TDP-43: a critical amplifier of Alzheimer's disease beyond amyloid and tau.

TAR DNA-binding protein 43 (TDP-43) proteinopathy has recently emerged as a pivotal, yet underrecognized, contributor to the multifaceted neuropathology of Alzheimer's disease (AD). While amyloid-β and tau have long been established as cardinal pathological hallmarks, growing evidence delineates TDP-43 as a critical participant of neurodegeneration, intricately interwoven with amyloid and tau pathologies. TDP-43 mislocalization, post-translational modifications, and aggregation potentiate neuronal loss through disruption of RNA metabolism, nucleocytoplasmic transport, and protein homeostasis.

Immersive digital art for well-being and ecological awareness: from a conceptual framework to practice.

BACKGROUND: The intensification of the consequences of ecological crises over the past decade has made them more visible, leading to a rise in eco-anxiety, especially in young adults. This psychological response is not pathological, but if left unaddressed, it may expose to mental health disorders. Evidence shows that art has mental health benefits, suggesting that harnessing artistic experiences may serve as catalysts for resilience and renewed connection to the living world. Young adults inhabit an increasingly digital world and, therefore, engaging them requires speaking their technological

Biological Embedding of Adverse Childhood Experiences: Neurobiological and Immune Mechanisms Linking Early-Life Stress to Addiction Vulnerability.

Adverse childhood experiences (ACEs) are associated with elevated risk for substance use disorders (SUDs), yet prevailing accounts often emphasize psychosocial pathways and reward-circuit recalibration without fully articulating immunobiological mechanisms that may converge on these processes. This narrative review advances a biological embedding framework linking early adversity to SUD vulnerability through two partially overlapping pathways: 1) stress-reward neurodevelopmental recalibration and 2) immune embedding with downstream neuroimmune feedback. Drawing on epide- miologic and mechanist

Brain-First vs. Body-First Models in Neurodegenerative Disease: A Perspective Review.

Recent advances in neurodegenerative disease research increasingly support the existence of multiple trajectory signatures underlying the heterogeneity of cognitive decline syndromes. Originally proposed in Parkinson's disease, the brain-first and body-first models have emerged as conceptual frameworks to explain variability in disease onset, prodromal features, and progression across dementia-related disorders. Brain-first phenotypes are defined by the early emergence of central nervous system pathology and cognitive symptoms, whereas body-first phenotypes are characterized by prominent perip

Mapping subcortical brain lesions, behavioral and acoustic analysis for early assessment of subacute stroke patients with dysarthria.

INTRODUCTION: Dysarthria is a motor speech disorder frequently associated with subcortical damage. However, the precise roles of the subcortical nuclei, particularly the basal ganglia and thalamus, in the speech production process remain poorly understood. METHODS: The present study aimed to better understand their roles by mapping neuroimaging, behavioral, and speech data obtained from subacute stroke patients with subcortical lesions. Multivariate lesion-symptom mapping and voxel-based morphometry methods were employed to correlate lesions in the basal ganglia and thalamus with speech produ

Barriers to Accessing Opioid Treatment in Rural Queensland: Perspectives From Consumers and Clinicians.

INTRODUCTION: Opioid treatment (OT) is highly effective for managing opioid use disorder (OUD) but remains underutilised worldwide, including in Australia. This issue is especially pronounced in regional and rural areas; however, there has been limited research focused on the unique barriers to OT access outside of metropolitan centres. In Queensland (QLD), Australia, OT is largely community-based, with dosing, prescription and supply all occurring outside hospital settings. The state's vast size and regional spread create unique access challenges, especially given jurisdictional differences i

An Evolutionary Psychological Approach Toward BDSM Interest and Behavior.

Bondage/discipline, Dominance/submission, and Sadism/Masochism (BDSM) have gained increased attention and discussion in recent years. This prevalence is accompanied by a shift in perceptions of BDSM, including the declassification of sadomasochism as a paraphilic disorder. Evolutionary psychology offers a unique perspective of why some individuals are interested in BDSM and why some prefer certain elements of BDSM over others (e.g., dominance versus submission). In this paper, we examine BDSM from an evolutionary standpoint, examining biopsychosocial factors that underlie the BDSM interests an

Genotype-phenotype characteristics of 57 patients with Prader-Willi syndrome: a single-center experience from Turkey.

OBJECTIVES: Prader-Willi syndrome (PWS) is a rare and complex genetic disorder caused by the loss of expression of the paternal copy of the imprinted genes on chromosome 15q11-q13. A variety of findings have been reported on the phenotypic differences between the genetic subtypes of PWS. This article compares the clinical findings of 57 PWS patients by genetic subtype and explores possible associations in this context. METHODS: Methylation‑specific multiplex ligation-dependent probe amplification and single nucleotide polymorphism microarrays were used to diagnose deletion and uniparental dis

Complicated Relationships between Anterior and Condylar Guidance and Their Clinical Implications-Comparison by Cone Beam Computed Tomography and Electronic Axiography-An Observational Cohort Cross-Sectional Study.

