Cloning of a gene expressed in human breast cancer and regulated by estrogen in MCF-7 cells.

Messenger RNAs (mRNAs) were prepared from MCF-7 breast cancer cells grown in the presence of estradiol. Complementary DNAs (cDNAs) were inserted into pBR322 plasmid and a library of 4400 recombinant bacterial clones was prepared. The clones were screened by in situ differential hybridization with cDNAs prepared from RNAs of MCF-7 cells grown either in the presence or absence of estradiol. Several estrogen-induced or estrogen-repressed clones were identified. One of them corresponded to a relatively frequent mRNA (0.8% of recombinant plasmids) of 650 nucleotides. The concentration of this mRNA

Domesticated bacterial origin genes regulate homeostasis of the insect gut microbiome and empower adaptation.

Sap-sucking insects, represented by the globally distributed leafhoppers, have evolved a wide range of adaptive traits and play multiple ecological roles. However, their successful survival strategies in nature remain poorly understood. To explore the adaptive mechanisms of sap-sucking insects, we established a research system using three representative leafhopper species from the same family and found that they have acquired multiple cholesterol-dependent cytolysins (CDCs) derived from gram-positive bacterial genes. Furthermore, we demonstrated that these CDCs have been domesticated and co-ev

Neuronal activity-regulated histone demethylase KDM7A controls hippocampal gene expression and behavior.

The role of lysine demethylase 7A (KDM7A) in the mammalian brain remains largely unexplored. Here, we demonstrate that KDM7A occupies thousands of genomic loci in hippocampal chromatin and that both its expression and chromatin binding are dynamically regulated by neuronal activity in culture and in vivo. To define its functional relevance, we generated mouse models with either complete loss of KDM7A or disrupted enzymatic activity and assessed neurological phenotypes. Integrating cellular and molecular analyses with transcriptomic and chromatin-occupancy profiling and behavioral assessments,

Hilpda gene expression in steatotic liver is regulated by peroxisome proliferator-activated receptor γ.

Hypoxia-inducible lipid droplet-associated protein (HILPDA) promotes hepatic lipid accumulation. Hilpda gene expression is directly regulated by hypoxia-inducible factors (HIF1A and HIF1B) and the nuclear receptor peroxisome proliferator-activated receptor alpha (PPARα). However, whether the expression of Hilpda is regulated by peroxisome proliferator-activated receptor gamma (PPARγ) in fatty liver remains unclear. In this study, we investigated the role of PPARγ in regulating Hilpda expression in fatty liver. The analysis of publicly available microarray datasets revealed that Hilpda expressi

Cancer Gene Therapy Utilizing NF-κB-regulated Expression of a miRNA Targeting RelA.

BACKGROUND: Cancer continues to threat human health and life because of issues such as drug resistance and low response rates. The transcription factor NF-κB has emerged as a promising target for cancer therapy because of its overactivation in various cancers. However, existing NF-κB inhibitors lack cancer-cell specificity, leading to widespread suppression of essential physiological roles in immune regulation and cell survival, which results in systemic toxicity. METHODS: In contrast to the direct inhibition of NF-κB activity, this study introduces a novel antitumour strategy, DMP-miR533, whi

HOXA10 regulates apoptosis and expression of receptivity-related genes in bovine endometrial epithelial cells.

HOXA10 is a crucial marker gene for endometrial receptivity in mammals, with significantly increased expression in endometrial epithelial cells during the implantation window. However, the molecular mechanisms by which HOXA10 regulates bovine endometrial epithelial cells remain unclear. In this study, we constructed RNA interference fragments targeting the bovine HOXA10 gene, transfected them into bovine endometrial epithelial cells, and performed RNA-seq analysis. We conducted EdU staining, flow cytometry, RT-qPCR, and Western blot to assess apoptosis and the FOXO1 signaling pathway. Addition

Polyphosphate as a novel regulator of super-enhancer complexes: disruption of phase separation and gene expression.

Inorganic polyphosphate (polyP) is a highly conserved linear polymer of orthophosphate, present across nearly all living organisms. Its functional diversity, particularly through recently discovered post-translational modifications, has garnered increasing scientific interest. However, its role in gene regulation, especially in mammalian systems, remains unexplored. In this study, we present findings that demonstrate that polyP multi-targets the super-enhancer complex to regulate gene expression, marking a novel mechanism of transcriptional control. We show that polyP modifies key components o

Regulation of ER Stress-Associated Gene Expression by Lifeguard in Human Breast Cancer Cells.

BACKGROUND: Despite significant advances in breast cancer therapy, resistance to apoptosis remains a major obstacle to successful treatment. Lifeguard (LFG), an antiapoptotic membrane protein, has been implicated in breast cancer progression; however, its role in endoplasmic reticulum (ER) stress-associated molecular responses and apoptosis-related gene regulation remains incompletely understood. AIMS: This study aimed to investigate molecular changes associated with LFG-targeted siRNA treatment and reduced LFG mRNA expression in human breast cancer cells. METHODS AND RESULTS: The triple-neg

A negative role for master regulator Bcr1 in Candida albicans hypha-associated gene expression.

