First report in Africa of two clinical isolates of Proteus mirabilis carrying Salmonella genomic island (SGI1) variants, SGI1-PmABB and SGI1-W.

Two Proteus mirabilis strains, designated PmTAN59 and PmKAF126, were isolated from two different Egyptian cities in 2014 and 2015, respectively. PmTAN59 was isolated from a sputum swab from a pneumonia patient in Tanta University Teaching Hospital. PmKAF126 was isolated from a patient with a diabetic foot infection in a hospital in the city of Kafr El-Sheikh. The two isolates were identified with bacterial small ribosomal RNA (16S rRNA) gene amplification and sequencing and tested for antimicrobial sensitivity with a Kirby-Bauer disk diffusion assay. The two strains were resistant to amoxicill

Commensal Escherichia coli isolate resistant to eight classes of antimicrobial agents in the United States.

To increase understanding of community-acquired resistance, stool samples from 477 nonhospitalized persons in Maryland and Michigan, from 2004 to 2008, were screened for ceftriaxone resistance. Seven (1.5%) yielded ceftriaxone-resistant Escherichia coli; one isolate was resistant to all eight antimicrobial classes routinely tested: aminoglycosides, β-lactam/β-lactamase inhibitor combinations, cephems, penicillins, folate pathway inhibitors, phenicols, quinolones, and tetracyclines. The extensively resistant isolate was from a 50-year-old woman who denied antimicrobial use, hospitalization, or

Dynamics of clinical Klebsiella pneumoniae strains over the COVID-19 pandemic in Qingdao, China.

The dynamics of multidrug-resistant pathogens in response to large-scale societal and environmental disruptions remain poorly understood. Here, we investigated 528 Klebsiella pneumoniae clinical isolates collected from a tertiary hospital in Qingdao, China, spanning 2016 to 2023, with a primary focus on 2018-2023, across pre-, during-, and post-COVID-19 pandemic periods. All isolates underwent whole-genome sequencing. We identified SL258 and SL23 as dominant sequence lineages, with prevalent serotypes including K1 and O1. A wide range of resistance and virulence determinants were detected, par

Prevalence of multidrug-resistant Gram-negative pathogens isolated from febrile neutropenic cancer patients with bloodstream infections in Egypt and new synergistic antibiotic combinations.

INTRODUCTION: Bloodstream infections with multidrug-resistant (MDR) Gram-negative bacteria (GNB) are among the most frequent complications in immunocompromised cancer patients because of their considerable morbidity and mortality. Several guidelines on antimicrobial therapy have addressed empirical treatment for such serious infections; however, the emergence of microbial resistance has become a significant problem worldwide. MATERIALS AND METHODS: In this study, starting from November 2015 to October 2016, a total of 529 blood specimens were collected from febrile neutropenic cancer patients

Multidrug-Resistant Gram-Negative Bacteria and Extended-Spectrum β-Lactamase-Producing Klebsiella pneumoniae from the Poultry Farm Environment.

The indiscriminate use and overuse of various antibiotics have caused the rapid emergence of antibiotic-resistant bacteria (ARB) in poultry products and the surrounding environment, giving rise to global public health issues. This study aimed to determine the prevalence of multidrug-resistant (MDR) Gram-negative bacteria (GNB) found in the environment of poultry farms and to evaluate the risk of contamination in these farms based on multiple antibiotic resistance (MAR) index values. Soil and effluent samples were collected from 13 poultry farms. The VITEK 2 system was used for bacterial identi

Gene network interaction analysis to elucidate the antimicrobial resistance mechanisms in the Clostridiumdifficile.

Antimicrobial resistance has caused chaos worldwide due to the depiction of multidrug-resistant (MDR) infective microorganisms. A thorough examination of antimicrobial resistance (AMR) genes and associated resistant mechanisms is vital to solving this problem. Clostridium difficile (C. difficile) is an opportunistic nosocomial bacterial strain that has acquired exogenous AMR genes that confer resistance to antimicrobials such as erythromycin, azithromycin, clarithromycin, rifampicin, moxifloxacin, fluoroquinolones, vancomycin, and others. A network of interactions, including 20 AMR genes, was

Genome-Based Analysis of Chromosomal Colistin Non-Susceptibility in Stenotrophomonas pavanii Isolated from the Phycosphere of Pectinodesmus pectinatus.

