Thr399Ile polymorphism and periodontal disease. A search of electronic databases identified previous studies evaluating the association of the polymorphisms of TLR4 and periodontitis risk. The association was evaluated by odds ratio (OR) and its 95% confidence interval (CI). The results showed that TLR4 Asp299Gly and Thr39

Total colonic aganglionosis in Hirschsprung disease.

Total colonic aganglionosis (TCA) is a relatively uncommon form of Hirschsprung disease (HSCR), occurring in approximately 2%-13% of cases. It can probably be classified as TCA (defined as aganglionosis extending from the anus to at least the ileocecal valve, but not >50 cm proximal to the ileocecal valve) and total colonic and small bowel aganglionosis, which may involve a very long segment of aganglionosis. It is not yet clear whether TCA merely represents a long form of HSCR or a different expression of the disease. There are many differences between TCA and other forms of HSCR, which requi

Reconstruction and expression of the MRI-contrast protein, ferritin, with recombinant rabies vectors.

Attenuated recombinant rabies vector could be an ideal system for delivery of contrast agent gene for Magnetic Resonance Imaging (MRI) because of its neurotropic nature. In this study, the gene of a biomolecular contrast agent, ferritin, was successfully cloned into two rabies virus vectors, vaccine-based pCTN and street strain-based pNH. Recombinant virus granules were obtained and proved to express ferritin by RT-PCR after transfection of CTN-ferritin and NH-ferritin vector systems in BHK-21 cells. The recovered rabies virus-rCTN-ferritin was of similar ability to rNH-ferritin, which suggest

Pseudotype-dependent lentiviral transduction of astrocytes or neurons in the rat substantia nigra.

Gene transfer to the central nervous system provides powerful methodology for the study of gene function and gene-environment interactions in vivo, in addition to a vehicle for the delivery of therapeutic transgenes for gene therapy. The aim of the present study was to determine patterns of tropism exhibited by pseudotyped lentiviral vectors in the rat substantia nigra, in order to evaluate their utility for gene transfer in experimental models of Parkinson's disease. Isogenic lentiviral vector particles encoding a GFP reporter were pseudotyped with envelope glycoproteins derived from vesicula

Efficient transduction of non-human primate motor neurons after intramuscular delivery of recombinant AAV serotype 6.

Retrograde transport of viral vectors in the rodent spinal cord provides a powerful means to administer a therapeutic transgene from the innervated musculature. With the aim of scaling up this approach to non-human primates, we have injected recombinant adeno-associated vectors (rAAV) serotype 6 expressing enhanced green fluorescent protein (eGFP) into the gastrocnemius muscle of African green monkeys to determine whether this results in efficient transgene delivery to lumbar motor neurons. Cells expressing eGFP were observed across more than 1 cm of the spinal cord 4 weeks after intramuscular

Enhanced transduction of colonic cell lines in vitro and the inflamed colon in mice by viral vectors, derived from adeno-associated virus serotype 2, using virus-microbead conjugates bearing lectin.

BACKGROUND: Virus-mediated delivery of therapeutic transgenes to the inflamed colon holds a great potential to serve as an effective therapeutic strategy for inflammatory bowel disease, since local, long-term expression of the encoded therapeutic proteins in the colorectal system is potentially achievable. Viral vectors, derived from adeno-associated virus (AAV), should be very useful for such therapeutic strategies, particularly because they can establish long-term expression of transgenes. However, few studies have been carried out to investigate the ability of AAV-based vectors to transduce

Regulatory T cells induced by rAAV carrying the forkhead box P3 gene prevent autoimmune thyroiditis in mice.

CD4+CD25+ regulatory T (Treg) cells are immunosuppressive and help maintain peripheral immune tolerance. The forkhead/winged helix family member, Foxp3, has been shown to be a critical regulator of CD4+CD25+ Treg cells. We demonstrate by quantitative real-time PCR that CD4+CD25+ T cells express higher levels of Foxp3 mRNA than other T cell populations. Recombinant adeno-associated virus vector carrying mouse Foxp3 gene under the control of the CMV promoter was generated and used to transduce CD4+CD25- T cells. These Foxp3-transduced T cells are similar to naturally occurring CD4+CD25+ regulato

Ex vivo adenovirus-mediated gene delivery leads to long-term expression in pancreatic islet transplants.

BACKGROUND: Replication-deficient adenovirus, one of the most efficient vectors in gene therapy, has been limited by transient transgene expression due to its episomal location and loss during cell division, as well as a host immune response against viral proteins. METHODS: Murine pancreatic islets were infected ex vivo with ad5-cytomegalovirus (CMV)-beta-galactosidase and transplanted into diabetic recipients with normalization of glucose metabolism. RESULTS: High levels of beta-galactosidase activity were detectable histologically for at least 20 weeks after transplant, and beta-galactosid

Enhancing in vivo B cell gene delivery using targeted lentiviral vectors and transduction enhancers.

B cells occupy a distinct niche in humoral immunity, and engineered B cells that leverage their unique biological functions hold significant promise for therapeutic applications. Here, we combine chemical and virological strategies to establish a method for in situ engineering of B cells by optimizing formulations of CD20-targeted Nipah paramyxovirus-pseudotyped lentiviral vectors (LVs) coupled with the use of transduction enhancers (TEs). Select TE combinations enabled greater than 3-fold improvements in transduction efficiency of isolated B cells as well as B cells within human PBMC populati

OsWOX13 functions as a heading signal transducer from integrator Ehd1 to florigen for the accurate regulation of rice heading date.

