Ionic activities of the inner ear fluid and ionic permeabilities of the cochlear duct in endolymphatic hydrops of the guinea pig.

Ionic activities (K+, Na+, and Cl-) of the perilymph and endolymph of the basal turn were measured using ion-selective microelectrodes in experimentally induced endolymphatic hydrops of the guinea pig. Three months following the obstruction of the endolymphatic duct and sac, the endocochlear potential (EP) of hydroptic ears was measured at 59.7 +/- 9.6 mV (N = 12) which was significantly lower than the EP of the contralateral control ears (84.4 +/- 2.8 mV, N = 12). A paired t-test (P greater than 0.05) showed no significant differences of ion concentrations of the inner ear fluid between the h

Localization of the atrial natriuretic peptide binding sites in the inner ear tissue--possibly an additional regulating system.

Specific high affinity atrial natriuretic peptide (ANP) binding sites were identified and localized by autoradiography (125J-ANP 1-28) in the inner ear of guinea pigs. ANP is a cardiac hormone known to mediate increased capillary permeability, vasodilation, and natriuresis. Since specific receptors for ANP exist in both eye and brain, possibly playing a role in regulation of fluid and pressure (14, 15), we hypothetized that ANP might be involved also in inner ear fluid dynamics via interaction with other volume/electrolyte balance regulating hormones including vasopressin, renin, and aldostero

Evidence of serum antibodies against inner ear tissues in the blood of patients with certain sensorineural hearing disorders.

In healthy human temporal bones, immunoglobulins (IgG, IgA) and the secretory component are demonstrated by two immunohistochemical methods in epithelial cells and in the lumen of the endolymphatic sac. The normal human inner ear does not otherwise show any localization of immunoglobulins. By means of indirect immunofluorescence, antibodies against healthy inner ear tissue were found in the serum of 15 out of 21 patients with double-sided progressive or fluctuating sensorineural hearing loss of undefined etiology. In 2 cases of Cogan's syndrome, it was also possible to demonstrate serum antibo

[Antibacterial therapy in surgery of the inner and middle ear. A study of co-trimoxazole penetration into the perilymph (author's transl)].

Studies of the type presented here have rarely been undertaken and should be useful in surgery and pathology of the inner and middle ear. Samples of perilymph were collected during stapedectomy in patients with otosclerosis. In view of the very small volume of perilymph obtainable from each patient's vestibule (2 to 4 microliter), the only assay method that could be used to measure drug levels was thin layer chromatography on silica gel. Despite pooling of the perilymphs of 5 patients, trimethoprim (TMP) levels could not be measured but the authors were able to demonstrate that sulfamethoxazol

Structural changes in the organ of Corti of the guinea pig after obstruction of the arterial blood flow to the inner ear.

Forty-one young guinea pigs were studied after obstruction of the anterior inferior cerebellar artery in order to ascertain the effects on the surface specimen of the organ of Corti. Most of the animals had damage in the third and fourth turns (22) and a minority of these had dizziness and destruction nystagmus (3). Our hypothesis is that one of the causes of Meniere's disease is an interference with the inner ear circulation, and our plans for testing this hypothesis are given.

AI-Assisted Compressed Sensing for Optimized Inner Ear Imaging: a Multi-Rater MRI Evaluation of CSAI T2-DRIVE.

PURPOSE: High-resolution T2-weighted imaging is essential for preoperative assessment before cochlear implantation. Compressed sensing (CS) with AI-based reconstruction (CSAI) reduces acquisition times whilst preserving image quality. Although CSAI has been established in various clinical applications, its performance in inner ear imaging remains unclear. This study assesses CSAI-optimized T2-DRIVE sequences at different resolutions and acquisition times for visualizing inner ear structures. METHODS: In 30 healthy participants, CS T2-DRIVE was acquired at isotropic resolutions of 0.65, 0.5, a

Diagnostic value of the inner ear computed tomography measurements in detecting congenital sensorineural hearing loss: a comparative study.

BACKGROUND: Congenital sensorineural hearing loss (CSNHL) is a very common subtype of pediatric hearing loss disorders. Less severe deformities in CSNHL might not be recognized during a simple visual inspection in computed tomography (CT) of the temporal bone. Therefore, the present study aimed to compare the inner ear measurements between CSNHL patients with grossly normal CT appearance and the healthy population. MATERIALS AND METHODS: Children with confirmed CSNHL were enrolled along with a control group of healthy children. Their CT scans of the temporal bone were retrospectively reviewed

Phenotypic Expansion of PPP1R12A-Related Syndrome: A Novel Splicing Variant Associated with Hearing Loss and Inner Ear Malformations.

Background: Pathogenic variants in the PPP1R12A gene have been associated with a malformation syndrome involving the brain and the genitourinary systems (GUBS, MIM #618820). To date, neither hearing loss (HL) nor inner ear malformations have been reported in affected individuals, and these features are therefore not currently regarded as part of the PPP1R12A-related phenotype. Moreover, functional evidence supporting the pathogenicity of several reported variants remains limited. Methods: We investigated a 12.5-year-old patient presenting with profound bilateral sensorineural hearing loss asso

Establishment of a human inner ear model reveals that gentamicin C2b is substantially less ototoxic than clinical gentamicin.

