Getting around the gut: a unique management challenge of thyroid storm precipitated by amphetamine-associated duodenal ischaemia leading to compromised enteric absorption.

Thyroid storm is a rare, life-threatening endocrine emergency with a high mortality rate of up to 30%. We present a unique management challenge of a critically ill patient who developed thyroid storm in the setting of a duodenal perforation from amphetamine-associated non-occlusive mesenteric ischaemia. The diagnosis of 'thyroid storm' was made based on clinical criteria and a Burch-Wartofsky score of 100. During emergent exploratory laparotomy, a 1 cm duodenal perforation with surrounding friable tissue was found and repaired. Intraoperatively, a nasogastric tube was guided distal to the area…

Maternal immune activation in mid-late gestation alters amphetamine sensitivity and object recognition, but not other schizophrenia-related behaviours in adult rats.

Maternal immune activation induced by Poly(I:C) administration is one of the most commonly used animal models of schizophrenia at present. P … ation, compared to controls, exhibited enhanced sensitivity to the locomotor-stimulating effects of amphetamine and reduced exploration of novel objects. These findings present a unique profile of effects, different to what has already been established for the same time point in rats, and different to what we had found at earlier and later time points. This w …

Sexual and reproductive health risks amongst female adolescents who use amphetamine-type stimulants and sell sex: a qualitative inquiry in Yunnan, China.

BACKGROUND: China, as other Southeast Asian countries, has witnessed an increased use in amphetamine-type stimulants (ATS) amongst urban youth. Amongst female adolescents who both sell sex and use ATS, risk behaviours are compounded resulting in even poorer health outcomes. However, limited knowledge exists on ATS use patterns and ATS-related risk behaviours, particularly in this context. This research aimed to improve the understanding of these issues amongst female adolescents who use ATS and sell sex, and to inform future programming. METHOD: This study utilised monthly focus group discuss…

Female sex workers who use amphetamine-type stimulants (ATS) in three cities of Vietnam: use and sexual risks related to HIV/AIDS.

Early evidence shows that amphetamine-type stimulant (ATS) use has been rapidly increasing in Vietnam. Female sex workers (FSWs) who use ATSs have increased sexual risks for HIV infection. This paper presents qualitative data from a mixed-method study conducted from 2010 to 2011 that aimed to explore the use of ATS among FSWs in three major cities and to identify HIV-related sexual risks among this group. A total of 37 in-depth interviews were conducted, and thematic analysis was performed using NVIVO 8.0 software. Study participants reported that they perceive ATS to be more 'stylish', 'highe…

Acute dextro-amphetamine administration does not alter brain myo-inositol levels in humans and animals: MRS investigations at 3 and 18.8 T.

The pathophysiological underpinnings of bipolar disorder are not fully understood. However, they may be due in part to changes in the phosphatidylinositol second messenger system (PI-cycle) generally, or changes in myo-inositol concentrations more specifically. Dextro-amphetamine has been used as a model for mania in several human studies as it causes similar subjective and physiological symptoms. We wanted to determine if dextro-amphetamine altered myo-inositol concentrations in vivo as it would clearly define a mechanism linking putative changes in the PI-cycle to the subjective psychologica…

A substrate trapping mutant form of striatal-enriched protein tyrosine phosphatase prevents amphetamine-induced stereotypies and long-term potentiation in the striatum.

BACKGROUND: Chronic, intermittent exposure to psychostimulant drugs results in striatal neuroadaptations leading to an increase in an array of behavioral responses on subsequent challenge days. A brain-specific striatal-enriched tyrosine phosphatase (STEP) regulates synaptic strengthening by dephosphorylating and inactivating several key synaptic proteins. This study tests the hypothesis that a substrate-trapping form of STEP will prevent the development of amphetamine-induced stereotypies. METHODS: A substrate-trapping STEP protein, TAT-STEP (C-S), was infused into the ventrolateral striatum…

[Determination of 15 fentanyl analogs and amphetamine-type stimulants in hair by liquid chromatography-tandem mass spectrometry and gas chromatography-mass spectrometry].

Drug abuse is a major global public health problem. Fentanyl analogs and amphetamine-type stimulants (ATS) are widely abused worldwide. Hair analysis can trace drug use history over months. This technique has unique advantages in forensic toxicology. However, its application is limited by a lack of matrix-certified reference materials (CRMs). At present, the existing CRMs are unable to simultaneously cover both fentanyl analogs and amphetamine-type stimulants. This study established a hair sample preparation method based on a water‑dimethyl sulfoxide‑hydrochloric acid soaking approach, targeti…

Amphetamine produces enhanced behavioral responding in sensitized male rats: Parallel changes in AMPA receptor surface expression and PKC signaling in the NAcc.

