pH and nitric oxide synthase activity and expression in bovine aortic endothelial cells.

AIM: Nitric oxide (NO) plays an important role in the transition from intrauterine to extrauterine life. If this transition fails, a condition called persistent pulmonary hypertension of the neonate (PPHN) may develop. The current treatment modalities for this disease include induction of alkalosis by hyperventilation or alkali infusion, inhaled nitric oxide (iNO) and extracorporeal membrane oxygenation. There is evidence from animal studies that the elevated pH, not the low pCO2 is responsible for the resultant pulmonary vasodilatation. In this study, we examined the effect of pH on the activ

Nitric oxide/cGMP-mediated effects in the outflow region of the lower urinary tract--is there a basis for pharmacological targeting of cGMP?

Treatment with alpha-adrenoceptor antagonists that reduce the tone of prostatic stromal and urethral smooth muscle has beneficial effects in patients with benign prostatic hyperplasia (BPH) and lower urinary tracts symptoms (LUTS) and has brought attention to regulatory mechanisms of smooth muscle contractility of the outflow region. The prostate, urethra and bladder neck are densely supplied by nitric oxide (NO)-synthase-containing nerves that cause relaxation upon activation. In various experimental models, altered function or activity of the NO/cGMP pathway of the bladder neck and urethra m

Enzyme-independent nitric oxide formation during UVA challenge of human skin: characterization, molecular sources, and mechanisms.

Many of the local UV-induced responses including erythema and edema formation, inflammation, premature aging, and immune suppression can be influenced by nitric oxide synthase (NOS)-produced NO which is known to play a pivotal role in cutaneous physiology. Besides NOS-mediated NO production, UV radiation might trigger an enzyme-independent NO formation in human skin by a mechanism comprising the decomposition of photo-reactive nitrogen oxides. Therefore, we have examined the chemical-storage forms of potential NO-generating agents, the mechanisms and kinetics of their decomposition, and their

Dopamine in the ink defence system of Sepia officinalis: biosynthesis, vesicular compartmentation in mature ink gland cells, nitric oxide (NO)/cGMP-induced depletion and fate in secreted ink.

The biosynthesis, localization and fate of catecholamines in the ink gland of the cuttlefish Sepia officinalis were investigated by combined biochemical and immunohistocytochemical methodologies. HPLC analysis of crude ink gland extracts indicated the presence of dopa (2.18+/-0.82 nmol/mg of protein) and DA (dopamine, 0.06+/-0.02 nmol/mg of protein), but no detectable noradrenaline or adrenaline. DA was shown to derive from L-tyrosine, according to experiments performed by incubating intact ink glands with [L-14C]tyrosine. The biosynthetic process involves a tyrosine hydroxylase and a dopa dec

[Plasma level of atrial natriuretic peptide following oral glycerol administration].

Atrial natriuretic peptide (ANP) is a cardiac hormone known to mediate increased capillary permeability, vasodilation, and natriuresis. In 1988 the authors' experimental results indicated the presence of ANP receptors in the inner ear. From this they hypothesized that ANP might be involved in the regulation of inner ear fluids and therefore investigated the effects of orally administered glycerol on ANP and cGMP plasma levels. As regards the initial values, there was no apparent significant difference between the 14 subjects with Ménière's disease and the 34 subjects without. During the first

Non-identity of cGMP as the guanine nucleotide stimulated to bind to ROS by light and ATP.

Light, in the presence of ATP, has been reported to stimulate cGMP binding to a 58 kDa protein in ROS (rod outer segments, Fesenko and Krapivinsky, 1986b, Photobiochem. Photobiophys. 13 345-58). This apparent light-related redistribution of ROS cGMP has been suggested to eliminate any requirement for phosphodiesterase-promoted hydrolysis of cGMP in the mechanism subserving phototransduction. Using conditions identical to those previously reported, this effect of light and ATP was examined further by characterizing the metabolic products that arise and the nucleotides that become liganded. [Nitrate tolerance].

Nitrate tolerance is defined as an attenuation or even loss of hemodynamic and anti-ischemic effects during continuous nitrate medication. T … the guanylate-cyclase, and stimulation of a specific phosphodiesterase may result in a decrease of cyclic GMP formation and hence to a decrease in nitrate induced vasodilatation. Tolerance development may be prevented by intermittent nitrate administration providing intervals with low plasma and tissue nitrate levels. In consequence, nitrates should be used …

Demonstration of neurochemical tolerance following chronic barbital treatment.

Control rats (C) and rats rendered barbital dependent (BD) by the consumption of barbital chronically in their food for 6 weeks were treated with saline or 1 of 3 acute dosages of barbital 16 minutes before killing. Cyclic guanosine monophosphate (cGMP) levels and circulating levels of barbital were measured. Significantly higher circulating levels of barbital were required to produce a comparable suppression of cerebellar cGMP in the BD as opposed to the C rats. These data provide evidence for the development of tolerance to barbital in the cerebellar cGMP system during chronic barbital treat

Regulation of abalone sperm cyclic AMP concentrations and the acrosome reaction by calcium and methylxanthines.

