Quantitative cellular changes in multiple system atrophy brains.

Multiple system atrophy (MSA) is a neurodegenerative disorder characterised by a combined symptomatology of parkinsonism, cerebellar ataxia, autonomic failure and corticospinal dysfunction. In brains of MSA patients, the hallmark lesion is the aggregation of misfolded alpha-synuclein in oligodendrocytes. Even though the underlying pathological mechanisms remain poorly understood, the evidence suggests that alpha-synuclein aggregation in oligodendrocytes may contribute to the neurodegeneration seen in MSA. The primary aim of this review is to summarise the published stereological data on the to…

Amyotrophic lateral sclerosis with dementia: the clinicopathological spectrum.

Amyotrophic lateral sclerosis with dementia (ALS-D) is a non-Alzheimer-type dementia characterized by both frontotemporal degeneration and motor neuron disease and marked by ubiquitin-positive, tau- and alpha-synuclein-negative intraneuronal inclusions and dystrophic neurites. New neuropathological diagnostic criteria for ALS-D are proposed on the basis of the present investigation of 28 autopsy cases. Clinical features included those of typical ALS-D, primary lateral sclerosis, atypical ALS with frontotemporal atrophy and atypical Pick's disease without Pick's bodies. Macroscopically anterior…

Internalized SNCA/α-synuclein fibrils become truncated and resist degradation in neurons while glial cells rapidly degrade SNCA fibrils.

Parkinson disease (PD) and other α-synucleinopathies are characterized by the intracellular aggregates of SNCA/α-synuclein (synuclein, alpha) thought to spread via cell-to-cell transmission. To understand the contributions of various brain cells to the spreading of SNCA pathology, we examined the metabolism of SNCA aggregates in neuronal and glial cells. In neurons, while the full-length SNCA rapidly disappeared following SNCA pre-formed-fibril (PFF) uptake, truncated SNCA accumulated with a half-life of days rather than hours. Epitope mapping and fractionation studies indicate that SNCA fibri

Proteomic analysis of heat-stable proteins revealed an increased proportion of proteins with compositionally biased regions.

Intrinsically disordered proteins (IDPs) have been in the spotlight for their unique properties, such as their lack of secondary structures and low sequence complexity. Alpha-synuclein and tau are representative disease-related IDPs with low complexity regions in their sequences, accumulating in the brains of patients with Parkinson disease and Alzheimer disease, respectively. Their heat resistance in particular was what attracted our attention. We assumed that there exist many other unidentified proteins that are resistant to heat-treatment, referred to as heat-stable proteins, which would al…

Coordination of Anle138b to Silver Results in Selective Reduction of a C-Terminal Truncated α-Synuclein Protein and Increased Aggregate Size.

Parkinson's disease (PD) is a prevalent age-related neurodegenerative syndrome, partially thought to be caused by a decrease in α-synuclein proteostasis. Anle138b = 5-(1,3-benzodioxol-5-yl)-3-(3-bromophenyl)-1H-pyrazole (HL) is undergoing clinical trials as a promising mitigator of α-synuclein aggregation. Because complexation to metals is known to modulate the activity of several drugs, we have prepared and characterized: H2L(ClO4), [CuI(µ-L)]3, and [AgI(µ-L)]3. To better understand the bioviability of these compounds, we monitored their effects in a cell culture model of α-synuclein protein

Authentic Phosphorylation of α-Synuclein at Ser129 Reveals Functional Differences Not Captured by the S129D Phosphomimetic.

Phosphorylation at serine 129 (pS129) is a dominant post-translational modification of α-synuclein (αSyn) and a widely used pathological marker in Parkinson's disease, yet its mechanistic consequences remain debated across physiological and pathological contexts. Most studies rely on phosphomimetic substitutions such as S129D, which approximate net charge but do not reproduce the steric, geometric, or hydrogen-bonding properties of authentic phosphorylation. Here, we establish a robust bacterial coexpression platform that generates homogeneous, site-specifically phosphorylated αSyn using its n

Proteoform-Resolved Cross-Linking Reveals Environment-Dependent Structural Effects of α-Synuclein S129 Phosphorylation.

Intrinsically disordered proteins (IDPs) drive many neurodegenerative disorders, but their structures remain difficult to define because they populate dynamic ensembles that change with the chemical environment. This problem is central for α-synuclein (aSyn), a Parkinson's disease-linked IDP in which S129 phosphorylation is highly enriched in disease-associated aggregates. Although S129 phosphorylation stabilizes a more compact aSyn ensemble in dilute solution, whether this structural effect persists across other biochemical environments has remained unknown. Here, we developed a quantitative

Decode the Ubiquitinome in Parkinson's Disease: From Pathological Aggregates to Targeted DUB Therapeutics.

Parkinson's disease (PD), a neurodegenerative disorder, is significantly influenced by genetic predispositions, aging, and environmental factors. Central to PD pathology are mechanisms such as aberrant α-synuclein aggregation, mitochondrial dysfunction, oxidative stress, neuroinflammation, and ferroptosis, all of which are closely associated with dysregulated protein post-translational modifications. Ubiquitination, a critical reversible modification, acts as a pivotal bridge connecting the ubiquitin-proteasome system and the lysosomal-autophagy pathway, with its dynamics finely counterbalance

α-Synuclein Promotes Atherosclerosis by Impairing Macrophage Autophagic Flux.