Repeated intermittent exposure to drugs like amphetamine and cocaine leads to long-lasting sensitization of their behavioral and neurochemical effects. Several lines of evidence suggest that dopamine (DA) and glutamate in the forebrain nucleus accumbens (NAcc) contribute to these effects and that AMPA receptors in this site play an essential role. Here, in experiments conducted in outbred male rats, we again show that, unlike cocaine, prior sensitizing exposure to amphetamine does not lead to an increase in basal cell surface expression of AMPA receptors. However, 30-min after an amphetamine c…

Amphetamine-type stimulant use among men who have sex with men (MSM) in Vietnam: Results from a socio-ecological, community-based study.

INTRODUCTION: Amphetamine-type-stimulants (ATS) use is associated with HIV-related sexual risk behaviours and is an emergent problem among men who have sex with men (MSM) in Vietnam. The purpose of this study is to describe ATS use patterns and understand the correlates of recent methamphetamine use from a socio-ecological perspective. METHODS: From September through December, 2014, 622 MSM were recruited in Hanoi and Ho Chi Minh City, Vietnam. We collected information on demographic characteristics, HIV testing behaviours, use of ATS and other recreational drugs (ever and recently), sexual s…

Olanzapine and risperidone disrupt conditioned avoidance responding in phencyclidine-pretreated or amphetamine-pretreated rats by selectively weakening motivational salience of conditioned stimulus.

The rat conditioned avoidance response model is a well-established preclinical behavioral model predictive of antipsychotic efficacy. All clinically approved antipsychotic drugs disrupt conditioned avoidance responding - a feature that distinguishes them from other psychotherapeutics. We previously showed that the typical antipsychotic drug haloperidol disrupts avoidance responding by progressively attenuating the motivational salience of the conditioned stimulus (CS) in normal rats. In this study, using two pharmacological rat models of schizophrenia [e.g. phencyclidine (PCP) or amphetamine s…

Reduced growth hormone responses to amphetamine in "endogenous" depressive patients: studies in normal, "reactive" and "endogenous" depressive, schizophrenic, and chronic alcoholic subjects.

Several pharmacological stimulation tests of the pituitary-hypothalamic system have been used to investigate psychiatric disorders. This study introduces amphetamine sulfate as a stimulus for human growth hormone (HGH) release in various psychiatric patients. Peak HGH release after a single intravenous administration of amphetamine sulfate, 0.1 mg/kg, was significantly lower in nine "endogenous" depressives (P = .01) and significantly higher in seven "reactive" depressives (P less than .05) as compared to normal subjects, whereas peak HGH release in eight schizophrenics and six chronic alcohol…

Bioanalysis of amphetamines in alternative matrices using a sensitive and validated liquid chromatography-tandem mass spectrometry method and its application to real samples.

Amphetamines abuse remains a major concern in both clinical and forensic toxicology. While conventional matrices like blood and urine are commonly used, their detection window is limited. This study aimed to validate a UHPLC-ESI-MS/MS method for amphetamines detection across both conventional and alternative biological specimens and apply it to real patient samples. A validated UHPLC-ESI-MS/MS method was used to quantify amphetamines in whole blood, urine, oral fluid, scalp hair, and fingernail specimens collected from fifty amphetamine abusers. Method validation followed international guideli…

Dopamine mediates a directionally opposite correlation between empathy and the reinforcing effects of amphetamine and gambling in people with gambling disorder vs. healthy controls.

Understanding the relationship between empathy, subjective effects of addictive reinforcers and dopamine function in people with gambling disorder (PGD) vs. healthy controls (HCs) may inform GD treatment. The current investigation addressed this issue via retrospective analysis of data from three studies using amphetamine and a slot machine (SLOTS) as reinforcers in PGD and HCs. The Empathy scale of Eysenck's Impulsiveness Questionnaire assessed trait Empathy. The Gamblers Beliefs Questionnaire assessed cognitive distortions. The Eysenck Lie scale assessed socially desirable responding. PET sc…

Modulation of locomotion and motor neuron response by the cohesive effect of acute and chronic feeding states and acute d-amphetamine treatment in zebrafish larvae.

Amphetamine (AMPH) increases locomotor activities in animals, and the locomotor response to AMPH is further modulated by caloric deficits such as food deprivation and restriction. The increment in locomotor activity regulated by AMPH-caloric deficit concomitance can be further modulated by varying feeding schedules (e.g. acute and chronic food deprivation and acute feeding after chronic food deprivation). However, the effects of different feeding schedules on AMPH-induced locomotor activity are yet to be explicated. Here, we have explored the stimulatory responses of acutely administered d-amp…
