3,4,5-Trimethoxycinnamin acid ameliorates restraint stress-induced anxiety and depression.

The present study investigated whether 3,4,5-trimethoxycinnamic acid (TMCA) treatment ameliorated restraint stress-provoked anxiety- and depression-like behaviors in mice. Fourteen consecutive days of restraint stress produced anxiety- and depression-like behaviors, including reduced time and frequency in the open arms of the elevated plus maze (EPM), as well as enhanced immobility times in the forced swim test (FST). However, TMCA (50 mg/kg) treatment ameliorated this effect; mice showed increased time and frequency of visits in the open arms of the EPM and showed reduced immobility in the FS

3,4,5-Trimethoxycinnamic acid (TMCA), one of the constituents of Polygalae Radix enhances pentobarbital-induced sleeping behaviors via GABAAergic systems in mice.

These experiments were performed to investigate whether 3,4,5-trimethoxycinnamic acid (TMCA), one of the constituents derived from Polygalae Radix, enhances pentobarbital-induced sleeping behaviors, and to alter sleep architecture through the γ-aminobutyric acid (GABA)ergic systems in mice. TMCA decreased the locomotor activity. TMCA prolonged total sleep time, and reduced sleep latency induced by pentobarbital, similar to muscimol, a GABAA agonist. From the electrocencephalogram recording for 6 h after TMCA administration, the number of sleep/wake cycles were reduced by TMCA. TMCA also increa

Pan-histone deacetylase inhibitors regulate signaling pathways involved in proliferative and pro-inflammatory mechanisms in H9c2 cells.

BACKGROUND: We have shown previously that pan-HDAC inhibitors (HDACIs) m-carboxycinnamic acid bis-hydroxamide (CBHA) and trichostatin A (TSA) attenuated cardiac hypertrophy in BALB/c mice by inducing hyper-acetylation of cardiac chromatin that was accompanied by suppression of pro-inflammatory gene networks. However, it was not feasible to determine the precise contribution of the myocytes- and non-myocytes to HDACI-induced gene expression in the intact heart. Therefore, the current study was undertaken with a primary goal of elucidating temporal changes in the transcriptomes of cardiac myocyt

Cinnamoyl derivatives of 7alpha-aminomethyl-6,14-endo-ethanotetrahydrothebaine and 7alpha-aminomethyl-6,14-endo-ethanotetrahydrooripavine and related opioid ligands.

A new series of ligands has been synthesized where the cinnamoyl group of the 14-cinnamoylamino morphinones has been introduced to the 7alpha-substituent of the 6,14-bridged oripavine series. In vitro the compounds were mostly low efficacy partial agonists or antagonists with some selectivity for the mu opioid receptor, with evidence of micro efficacy in vivo. The similarity in SAR between these 6,14-bridged oripavines and the 14-cinnamoylamino series suggests a similar mode of interaction with the micro opioid receptor.

Modification of endotoxin-induced haemodynamic and haematological changes in the rabbit by methylprednisolone, F(ab')2 fragments and rosmarinic acid.

The effects of methylprednisolone, F(ab')2 fragments of human gamma globulins and rosmarinic acid, an inhibitor of complement activation, were tested on endotoxin-induced haemodynamic and haematological changes in the rabbit. Their effects were compared with complement depletion by cobra venom factor (CVF) pretreatment. The results provide further evidence for the role of complement activation and the concomitant triggering of the arachidonic acid cascade in the early phase of shock. The formation of vasoactive prostanoids (prostacyclin and thromboxane A2), the arterial hypotension and the thr

Foliar application of allantoin and serotonin mitigates salinity stress in bitter melon (Momordica charantia L.).

Bitter melon (Momordica charantia L.) is a climbing species frequently employed in traditional medicine to treat a wide number of diseases. Soil salinity represents a primary abiotic constraint on plant growth, impacting nearly 50% of irrigated farmland and constantly increasing. Previous studies revealed the individual benefits of allantoin and serotonin, in mitigating abiotic stresses. Therefore, in the current study, we investigated their ability, both individually and combination, ameliorating the damaging consequences of salt stress on the development and physiological and biochemical att

Paired genomic and proteomic analysis of Acinetobacter towneri from waste sludge reveals an extensive repertoire of enzymes for breakdown of hydrocarbons.

Crude oil, primarily consisting of alkanes, and plastic derivatives are integral to modern society, yet their widespread use has led to persistent pollution that devastates ecosystems worldwide. Enzymatic and microbial biodegradation are highly desirable to become alternatives to conventional methods for the control of these pollutants in terrestrial and aquatic environments. Consequently, investigating the enzymatic machinery of specialized microbes that thrive in contaminated areas and possess the catabolic potential to degrade alkanes and plastics is of considerable interest. In this study,

Exploring the potential of hydro alcoholic crude extract of beeswax as antibacterial antifungal antiviral antiinflammatory and antioxidant agent.

