Anti-LCN2 antibody treatment is associated with attenuated hearing loss and blood-labyrinth barrier disruption in a mouse model of chronic kidney disease.

Chronic kidney disease (CKD) is associated with sensorineural hearing loss, but the mechanisms linking renal injury to cochlear dysfunction remain incompletely defined. Because lipocalin-2 (LCN2/NGAL) is markedly increased in CKD and has been implicated in endothelial dysfunction and barrier injury, we investigated whether LCN2 contributes to cochlear damage in a mouse model of CKD. Male C57BL/6J mice underwent 5/6 nephrectomy, and CKD mice received either an anti-LCN2 monoclonal antibody or an isotype control three times weekly for seven weeks beginning one week after completion of surgery. Poria cocos polysaccharide alleviates multidrug-resistant Klebsiella pneumoniae-induced acute lung injury by modulating the gut-lung axis via gut microbiota remodeling.

The emergence and spread of multidrug-resistant Klebsiella pneumoniae (MDR-KP), which leads to acute lung injury (ALI) and severe pneumonia, pose a huge challenge to clinical treatment and public health. Non-antibiotic strategies have recently garnered significant attention as promising avenues for combating antibiotic resistance. In this study, whole-genome sequencing of an MDR-KP strain isolated from a deceased goat revealed key genetic features underlying its pathogenicity, including 31 resistance genes mediating broad-spectrum resistance to 14 antibiotic classes, 71 pathogen-host interacti

Therapeutic impacts of oral anti-diabetic drugs on Inflammatory Bowel Disease (IBD), a comprehensive review of clinical and preclinical studies.

Inflammatory bowel disease (IBD) is a chronic condition with a rising incidence linked to inflammation and oxidative stress. This disorder includes two main subtypes: ulcerative colitis (UC) and Crohn's disease (CD). Current treatment plans lack effectiveness; as a result, finding novel approaches is encouraged. Various groups of oral anti-diabetic medications are employed in clinical practice, and they have shown beneficial effects against conditions beyond diabetes mellitus (DM). This review is aimed at evaluating the hypothesis that oral anti-diabetic medications may serve as potential ther

The impact of active components from Piper sarmentosum on the growth, intestinal barrier function, and immunity of broiler chickens.

OBJECTIVE: Pellitorine (PT) and vitexin-2-O-rhamnoside (VR) are two bioactive compounds found in Piper sarmentosum (PS). Their contents are relatively high in the ethanol extract of PS (PSE). However, it remains unknown whether PT and VR are the primary components of PS that exert beneficial effects on gut health. In this study, we aimed to confirm that these two compounds are the primary anti-inflammatory active ingredients in PSE. METHODS: Total of 300 female one-day-old Danzhou chickens were randomly divided into five groups with five replicates each. Chickens were given a basal diet (CON

The Improvement of Intestinal Mucosal Epithelial Barrier Integrity by 1,3,4-Oxadiazole Derivatives of Pyrrolo[3,4-d]pyridazinone in Rat Experimental Colitis.

BACKGROUND AND PURPOSE: The intestinal mucosal barrier is a complex structure that separates the internal and lumen environments. An impaired intestinal barrier may lead to excessive mucosal immune system activation and further to intestinal diseases, including inflammatory bowel disease (IBD). Therefore, improving the integrity of the intestinal barrier may be a therapeutic approach to prevent or treat IBD. In this study, we aimed to elucidate the effects of the new 1,3,4-oxadiazole derivatives of pyrrolo[3,4-d]pyridazinone, compounds 7b, 10b, and 13b, in intestinal epithelial damage. METHOD

Itaconic Acid Alleviates Perfluorooctanoic Acid-Induced Oxidative Stress and Intestinal Damage by Regulating the Keap1/Nrf2/Ho-1 Pathway and Reshaping the Gut Microbiota.

Itaconic acid (IA) is recognized for its potential application in treating intestinal diseases owing to the anti-inflammatory and antioxidant properties. Perfluorooctanoic acid (PFOA) can accumulate in animals and result in oxidative and inflammatory damages to multi-tissue and organ, particularly in the intestinal tract. This study aimed to explore whether IA could mitigate intestinal damage induced by PFOA exposure in laying hens and elucidate its potential underlying mechanisms. The results showed that IA improved the antioxidant capacity of laying hens and alleviated the oxidative damage i

Novel organic selenium source hydroxy-selenomethionine counteracts the blood-milk barrier disruption and inflammatory response of mice under heat stress.

Heat stress (HS) in summer has caused huge economic losses to animal husbandry production recently. When mammary gland is exposed to high temperatures, it will cause blood-milk barrier damage. Hydroxy-selenomethionine (HMSeBA) is a new selenium source with better guarantee of animals' production performance under stress, but whether it has protective effect on heat stress-induced blood-milk damage is still unclear. We established mammary epithelial cells and mice heat stress injury models to fill this research gap, and hope to provide theoretical basis for using HMSeBA to alleviate heat stress

Hepatitis C Virus Infection of Human Thyrocytes: Metabolic, Hormonal, and Immunological Implications.

CONTEXT: Hepatitis C virus (HCV) infection is a prevalent disease worldwide. Thyroid dysfunction is one of the most common extrahepatic manifestations of HCV infection. We hypothesized that HCV can directly infect human thyrocytes thereby causing thyroid dysfunction. SETTING: Human thyrocytes in primary cell culture, ML-1 human thyroid cell line, and Huh7.5 human hepatocyte cell line were infected with HCV using the Huh7.5JFH1 cell line that releases infectious HCV virions. After infection, the release of new virions, production of proinflammatory cytokines, and expression of miR-122 were eva