UNLABELLED: Filamentation, a central virulence trait of the fungal pathogen Candida albicans, is required for host cell damage, tissue invasion, and biofilm formation. The transcription factor Bcr1 was first found to be required for biofilm formation but not filamentation. Subsequent studies revealed a negative role for Bcr1 in filamentation in opaque cells, a cell type that is required for mating, under hypoxic conditions, and in a wor2 Δ/Δ mutant background. Here we characterize a new context in which Bcr1 negatively regulates filamentation and present its associated gene expression impact.

Investigating the Role of MicroRNA396 (miR396) Gene in Regulating Wheat Yield and Grain Nitrogen Concentration.

Nitrogen (N) is essential for crop growth, yet excessive fertilization causes environmental issues, highlighting the need to sustain yield and grain N concentration under reduced N input. miR396s are known to regulate plant development and stress responses. Here, we examined whether and how miR396 affects wheat yield and N status under high and low N conditions. TaMIM396 (transforming with the target mimicry construct of miR396) overexpression significantly increased plant height, spike length, grain yield, and grain N concentration under both N treatments. Physiological data showed TaMIM396 e

EL222-Based Optogenetic Gene Regulation in Methylotrophic Yeasts: Mechanisms, Applications, and Future Directions.

Methylotrophic yeasts such as Pichia pastoris are widely used for heterologous protein production because they contain strong and tightly regulated promoters. However, the use of methanol as an inducer presents several practical challenges, including toxicity, flammability, high oxygen demand during fermentation, and increased production costs. To overcome these limitations, researchers have been working on redesigning the AOX1 regulatory system and developing alternative induction strategies that do not rely on methanol. One promising approach is optogenetics, which uses light to control gene

Proline Dehydrogenase Gene TaProDH1 Positively Regulates Wheat Resistance to Stripe Rust Disease.

Proline dehydrogenase (ProDH) is a rate-limiting enzyme in the proline metabolism cycle that plays an important role in plant stress response and disease resistance. However, the function and mechanism of ProDH1 in the wheat-Puccinia striiformis f. sp. tritici (Pst) interaction remain poorly understood. In this study, a wheat proline dehydrogenase gene TaProDH1 was induced by stripe rust. qRT-PCR analyses showed that the transcript levels of TaProDH1 were highly increased during early infection. The transient expression of TaProDH1 in Nicotiana benthamiana leaves revealed that TaProDH1 increas

High-mobility group N proteins are required for regulation of antigen-presentation genes in macrophages.

The expression, localization, and secretion of certain chromatin proteins are tightly regulated in response to inflammatory challenges or tissue damage. High mobility group (HMG) nucleosome-binding proteins can translocate out of the nucleus to act as alarmins in immune cell communications. The consequences of nuclear depletion of HMGs in genome organization and gene regulation are not understood in the context of innate immunity. Here, we report alterations of tissue-specific macrophage transcriptome using a genetic knockout (KO) of HMGN1 and HMGN2, an HMGN double knockout mouse line. Lack of

Mechanisms of Diapause in Major Crustacean Taxa: Environmental Cues, Gene Regulation, and Adaptive Strategies.

Diapause serves as an adaptive dormancy strategy in freshwater, hypersaline, and marine crustaceans, allowing them to survive harsh conditions like temperature extremes, salinity fluctuations, hypoxia, and food scarcity. This review focuses on diapause across major crustacean taxa, with particular emphasis on branchiopods and copepods. However, detailed mechanistic evidence is more prevalent in well-studied species like Artemia, Daphnia, and calanoid copepods, leading to an uneven distribution of evidence across these taxa. Thus, the molecular and physiological mechanisms discussed in this rev

Cornea Arcus and Dyslipidemia in Unrelated Adults With Monoallelic Germline Variants in the Glucokinase Regulator Gene.

Variants in the Glucokinase Regulator (GCKR) gene are increasingly being detected with growing applications of genomic sequencing technologies. Despite the crucial roles of the GCKR gene in energy homeostasis, the clinical applications of GCKR variants are currently limited. We observed that a large proportion of GCKR variants that are available in gnomAD database (https://gnomad.broadinstitute.org/) lacked adequate evidence of pathogenicity or benign impact. We highlight the clinical need for an improved understanding of GCKR variants by presenting 2 compelling cases of unrelated families (3

Hormonal regulation of cochlear gene expression: effects of the estrous cycle, ovariectomy, and estradiol treatment.

BACKGROUND: The significance of sex differences in auditory function and vulnerability to hearing disorders has gained increasing attention. However, the underlying biological mechanisms remain unclear. Estrogen and other female hormones are known to regulate gene expression in various organs and tissues. However, their role in regulating gene expression in the cochlea remains to be determined. The current study investigated how female hormonal status, through natural fluctuations of the estrous cycle and hormonal deprivation via ovariectomy, modulates cochlear gene expression in mice. PPARα and RXRα in the regulation of neuronal ceroid lipofuscinosis genes: implications for Batten disease therapy.

Neuronal ceroid lipofuscinosis or Batten disease comprises a category of autosomal recessive neurodegenerative disorders that primarily affect children. Mutations in different genes lead to different forms of neuronal ceroid lipofuscinoses (CLN1-14). At present, there is no established therapy to cure most of the neuronal ceroid lipofuscinoses and the treatments are symptomatic. Enzyme replacement therapy, gene therapy, stem cell transplantation, and pharmacological chaperone therapy are being tested in different animal models and human patients.