Appropriate heading date is an important safeguard for stabilizing rice yield and quality. Heading integrator Ehd1 and florigen Hd3a are crucial for rice heading. However, the mechanism by which Ehd1 transmits heading signals to Hd3a and the accurate regulation of Hd3a expression remains unclear. Here, we found that OsWOX13 functions as a signal transducer from Ehd1 to Hd3a for the accurate regulation of rice heading date. Ehd1 can directly bind to the OsWOX13 promoter, OsWOX13, in turn, binds to the Hd3a promoter to activate its expression. OsWOX13 interacts with Hd3a to inhibit OsWOX13-media

Fungal Infections in Disorders of Inborn Errors of Immunity.

Emerging infectious threats continually challenge the adaptability of the human immune system. Inborn errors of immunity (IEIs) provide a unique framework to dissect host-pathogen interactions, revealing how single-gene defects uncover critical pathways in antifungal defence. This review offers an overview of fungal immunity through the lens of IEIs, and integrates recent advances in innate fungal recognition, neutrophil and mononuclear phagocyte effector mechanisms, CARD9/NF-kB signalling, IL-17/IL-23 and IL-12/IFN-gamma cytokine axes, and transcription-factor defects that shape susceptibilit

The Role of SAMHD1 in Viral Resistance and Transduction Efficiency Challenges in Pediatric Hematological Malignancies: Mechanistic Insights and Clinical Perspectives.

Sterile alpha motif and HD domain-containing protein 1 (SAMHD1) is a dNTPase that regulates intracellular nucleotide pools, preserves genomic stability, and mediates intrinsic antiviral immunity. While its role in adult hematologic malignancies is established, its implications for pediatric leukemia-particularly acute myeloid leukemia (AML) and acute lymphoblastic leukemia (ALL)-remain underexplored. This review integrates transcriptomic, protein interaction, and immune correlation analyses to examine the dual role of SAMHD1 in pediatric hematological malignancies: as an antiviral restriction

Elucidating the molecular symphony: unweaving the transcriptional & epigenetic pathways underlying neuroplasticity in opioid dependence and withdrawal.

The persistent use of opioids leads to profound changes in neuroplasticity of the brain, contributing to the emergence and persistence of addiction. However, chronic opioid use disrupts the delicate balance of the reward system in the brain, leading to neuroadaptations that underlie addiction. Chronic cocaine usage leads to synchronized alterations in gene expression, causing modifications in the Nucleus Accumbens (NAc), a vital part of the reward system of the brain. These modifications assist in the development of maladaptive behaviors that resemble addiction. Neuroplasticity in the context

Genetic Modification of the AAV5 Capsid with Lysine Residues Results in a Lung-Tropic Liver-Detargeted Gene Transfer Vector.

Intravenous (IV) administration of naturally occurring adeno-associated virus (AAV) vectors are liver tropic, with a significant proportion of the total vector dose mediating gene expression in liver hepatocytes. AAV capsids that are directed toward other organs such as lung may be useful for therapy of nonliver-based diseases. Based on the knowledge that the lung capillary endothelium is the first capillary bed encountered by an intravenously administered AAV vector, and that the lung endothelium glycocalyx is enriched in negatively charged sialic acid, we hypothesized that adding positively

Enhanced CNS transduction from AAV.PHP.eB infusion into the cisterna magna of older adult rats compared to AAV9.

The development of high efficiency, central nervous system (CNS) targeting AAV-based gene therapies is necessary to address challenges in both pre-clinical and clinical investigations. The engineered capsids, AAV.PHP.B and AAV.PHP.eB, show vastly improved blood-brain barrier penetration compared to their parent serotype, AAV9, but with variable effect depending on animal system, strain, and delivery route. As most characterizations of AAV.PHP variants have been performed in mice, it is currently unknown whether AAV.PHP variants improve CNS targeting when delivered intrathecally in rats. We eva

Genes affecting progression of bacteriophage P22 infection in Salmonella identified by transposon and single gene deletion screens.

Bacteriophages rely on their hosts for replication, and many host genes critically determine either viral progeny production or host success via phage resistance. A random insertion transposon library of 240,000 mutants in Salmonella enterica serovar Typhimurium was used to monitor effects of individual bacterial gene disruptions on bacteriophage P22 lytic infection. These experiments revealed candidate host genes that alter the timing of phage P22 propagation. Using a False Discovery Rate of < 0.1, mutations in 235 host genes either blocked or delayed progression of P22 lytic infection, inclu

Advances in molecular genetics of Hirschsprung's disease.

Hirschsprung's disease (HSCR) is a developmental disorder of the enteric nervous system, which occurs due to the failure of neural crest cells to fully colonize the gut during embryonic development. It is characterized by the absence of the enteric ganglia in a variable length of the intestine. Substantial progress has been made in understanding the genetic basis of HSCR with the help of advanced genetic analysis techniques and animal models. More than 11 genes have been found to be involved in the pathogenesis of HSCR. The RET gene is the most important susceptibility gene involved in HSCR wi
