Preparation and immunogenicity studies of NvIBDV VP2-ferritin nanoparticles.

BACKGROUND: Infectious bursal disease (IBD), caused by infectious bursal disease virus (IBDV), is a highly contagious disease that is prevalent worldwide and poses a significant threat to the poultry industry. While commercially available vaccines are used for prevention, IBD outbreaks remain frequent. OBJECTIVE: The continuous mutation of virulent strains and their ability to evade traditional vaccine protection complicate IBD control, which necessitates the development of novel vaccines and a deeper understanding of viral mutation mechanisms. METHOD: Utilizing the self-assembly capability

Construction and efficacy evaluation of novel swine leukocyte antigen (SLA) class I and class II allele-specific poly-T cell epitope vaccines against porcine reproductive and respiratory syndrome virus.

Porcine reproductive and respiratory syndrome virus (PRRSV) causes an economically important global swine disease. Here we report the development of subunit PRRSV-2 vaccines by expressing swine leucocyte antigen (SLA) class I and class II allele-specific epitope antigens in a robust adenovirus vector. SLA I-specific CD8 and SLA II-specific CD4 T cell epitopes of PRRSV-2 NADC20 were predicted in silico. Stable murine leukaemia cell lines (RMA-S), which are TAP-deficient and lacking endogenous class I epitope loading, were established to express different SLA I alleles. The binding stability of

Immunization with recombinant Brucella species outer membrane protein Omp16 or Omp19 in adjuvant induces specific CD4+ and CD8+ T cells as well as systemic and oral protection against Brucella abortus infection.

Available vaccines against Brucella spp. are live attenuated Brucella strains. In order to engineer a better vaccine to be used in animals and humans, our laboratory aims to develop an innocuous subunit vaccine. Particularly, we are interested in the outer membrane proteins (OMPs) of B. abortus: Omp16 and Omp19. In this study, we assessed the use of these proteins as vaccines against Brucella in BALB/c mice. Immunization with lipidated Omp16 (L-Omp16) or L-Omp19 in incomplete Freund's adjuvant (IFA) conferred significant protection against B. abortus infection. Vaccination with unlipidated Omp

mRNA Vaccines Targeting the Conserved SARS-CoV-2 Fusion Machinery Elicit Cross-Variant and Robust Immunity.

BACKGROUND/OBJECTIVES: Despite the gradual waning of global interest in SARS-CoV-2, the continued evolution and emergence of viral variants underscore the need for broadly protective vaccines. Vaccines targeting the highly mutable S1 subunit of the spike protein have shown limited breadth of protection, whereas conserved regions within the S2 fusion machinery remain critical targets for the development of vaccines effective against future variants. This study evaluated two mRNA-lipid nanoparticle (mRNA-LNP) vaccines encoding HR121, a conserved HR1-HR2-HR1 fusion-intermediate immunogen, in comp

Recent Progress and Challenges in the Development of Edible Vaccines Produced by Genetically Modified Plants.

Biotechnologists have pioneered the idea of an edible vaccination in recent years. Subunit vaccines, such as those used to create edible vaccines, involve the introduction of certain genes into transgenic plants, which are subsequently coaxed into producing the corresponding protein. Bananas, potatoes, legumes, lettuce, soybeans, corn, and rice are all examples of foods that fall under this category. They have a low unit cost, can be stored conveniently, and are simple to administer to patients of varying ages. There is great hope that the use of edible vaccinations, particularly in underdevel

Immunogenicity of the tuberculosis vaccine MVA85A is reduced by coadministration with EPI vaccines in a randomized controlled trial in Gambian infants.

New tuberculosis vaccines are urgently needed to curtail the current epidemic. MVA85A is a subunit vaccine that could enhance immunity from BCG vaccination. To determine MVA85A safety and immunogenicity as well as interactions with other routine vaccines administered in infancy, we randomized healthy 4-month-old infants who had received Bacille Calmette-Guérin at birth to receive Expanded Program on Immunization (EPI) vaccines alone, EPI and MVA85A simultaneously, or MVA85A alone. Adverse events were monitored throughout. Blood samples obtained before vaccination and at 1, 4, and 20 weeks afte

A review of the current status of enteric vaccines.

