SynOmega: Simplifying Retrosynthesis for Efficient Synthesizability Scoring.

Retrosynthesis-based synthesizability scoring triages molecules from generative design but is expensive: every score requires a multi-step search. We present SynOmega, an open-source toolkit that couples a single-step template model, an AND-OR route search, and a route-based synthesizability score (SynScore). Its single-step model can be restricted, at the reaction-template level, to simplifying disconnections that split the target into smaller precursors. On 1000 ChEMBL drug molecules this yields two findings. (1) The simplifying constraint cuts node expansions by about 30% on jointly solved

Efficient Mesh Reconstruction and Texturing of Oracle Bones.

The high-fidelity 3D digitization of small, detailed cultural heritage objects, such as Oracle Bones, presents significant challenges for which existing reconstruction workflows are often inadequate. Methods based on Structure-from-Motion (SfM) often lack the geometric density required to capture fine inscription details, while Light Detection and Ranging and RGB-Depth approaches may introduce high data overhead and unstable color mapping. Recent specialized studies have utilized multi-shading-based techniques to extract such hidden surface textures, yet integrating these results into a cohesi

Efficient methods for wave propagation in electron microscopy.

Accurate wave-optical simulation in electron microscopy is severely constrained by the extreme sampling requirements imposed by short wavelengths and relatively large convergence angles. Conventional implementations of the angular spectrum method (ASM) rapidly become computationally intractable, often exceeding realistic memory and time limits. We present two numerical approaches - the scaling angular spectrum method (SASM) and the no-lensing angular spectrum method (NLASM) - that systematically reduce the sampling requirements while retaining the essential physics of wave propagation. SASM re

PdYb/CeO2 Catalyst for Efficient Hydroaminocarbonylation of Alkenes.

The hydroaminocarbonylation of alkenes is one of the most promising methods for economically utilizing carbon monoxide (CO) to form high-value organic molecules. Here, we reported that a dual-atom catalyst named PdYb/CeO2 was prepared by impregnation method for the one step synthesis of amides from CO, alkenes with amines. Compared with Pd/CeO2, PdYb/CeO2 nearly doubled the amide yield. And the latter exhibited improved chemical selectivity and functional group tolerance toward diverse olefin and amine substrates. Characterization indicated that Pd and Yb species were embedded into the CeO2 su

Efficient CO2 Activation Mediated by Triplet Pnictinidenes.

The present investigation delineates the mechanistic landscape governing the [1 + 2] cycloaddition of CO2 with triplet monomeric pnictinidenes ([G15-Rea]3; G15 = Group 15 element) and identifies the principal energetic factors controlling their periodic reactivity. The computed potential-energy profiles support a stepwise mechanism in which cycloaddition is initiated on the triplet potential-energy surface through [G15-TS]3, followed by a singlet-triplet surface-crossing event at [G15-T/S] and subsequent relaxation to the singlet heterocyclic products [G15-Prod]1. ASM analysis reveals that the

Dynamics of energy-efficient coding in visual cortex.

Sparse coding enables cortical populations to represent sensory inputs efficiently, yet its temporal dynamics remain poorly understood. Here, we provide direct evidence that stimulus onset initially drives broad cortical activation, transiently reducing sparseness while increasing mutual information. Over time, competitive interactions refine the population response, maintaining high mutual information as activity declines and sparseness increases. Critically, coding efficiency, quantified as the ratio of mutual information to metabolic cost, steadily improves throughout stimulus presentation,…

Exploring Dynamic Transformer for Efficient Object Tracking.

The speed-precision tradeoff is a critical problem in visual object tracking, as it typically requires low latency and is deployed on resour … ayers of the model. Furthermore, we propose a feature recycling mechanism to maximize computational efficiency by reusing the outputs of predecessors. Additionally, a target-aware self-distillation strategy is designed to enhance the discriminating capabilities of early-stage predictions by mimicking the representation patterns of the deep model. Towards the efficient preparation of tough cellulose nanopapers.