Apis mellifera beeswax is synthesized from honey sugars and secreted by specialized glands located on the ventral side of the abdomen of worker bees aged 12-18 days. This beeswax possesses various therapeutic properties beneficial to human health. Given the priority in pharmacological research to develop new drugs from natural sources with minimal side effects, the absence of studies on the biological activities of A. mellifera beeswax is notable. Therefore, this study aims to investigate the chemical composition, as well as the anti-inflammatory, antiviral, cytotoxic, antimicrobial, and antio

Metabolomics analysis of mechanism of improving quality of Schisandrae chinensis fructus by NO combining with high-temperature stress.

The fruits of Schisandra chinensis (Turcz.) Baill. (Schisandrae chinensis fructus) are a well-known herbal medicine, known for its hepatoprotective, antidepressant, antioxidant, and sedative-hypnotic properties. Over-exploitation of wild resources led to the rise of cultivation, along with a decrease in quality. Exposure of plants to adversity must generate substantial quantities of reactive oxygen species (ROS) and result in cellular damage. In response, secondary metabolites are produced to neutralize ROS; these secondary metabolites are usually the active ingredient of herbal medicine, so t

An in vitro study elucidating the synergistic effects of aqueous cinnamon extract and an anti-TNF-α biotherapeutic: implications for a complementary and alternative therapy for non-responders.

BACKGROUND: Tumor necrosis factor-alpha (TNF-α) is a critical pro-inflammatory cytokine, and its abnormal production is associated with several immune mediated inflammatory diseases (IMID). Biological anti-TNF-α therapy includes treatment with monoclonal antibodies such as infliximab which have proven successful and are well-tolerated in most patients. Unfortunately, some patients may not respond to therapy (primary non-responders) or may lose sensitivity to the biological agent over time (early and late secondary non-responders). Natural products can reduce inflammation and act synergisticall

Multi-omics strategies uncover the molecular mechanisms of nitrogen, phosphorus and potassium deficiency responses in Brassica napus.

BACKGROUND: Nitrogen (N), phosphorus (P) and potassium (K) are critical macronutrients in crops, such that deficiency in any of N, P or K has substantial effects on crop growth. However, the specific commonalities of plant responses to different macronutrient deficiencies remain largely unknown. METHODS: Here, we assessed the phenotypic and physiological performances along with whole transcriptome and metabolomic profiles of rapeseed seedlings exposed to N, P and K deficiency stresses. RESULTS: Quantities of reactive oxygen species were significantly increased by all macronutrient deficienci

Magnesium Nutrient Application Induces Metabolomics and Physiological Responses in Mulberry (Morus alba) Plants.

Mulberry (Morus alba) is a significant plant with numerous economic benefits; however, its growth and development are affected by nutrient levels. A high level of magnesium (Mg) or magnesium nutrient starvation are two of the significant Mg factors affecting plant growth and development. Nevertheless, M. alba's metabolic response to different Mg concentrations is unclear. In this study, different Mg concentrations, optimal (3 mmol/L), high (6 mmol/L and 9 mmol/L), or low (1 and 2 mmol/L) and deficient (0 mmol/L), were applied to M. alba for three weeks to evaluate their effects via physiologic

Induction, titration, and maintenance dosing regimen in a phase 2 study of pegvaliase for control of blood phenylalanine in adults with phenylketonuria.

BACKGROUND: Phenylketonuria (PKU) is caused by a deficiency in phenylalanine hydroxylase enzyme activity that leads to phenylalanine (Phe) accumulation in the blood and brain. Elevated blood Phe levels are associated with complications in adults, including neurological, psychiatric, and cognitive issues. Even with nutrition and pharmacological management, the majority of adults with PKU do not maintain blood Phe levels at or below guideline recommended levels. Pegvaliase, PEGylated recombinant Anabaena variabilis phenylalanine ammonia lyase (PAL), converts Phe to trans-cinnamic acid and ammoni

Further development of the NCTC 2544 IL-18 assay to identify in vitro contact allergens.

Several European Union legislations request the use of in vitro methods for toxicological evaluations, including sensitization, in order to increase consumer safety but also to reduce the use of animals. The EU project SENS-IT-IV addresses the need of developing predictive in vitro tests to assess contact and respiratory hypersensitivity reactions. In this context, we have recently reported the possibility to use IL-18 production in the human keratinocyte cell line NCTC 2544 to discriminate contact sensitizer from irritants and low molecular weight respiratory allergens. The aims of the presen