Much progress has been made in developing vaccines against the most important enteric infections. Two new vaccines against typhoid fever (oral Ty21a and parenteral Vi polysaccharide) have been licensed in many countries. Newer, more sophisticated typhoid vaccines undergoing clinical testing include recombinant attenuated Salmonella typhi strains and Vi polysaccharide-carrier-protein conjugate vaccines. Two inactivated oral cholera vaccines, consisting of inactivated Vibrio cholerae O1 bacteria alone or in combination with B subunit of cholera toxin, each conferred 50-53% protection over three

Anti-laminin-332 mucous membrane pemphigoid developing after a diphtheria tetanus vaccination.

IMPORTANCE: Bullous pemphigoid (BP) has been previously described to develop after vaccination in 26 patients. Immunoblotting or enzyme-linked immunosorbent assays (ELISAs), which were performed for 7 of these patients, have always shown circulating autoantibodies against BP180 and/or BP230 antigens. A case of anti-laminin-332 mucous membrane pemphigoid (MMP) that developed shortly after a diphtheria tetanus vaccination is described, with a review of the literature on postvaccination BP. OBSERVATIONS: A 29-year-old man developed an acute eruption of oral and cutaneous blisters and erosions 2

Review on anticoccidial vaccine development for chickens: Past progress, modern methods, and production bottlenecks.

Poultry coccidiosis is an economically significant intestinal disease caused by parasitic protozoa in the genus Eimeria. Developing Eimeria vaccines is a possible way to control and prevent coccidiosis. This review discusses the different methods and challenges of anticoccidia vaccine production for poultry. The production of anticoccidial vaccines involves several methods with challenges. Live Eimeria vaccines are produced using non-attenuation without any alteration or modifications in terms of pathogenicity, and attenuation methods reduce virulence through serial passage in embryonated eggs

Development of a single-dose Q fever vaccine with an injectable nanoparticle-loaded hydrogel: effect of sustained co-delivery of antigen and adjuvant.

Q fever is a zoonotic infectious disease caused by Coxiella burnetii, and there is currently no FDA-approved vaccine for human use. The whole-cell inactivated vaccine Q-VAX, which is only licensed in Australia, has a risk of causing severe adverse reactions, making subunit vaccines a good alternative. However, most subunit antigens are weak immunogens and require two or more immunizations to elicit an adequate level of immunity. We hypothesized that by combining a nanoparticle to co-deliver both a protein antigen and an adjuvant, together with a hydrogel depot for sustained-release kinetics, a

Recombinant African swine fever virus p30-flagellin fusion protein promotes p30-specific humoral and cellular immune responses in mice.

African swine fever (ASF) is a transmissible and deadly viral disease caused by the African swine fever virus (ASFV) that has considerably affected the global pig industry. Vaccination is considered a potentially effective method to control ASF. However, live attenuated vaccines can't protect against all circulating virus isolates. Subunit vaccines can induce both cellular and humoral immune responses, but often require the addition of adjuvants. Flagellin, a stimulator of Toll-like receptor 5 (TLR5), functions as a potent adjuvant by enhancing cellular and humoral immune responses. However, i

A conserved motif in the immune-subdominant RAP-1 related antigen of Babesia bovis contains a B-cell epitope recognized by antibodies from protected cattle.

INTRODUCTION: Babesia bovis, a tick-borne apicomplexan parasite causing bovine babesiosis, remains a significant threat worldwide, and improved and practical vaccines are needed. Previous studies defined the members of the rhoptry associated protein-1 (RAP-1), and the neutralization-sensitive rhoptry associated protein-1 related antigen (RRA) superfamily in B. bovis, as strong candidates for the development of subunit vaccines. Both RAP-1 and RRA share conservation of a group of 4 cysteines and amino acids motifs at the amino terminal end (NT) of these proteins. METHODS AND RESULTS: Sequence

Vaccinating patients with autoimmune diseases.

PURPOSE: Vaccine preventable diseases, especially respiratory infections, occur at higher frequency in patients with rheumatic diseases who are immunosuppressed. The focus of this review is to highlight vaccinations that patients with rheumatic diseases should optimally receive, with a focus on efficacy and safety of vaccines. MAIN FINDINGS: In general, vaccines are effective in reducing the burden of infection. However, due to underlying immunosuppression, their efficacy is most likely reduced compared to the general population. This is even true of the novel vaccines, mRNA and subunit vacci

Approaches to Next-Generation Capripoxvirus and Monkeypox Virus Vaccines.