Cellulose nanopapers are attractive materials with outstanding mechanical and optical properties, yet their production is slow. The use of non-aqueous suspension media, in particular ethanol, for nanopaper production reduced filtration times by 73 %. Nanopapers prepared from ethanolic suspensions possessed higher porosities than those prepared from aqueous suspensions, reducing their transparency and tensile properties. Rewetting nanopapers prepared from ethanolic suspensions with water and subsequent drying yielded nanopapers with densities essentially the same as those prepared from aqueous

Vascular embolic nanobiomaterials for efficient tumor treatment.

Embolization is a minimally invasive cancer treatment method. Embolization involves artificially blocking blood flow using an embolic agent to block abnormal blood vessels that supply nutrients or oxygen to a specific lesion, thereby killing the lesion, inhibiting its growth, and stopping bleeding. Currently, polyvinyl alcohol (PVA) and gelatin are the most popular embolic agents. These substances are available in various sizes and shapes that physically obstruct blood flow to cause vascular embolization. They are commonly used due to their ease of use and low cost. However, they can cause sid

Ultra-Efficient Solid-Phase Peptide Synthesis.

Microwave peptide synthesis offers significant advantages for peptide production in speed, efficiency, and overall purity. This chapter surveys key developments including the introduction of a wash-free methodology that has enabled 95% waste reduction in peptide synthesis at both research and production level solid-phase peptide synthesis (SPPS). Microwave SPPS is a widely used technique for R&D scale peptide synthesis with more recent expansion into larger scale current good manufacturing practice (cGMP).

A Degradable Macromolecular Antioxidant for Efficient Arthritis Treatment.

Reactive oxygen species (ROS) are vital but harmful in excess, and numerous small molecule antioxidants (e.g., polyphenols) can help to maintain an optimal ROS balance. Considering factors like biocompatibility, it is more commonly preferred to transform these natural antioxidants into nanoscavengers through a macromolecular engineering strategy. However, conventional macromolecular antioxidants typically exhibit decreased scavenging capacity compared to monomers due to steric hindrance and oxidation during the polymerization. To tackle this issue, we developed robust degradable macromolecular

Thiazolidinones and related analogues as efficient antitrypanosomal agents.

We designed and synthesised a series of thiazolidinediones and related analogues and evaluated their antiparasitic activity. A structure-activity relationship (SAR) study focused on modifications of specific parts of the molecule revealed derivatives that displayed significant activity against Trypanosoma brucei species. Notably, the analogue 6i exhibited exceptional activity, with an EC50 value of 30 nM and a selectivity index of >2000, against the protozoan Trypanosoma brucei rhodesiense, which causes human African trypanosomiasis. Additionally, compounds 6a, 6k, 7e, and 18 demonstrated anti

An efficient heuristic for geometric analysis of cell deformations.

Sickle cell disease causes erythrocytes to become sickle-shaped, affecting their movement in the bloodstream and reducing oxygen delivery. It has a high global prevalence and places a significant burden on healthcare systems, especially in resource-limited regions. Automated classification of sickle cells in blood images is crucial, allowing the specialist to reduce the effort required and avoid errors when quantifying the deformed cells and assessing the severity of a crisis. Recent studies have proposed various erythrocyte representation and classification methods (Jennifer et al., 2023 [1])

Efficient Photolysis of Multidrug-Resistant Polymicrobial Biofilms.

Chronic wounds are prone to infections with multidrug-resistant bacteria, forming polymicrobial biofilms that limit treatment options and increase the risk of severe complications. Current cleansing options are insufficient to disrupt and remove tenacious biofilms; antibiotic treatments, on the other hand, often fall short against these biofilm-embedded bacteria. This study explores an non-antibiotic approach that extends beyond conventional porphyrin-based phototherapy by using blue light (BL) in conjunction with ferric ions (Fe(III)) to disrupt and eradicate biofilms. The dual not only degra

Designing efficient randstrobes for sequence similarity analyses.

MOTIVATION: Substrings of length k, commonly referred to as k-mers, play a vital role in sequence analysis. However, k-mers are limited to exact matches between sequences leading to alternative constructs. We recently introduced a class of new constructs, strobemers, that can match across substitutions and smaller insertions and deletions. Randstrobes, the most sensitive strobemer proposed in Sahlin (Effective sequence similarity detection with strobemers. Genome Res 2021a;31:2080-94. https://doi.org/10.1101/gr.275648.121), has been used in several bioinformatics applications such as read clas