A complex prosthodontic treatment is believed to be more successful when the condylar path is replicated using the articulator. However, there is an ongoing major disagreement between the researchers as the exact relationship between the posterior and anterior determinants has not been clear. The purpose of this study was to investigate whether the protrusive movement of the mandible does correlate with the temporomandibular joint (TMJ) anatomy or with incised features. Subjects (15 males and 15 females) were qualified for this study based on an initial interview including the following criter

Canine Rise Method: A Conservative Approach for Worn Teeth Rehabilitation with Different Adhesive Restorative Materials.

BACKGROUND: In this article, two cases with generalized dental wear and four cases with localized dental wear are reported. In some of the cases, the worn teeth were restored with direct composite techniques with no mock-up and articulator mounting according to the canine rise method. This method is used without any change in the intercuspation of posterior teeth without a muscle relaxation appliance. It introduces a stable occlusal condition that can alter neuromuscular reflex activity, leading to improvements in certain muscle pain disorders. It is also used to protect the teeth and supporti

A comparison of the functional connectome in mild traumatic brain injury and post-traumatic stress disorder.

Post-traumatic stress disorder (PTSD) and mild traumatic brain injury (mTBI) often co-occur in the context of threat to one's life. These conditions also have an overlapping symptomatology and include symptoms of anxiety, poor concentration and memory problems. A major challenge has been articulating the underlying neurobiology of these overlapping conditions. The primary aim of this study was to compare intrinsic functional connectivity between mTBI (without PTSD) and PTSD (without mTBI). The study included functional MRI data from 176 participants: 42 participants with mTBI, 67 with PTSD and

Update on the parameters influencing the adjustment of the sagittal and transversal condylar inclination of dental articulators.

OBJECTIVES: The literature review aimed to compile and summarize the results of research relating to the recordings of condylar displacements obtained with extraoral devices, to guide clinicians to set dental (virtual) articulator parameters. The meta-analysis was undertaken to assess the sagittal condylar inclination (SCI) and transversal condylar inclination (TCI, also known as Bennett angle) values according to horizontal reference planes, movement studied, and patient characteristics: dental status, interocclusal relationship, skeletal pattern, and signs and symptoms of temporomandibular d

Gambling disorder and dissociative features: A systematic review and meta-analysis.

OBJECTIVE: Multilevel consequences related to gambling disorder (GD) are glaring enough to make gambling a worldwide public health issue. Dissociation has been pointed out in clinical, empirical, and theoretical contributions as a key variable accounting for the development and maintenance of GD. However, we still lack a systematization of available empirical data that may facilitate further accurate conclusions. METHOD: A systematic review and meta-analysis were performed with the goal to answer to open questions. We followed the PRISMA guidelines conducting a systematic search of 5 scientif

Speech Vision: An End-to-End Deep Learning-Based Dysarthric Automatic Speech Recognition System.

Dysarthria is a disorder that affects an individual's speech intelligibility due to the paralysis of muscles and organs involved in the articulation process. As the condition is often associated with physically debilitating disabilities, not only do such individuals face communication problems, but also interactions with digital devices can become a burden. For these individuals, automatic speech recognition (ASR) technologies can make a significant difference in their lives as computing and portable digital devices can become an interaction medium, enabling them to communicate with others and

Conceptualizing a quality of life framework for girls with Rett syndrome using qualitative methods.

Rett syndrome is a neurodevelopmental disorder mainly affecting females and associated with a mutation on the MECP2 gene. There has been no systematic evaluation of the domains of quality of life (QOL) in Rett syndrome. The aims of this study were to explore QOL in school-aged children with Rett syndrome and compare domains with those identified in other available QOL scales. The sample comprised 21 families registered with the Australian Rett Syndrome Database whose daughter with Rett syndrome was aged 6-18 years. Semi-structured telephone interviews were conducted with each parent caregiver

Otolaryngologic manifestations of Noonan syndrome.

Noonan syndrome is an autosomal dominant disorder with associated anomalies that include short stature, congenital heart defects, developmental delay, and characteristic facial features among other abnormalities. Articulation deficiency and language delay are often present and require speech therapy. Otitis media and hearing loss have been reported to be common in these patients. We performed a retrospective chart review of pediatric patients who were diagnosed with Noonan syndrome at our tertiary care center from January 1979 through December 2009. We found 19 such patients. Of these, 8 had r

[Is progressive anarthria a clinical form of Pick complex?].

INTRODUCTION: Progressive anarthria is defined as a clinical entity with a degenerative origin consisting in progressive difficulty in articulating while grammatical, semantic and graphic aspects of language are preserved. It is included within the group of processes affecting restricted areas of the brain although its exact nosological location is not clear. We report two cases that progressed clinically towards frontotemporal dementia and corticobasal degeneration, respectively. CASE REPORTS: Case 1: a male who, at the age of 72, began with speech difficulties while comprehension and readin