Globally, there are two major poxvirus outbreaks: mpox, caused by the monkeypox virus, and lumpy skin disease, caused by the lumpy skin disease virus. While vaccines for both diseases exist, there is a need for improved vaccines. The original vaccines used to eradicate smallpox, which also protect from the disease now known as mpox, are no longer acceptable. This is mainly due to the risk of serious adverse events, particularly in HIV-positive people. The next-generation vaccine for mpox prevention is modified vaccinia Ankara, which does not complete the viral replication cycle in humans and,

Advances in clinical immunogenicity evaluation of influenza vaccines.

Influenza vaccination is an effective intervention for preventing severe influenza, and assessing its immunogenicity is a critical step in determining vaccine protective efficacy. With the widespread use of influenza vaccines-including split-virion, subunit, recombinant protein, and live attenuated formulations-methods for evaluating immunogenicity have grown increasingly diverse and complex. This article systematically examines the advantages and limitations of various immunogenicity assessment approaches for influenza vaccines. It draws upon multidimensional evaluation frameworks covering hu

Current status on encephalitic alphavirus vaccines development: Advances, challenges, and global health perspectives.

Alphaviruses are a globally distributed group of arboviruses responsible for two distinct disease manifestations, namely inflammatory polyarthritis and encephalitis. Among the arthritogenic viruses, chikungunya virus (CHIKV) is the most prevalent and has caused epidemics in 119 countries. Notably, two human vaccines against CHIKV have now received FDA approval. In contrast, the encephalitic alphaviruses, including Venezuelan equine encephalitis virus (VEEV), eastern equine encephalitis virus (EEEV), and western equine encephalitis virus (WEEV), are associated with substantially higher case fat

Plant-Derived Recombinant Vaccines against Zoonotic Viruses.

Emerging and re-emerging zoonotic diseases cause serious illness with billions of cases, and millions of deaths. The most effective way to restrict the spread of zoonotic viruses among humans and animals and prevent disease is vaccination. Recombinant proteins produced in plants offer an alternative approach for the development of safe, effective, inexpensive candidate vaccines. Current strategies are focused on the production of highly immunogenic structural proteins, which mimic the organizations of the native virion but lack the viral genetic material. These include chimeric viral peptides,

Fowl adenovirus serotype 4: Epidemiology, pathogenesis, diagnostic detection, and vaccine strategies.

Fowl adenovirus (FAdV) serotype-4 is highly pathogenic for chickens, especially for broilers aged 3 to 5 wk, and it has emerged as one of the foremost causes of economic losses to the poultry industry in the last 30 years. The liver is a major target organ of FAdV-4 infections, and virus-infected chickens usually show symptoms of hydropericardium syndrome. The virus is very contagious, and it is spread both vertically and horizontally. It can be isolated from infected liver homogenates and detected by several laboratory diagnostic methods (including an agar gel immunodiffusion test, indirect i

Protective Efficacy of Vaccines Against Grass Carp Hemorrhagic Disease: A Systematic Review and Meta-Analysis.

Grass carp hemorrhagic disease, mainly caused by grass carp reovirus (GCRV), is a major threat to grass carp aquaculture, and vaccination is considered a sustainable strategy for disease prevention and control. However, the protective efficacy of GCRV vaccines and the factors influencing it have not been systematically synthesised. In this study, literature on GCRV vaccines was retrieved from CNKI, ScienceDirect, and Web of Science in accordance with the PRISMA 2020 statement, covering database inception to March 10, 2026, followed by meta-analysis. After screening, 44 studies were included, o

A systematic review and expert Delphi Consensus recommendation on the use of vaccines in patients receiving dupilumab: A position paper of the American College of Allergy, Asthma and Immunology.

BACKGROUND: Dupilumab is a monoclonal antibody that targets the interleukin (IL)-4 receptor alpha subunit, thus blocking the effects of IL-4 and IL-13, and has shown efficacy in treating various conditions including asthma, atopic dermatitis, eosinophilic esophagitis, and others. Because of its immune modulatory effects, clinical trials that studied dupilumab did not allow patients to receive live vaccines during the clinical trials because of an abundance of caution, and thus package inserts recommend that patients who are being treated with dupilumab should avoid live vaccines. Because dupil