When Ca2+ is added to abalone sperm (Haliotis rufescens) in Ca2+-free artificial seawater (CaFASW) to a final concentration of 9.6 mM a 4-fold elevation in sperm cAMP occurs within 15-30 sec. The methylxanthines, theophylline and 1-methyl-3-isobutylxanthine (MIX), also elevate sperm cAMP concentrations. In CaFASW, either compound causes up to a 3-fold increase in cAMP within 15-30 sec. MIX (150 microM), added to sperm in the presence of 9.6 mM Ca2+, elevates sperm cAMP 100-fold within 15-30 sec and also triggers the acrosome reaction (AR) in the same period. Under identical conditions theophyl

The release of surfactant in rat lung by brief periods of hyperventilation.

We investigated the release of surfactant-type phospholipids (S) using the isolated perfused rat lung (IPL). Following a 20 min equilibration period the lungs were hyperventilated for up to 15 min and then lavaged. Changing the peak inspired pressure (PIP) from 10 to 20 cm H2O rapidly increased the rate of release of S; this rate declined after 2 min. In contrast, doubling frequency of ventilation while maintaining the control tidal volume had no effect. The increase in alveolar S reflected release. rather than redistribution, and after 2 min amounted to about 8% of total S in lung tissue. Equ

Central blood pressure effects of guanylyl-imido-diphosphate and cyclic guanosine-monophosphate.

Injections of guanylyl-imido-diphosphate (250, 500 and 1,000 microgram/kg) into the lateral cerebral ventricle of the anaesthetized cat induced increases in blood pressure and heart rate while the intravenous injections of the same doses were ineffective, thus indicating a central mechanism of action of this compound which activates adenylcyclase at the catalytic subunit. The results support the hypothesis that the activity of cardiovascular centres depends on the prevailing concentration of cAMP. Intracerebroventricular injection of cGMP (125, 250 and 500 microgram/kg) caused hypotension and

Immunohistochemical distribution of cyclic nucleotide phosphodiesterase (PDE) isoenzymes in the human vagina: a potential forensic value?

OBJECTIVES: Phosphodiesterase (PDE) isoenzymes are key proteins involved in the maintenance of the normal function of various tissues of the human body including those of the male and female urogenital tract. More recently, PDEs and their main substrates, cyclic GMP and cyclic AMP, have also been assumed to play a crucial role in the control of the human vagina. In order to elucidate the potential significance of phosphodiesterases as marker proteins in female genital organs, it was the aim of the present study to evaluate by means of immunohistochemistry the distribution of cGMP- and cAMP-PDE…

cGAS-STING Signalling in the tumour microenvironment: Implications for immune surveillance and therapeutic strategies.

The cyclic GMP-AMP synthase-stimulator of interferon genes (cGAS-STING) pathway links cytosolic DNA sensing to type I interferon production and inflammatory signalling, making it central to innate immunity. In addition to defending against pathogens, it shapes the tumour microenvironment by regulating immune surveillance, antitumour activity, and tumour progression. Short-term cGAS-STING activation enhances dendritic cell maturation, antigen presentation, macrophage polarisation, and cytotoxic lymphocyte recruitment. However, prolonged activation shifts the TME into an immunosuppressive state …

cGAS-agonistic spherical nucleic acids reprogram the glioblastoma immune microenvironment and promote antitumor immunity.

The cyclic GMP-AMP synthase-Stimulator of Interferon Genes (cGAS-STING) pathway is an important DNA-sensing mechanism that increases T cell trafficking and activation in tumors and reverses the immunosuppressive phenotype of myeloid cells. Therefore, direct STING targeting using synthetic cyclic dinucleotides (CDNs) is an attractive strategy for treating lymphocyte-depleted and myeloid cell-enriched tumors, such as glioblastoma (GBM). However, inadequate bioavailability and poor cellular accumulation limit the clinical development of CDNs, particularly for noninvasive administration strategies…

Endothelium- and beta-2 adrenoceptor-independent relaxation of rat aorta by tyramine and certain other phenylethylamines.

Molecular pathobiology of scleritis and its therapeutic implications.

Scleritis and other autoimmune diseases are characterized by an imbalance in the levels of pro-inflammatory and anti-inflammatory molecules with the balance tilted more towards the former due to the failure of recognition of self. The triggering of inflammatory process could be ascribed to the presence of cytoplasmic DNA/chromatin that leads to activation of cytosolic DNA-sensing cGAS-STING (cyclic GMP-AMP synthase linked to stimulator of interferon genes) pathway and enhanced expression of NF-κB that results in an increase in the production of pro-inflammatory bioactive lipids. Stimulatory effects of human pancreatic polypeptide on rat pancreatic acini.

The effect of human pancreatic polypeptide (HPP) on rat pancreatic acini has been studied. It was found that HPP stimulated amylase and lipa … se release stimulated by HPP was accompanied by an increase in 45Ca2+ efflux. Atropine or dibutyryl cyclic GMP did not influence the effect of HPP. It is concluded that HPP stimulates the release of enzymes from rat pancreatic acini and that Ca2+ may be a mediator for this secretion.