Atherosclerosis, recognized as a prototypical age-associated vascular disease, is driven by macrophage foam cell formation. Individuals with Parkinson's disease (PD) exhibit an elevated risk for stroke. While α-synuclein (α-Syn) is known for its neuronal role in PD, its presence in the circulation and potential vascular effects remain unexplored. Here, we identify α-Syn as a previously unrecognized circulating factor that promotes atherosclerosis. α-Syn was detected in human atherosclerotic plaques. Genetic deletion of α-Syn reduced atherosclerotic lesion size and macrophage lipid accumulation

Zero-Dalton Resolution in Nanopore Peptide Recognition.

Achieving zero-dalton resolution─the discrimination of biomolecules with identical mass─remains a fundamental challenge in molecular analysis. Nanopore sensing can resolve short peptide isomers, but whether discrimination persists when peptide length approaches the nanopore saturation limit is unclear. Here, we demonstrate robust nanopore-based discrimination of two 13-residue α-synuclein isomers using wild-type aerolysin at lengths approaching full nanopore sensing region occupancy. Isomer separation requires full peptide accommodation within the nanopore and arises from intrinsic structural

Residue-specific early-stage aggregation dynamics of α-synuclein (61-95) in monolayers: A pMAIRS and AFM study.

To better understand the misfolding behavior of α-synuclein (α-syn) at a controlled amphiphilic interface that models key features of membrane-associated confinement, we employed p-polarized Multiple-Angle Incidence Resolution Spectrometry (pMAIRS) in combination with Langmuir-Blodgett monolayer methods. The pMAIRS spectra were acquired at different time points during film compression to monitor the temporal evolution of supramolecular aggregation of the α-syn segment corresponding to residues 61-95 (α-syn(61-95)) at the air-water interface. Initially, α-syn(61-95) adopts an α-helical conforma

Removing the noise: endotoxin-free α-synuclein for accurate in vitro screening of fibrillation modulators.

The presence of lipopolysaccharide (LPS) contaminant from the vector expression bacteria Escherichia coli was described as a factor responsible for accelerating the in vitro fibrillation of recombinant α-synuclein (αS), a key protein involved in the neuropathogenesis of Parkinson's disease. Herein, we describe an efficient protocol for the removal of LPS from αS samples based on phase separation using Triton X-114. Importantly, this method was more feasible, faster and cheaper in eliminating LPS than the commercially available protocol using an affinity matrix of modified polymyxin B. Endotoxi

Fused framework for predicting binding affinity to ɑ-synuclein using three-dimensional molecular graphs and attention.

The aberrant aggregation of α‑synuclein (α‑syn) is a pathological hallmark of synucleinopathies, driving the need for small‑molecule inhibitors that block its oligomerization and fibrillization. In computer‑aided drug discovery, accurate affinity prediction remains critical for early‑stage screening. However, current deep learning models predominantly rely on 1D sequences or 2D topological graphs, failing to exploit stereochemical and geometric cues from 3D space. Moreover, dedicated high‑precision predictors for intrinsically disordered targets such as α‑syn are scarce. To bridge this gap, we

Heat shock protein 10 as a chaperone modulating α-synuclein amyloid fibril formation.

HSP10 is a well-known human co-chaperone that interacts with HSP60 to comprise the HSP60/10 chaperonin complex which upholds mitochondrial proteostasis. HSP10 also demonstrates independent roles in binding to misfolded proteins and interacts with several amyloidogenic client proteins. Using a variety of biophysical and biochemical methods, we studied the interactions of HSP10 with the amyloidogenic protein α-synuclein (α-syn) associated with Parkinson's disease. HSP10 efficiently inhibited fibril formation of wild type (WT) and disease-mutant A30P α-syn at sufficient concentrations of chaperon

How Are Synaptic Vesicle Exocytosis and Endocytosis Affected in Parkinson Disease?

Synaptic vesicle exocytosis and endocytosis are 2 fundamental processes that involve the coordinated and timely activity of many proteins and protein-lipid interactions. Alpha-synuclein (α-Syn) regulates several steps of these processes. Impaired exocytosis and endocytosis are important consequences of Parkinson disease (PD), reflecting both loss of function of normal α-Syn and toxic gain of function of abnormal α-Syn aggregates. Studies in familial forms of PD indicate that abnormal functions of other critical proteins such as leucine repeat rich kinase 2 also impair normal synaptic vesicular…

Early α-synuclein-mediated mitochondrial dysfunction in a human cell model of Parkinson's disease dementia.

Parkinson's disease (PD) is a progressive neurodegenerative disorder characterised by the misfolding and accumulation of α-synuclein (α-syn) into pathological aggregates known as Lewy bodies. PD remains incurable, partly due to limited physiologically relevant models that recapitulate human pathology to enable therapeutic development. We developed a novel in vitro PD dementia model using fetal human cortical neurons seeded with α-syn preformed fibrils (PFFs). This model successfully replicates key PD features, including α-syn aggregation and mitochondrial gene dysregulation. Importantly, RNA s