Clinically used gentamicin, a widely used aminoglycoside antibiotic, is a mixture of 5 major subtypes with potentially differential contributions to the drug's known ototoxicity. The gentamicin subtype C2b has been previously associated with reduced ototoxicity in animal models in vitro, but its effects in humans remain uncertain due to the lack of appropriate in vitro models of the human inner ear. To address this gap, we generated otic progenitor cells (OPCs) from human induced pluripotent stem cells and used transcriptomic and immunocytochemical analyses to confirm their otic lineage identi

Pathological Insights into Lassa Virus-Induced Vestibular Dysfunction in Mice: Histopathological Analysis of the Inner Ear Vestibular Apparatus.

Lassa fever (LF), caused by Lassa virus (LASV) infection, typically leads to mild symptoms in humans, but some survivors experience audiovestibular problems. Here we present vestibular histopathological insights in our LF model mice. We observed (1) hemorrhage within the vestibular ganglion and stroma beneath the sensory epithelium, (2) preserved hair cells and supporting cells, (3) LASV antigen presence in the vestibular ganglion cells and the stroma beneath the sensory epithelium, and (4) CD3-positive T-lymphocyte infiltration in the vestibular ganglion and the stroma underlying the sensory

Automated volumetric analysis of the inner ear fluid space from hydrops magnetic resonance imaging using 3D neural networks.

Due to the development of magnetic resonance (MR) imaging processing technology, image-based identification of endolymphatic hydrops (EH) has played an important role in understanding inner ear illnesses, such as Meniere's disease or fluctuating sensorineural hearing loss. We segmented the inner ear, consisting of the cochlea, vestibule, and semicircular canals, using a 3D-based deep neural network model for accurate and automated EH volume ratio calculations. We built a dataset of MR cisternography (MRC) and HYDROPS-Mi2 stacks labeled with the segmentation of the perilymph fluid space and end

Comparison between audio-vestibular findings and contrast-enhanced MRI of inner ear in patients with unilateral Ménière's disease.

OBJECTIVE: The diagnosis of Ménière's disease (MD), characterized by idiopathic endolymphatic hydrops (ELH), remains a clinical priority. Many ancillary methods, including the auditory and vestibular assessments, have been developed to identify ELH. The newly emerging delayed magnetic resonance imaging (MRI) of the inner ear after intratympanic gadolinium (Gd) has been used for identifying ELH in vivo. We aimed to investigate the concordance of audio-vestibular and radiological findings in patients with unilateral MD. METHODS: In this retrospective study, 70 patients with unilateral definite

Findings of Intravenous Gadolinium Inner Ear Magnetic Resonance Imaging in Patients With Acute Low-Tone Sensorineural Hearing Loss.

OBJECTIVES: Acute low-tone sensorineural hearing loss (ALHL) is thought to have a different etiology from that of idiopathic sudden sensorineural hearing loss. We hypothesized that endolymphatic hydrops (EH) in the inner ear organ contributes to ALHL, even in patients without vertigo. This study investigated the presence of EH in ALHL and compared the clinical characteristics of patients with or without EH. METHODS: We retrospectively reviewed 38 patients diagnosed with ALHL without vertigo from January 2017 to March 2022. EH was measured in all patients using inner ear magnetic resonance ima

Single-cell transcriptomic profiling of the zebrafish inner ear reveals molecularly distinct hair cell and supporting cell subtypes.

A major cause of human deafness and vestibular dysfunction is permanent loss of the mechanosensory hair cells of the inner ear. In non-mammalian vertebrates such as zebrafish, regeneration of missing hair cells can occur throughout life. While a comparative approach has the potential to reveal the basis of such differential regenerative ability, the degree to which the inner ears of fish and mammals share common hair cells and supporting cell types remains unresolved. Here, we perform single-cell RNA sequencing of the zebrafish inner ear at embryonic through adult stages to catalog the diversi

Oridonin employs interleukin 1 receptor type 2 to treat noise-induced hearing loss by blocking inner ear inflammation.

NOD-like receptor protein 3 (NLRP3) inflammasomes trigger the inflammatory cascades and participate in various inflammatory diseases, including noise-induced hearing loss (NIHL) caused by oxidative stress. Recently, the anti-inflammatory traditional medicine oridonin (Ori) has been reported to provide hearing protection in mice after noise exposure by blocking the NLRP3-never in mitosis gene A-related kinase 7 (NEK7)-inflammasome complex assembly. Using RNA sequencing analysis, we further elucidated that interleukin 1 receptor type 2 (IL1R2) may be another crucial factor regulated by Ori to pr

X-Linked Gusher Disease DFNX2 in Children, a Rare Inner Ear Dysplasia with Mixed Hearing and Vestibular Loss.

Conductive hearing losses are typically present in disorders of the external/middle ear. However, there is a rare group of inner ear conditions called third windows that can also generate a conductive hearing loss. This is due to an abnormal connection between the middle and the inner ear or between the inner ear and the cranial cavity. X-linked gusher disorder is an extremely rare congenital inner ear dysplastic syndrome with an abnormal connection due to a characteristic incomplete cochlear partition type III and an incomplete internal auditory meatus fundus. The disorder is inherited in an

Physical effects of sound exposure from underwater explosions on Pacific mackerel (Scomber japonicus): Effects on the inner ear.

Studies of the effects of sounds from underwater explosions on fishes have not included examination of potential effects on the ear. Caged Pacific mackerel (Scomber japonicus) located at seven distances (between approximately 35 and 800 m) from a single detonation of 4.5 kg of C4 explosives were exposed. After fish were recovered from the cages, the sensory epithelia of the saccular region of the inner ears were prepared and then examined microscopically. The number of hair cell (HC) ciliary bundles was counted at ten preselected 2500 μm2 regions. HCs were significantly reduced in fish exposed