BACKGROUND/OBJECTIVES: Freshwater microalgae-bacteria consortia are increasingly utilized in wastewater treatment and biomass production. However, bacteria associated with the algal phycosphere may act as environmental reservoirs of multidrug-resistant (MDR) phenotypes and antibiotic resistance genes (ARGs), including resistance to last-resort antibiotics such as colistin. METHODS: An axenic culture of the freshwater microalga Pectinodesmus pectinatus was established using a NaClO-based cleaning protocol. Three phycosphere-associated bacterial strains (Chryseobacterium sp., Pseudomonas montei

Eco-evolutionary responses to plasmid-dependent phage constrain the spread of multidrug-resistance plasmids.

Phage therapy offers an alternative to antibiotics for treating multidrug-resistant infections. Plasmid-dependent phages (PDPs) are promising therapeutics as they can kill targeted pathogens and prevent the spread of plasmid-encoded antibiotic resistance genes. However, the evolutionary trajectories of multidrug-resistance (MDR) plasmids under the selective pressure of PDPs remain poorly understood, particularly in eco-evolutionary contexts that remain permissive to plasmid conjugation. We experimentally evolved populations of Escherichia coli carrying the MDR plasmid RP4 in the presence of th

Phenotypic and genotypic screening of multidrug resistant Klebsiella pneumoniae isolated from ready to eat street food in Tanta, Egypt.

BACKGROUND: Ready-To-Eat-Street-Foods (RTESF) have food safety concerns, since they are prepared with less-structured food safety guidelines in small and roadside outlets. Klebsiella pneumoniae has become a dangerous foodborne-pathogen worldwide due to its virulence and resistance profile. OBJECTIVE: This study aimed at evaluating the potential burden of antibiotic-resistant Klebsiella pneumoniae contaminating RTESF and assessing the microbiological quality of RTESF in Egypt. METHODS: A total of 242 RTESF (green salad) samples was collected, different media were used for isolation of differe

Integrating computational approaches to uncover β-lactamase-associated resistance in diarrheagenic Escherichia coli from pediatric patients.

Multidrug-resistant diarrheagenic Escherichia coli (MDR-DEC) increasingly complicates treatment of pediatric infections, largely due to β-lactamase-mediated resistance. Despite their clinical relevance, the evolutionary diversity, functional mechanisms, and therapeutic targeting of β-lactamase enzymes in DEC remain poorly defined. This study addresses that gap by evaluating the prevalence of β-lactamase genes in DEC isolates and exploring their molecular interactions with key antibiotics. This study aims to detect and characterize β-lactamase-producing diarrheagenic Escherichia coli (DEC) isol

Comprehensive antibiotic resistome comparison of Escherichia coli from irritable bowel syndrome and ulcerative colitis.

The emergence of multidrug-resistant Escherichia coli (MDR E. coli), particularly enteropathogenic E. coli, is closely associated with therapeutic interventions for irritable bowel syndrome (IBS) and ulcerative colitis (UC) in clinical practice. However, a comprehensive characterization of their resistome differences remains limited. Exploring their resistance profiles and virulence gene repertoires is crucial for informing improved treatment strategies and controlling the dissemination of MDR E. coli in healthcare settings. Here, we analyzed 70 E. coli strains isolated from a single-center, c

Prediction of Phenotypic Antimicrobial Resistance Profiles From Whole Genome Sequences of Non-typhoidal Salmonella enterica.

Surveillance of antimicrobial resistance (AMR) in non-typhoidal Salmonella enterica (NTS), is essential for monitoring transmission of resistance from the food chain to humans, and for establishing effective treatment protocols. We evaluated the prediction of phenotypic resistance in NTS from genotypic profiles derived from whole genome sequencing (WGS). Genes and chromosomal mutations responsible for phenotypic resistance were sought in WGS data from 3,491 NTS isolates received by Public Health England's Gastrointestinal Bacteria Reference Unit between April 2014 and March 2015. GES-11-producing Acinetobacter baumannii clinical isolates from Tunisian hospitals: Long-term dissemination of GES-type carbapenemases in North Africa.

Acinetobacter baumannii is an emerging threat in healthcare facilities owing to its ability to be multidrug-resistant (MDR) and to be involved in outbreaks. GES-type extended-spectrum β-lactamases (ESBLs) have been increasingly identified in A. baumannii. In this study, clinical A. baumannii isolates were characterised using standard biochemical methods and antibiotic susceptibility testing. Antibiotic resistance genes were sought by PCR and sequencing. Genetic support was characterised using S1 nuclease pulsed-field gel electrophoresis (PFGE) mapping, conjugation and electroporation assays. T
