Combined association of genetic predisposition and healthy lifestyle with lung cancer risk: a population-based cohort study.

BACKGROUND: Polygenic susceptibility and modifiable lifestyle behaviors contribute to lung cancer, but their interaction and combined impact on lung cancer hazard and potential biological correlations remain incompletely characterised. This study aimed to evaluate the independent and combined associations of polygenic risk and healthy lifestyle with lung cancer hazard and to explore potential biological correlations underlying these associations. METHODS: We analyzed data of 254,874 UK Biobank (UKB) participants. A five-component lifestyle score (smoking, alcohol intake, diet, physical activi

Exploring genetic diversity in genome-wide association studies and polygenic risk scores in the admixed Brazilian population.

The Brazilian population has a complex genetic background resulting from admixture of Native American, African, and European ancestries. This review examines how such diversity influences the findings of genome-wide association studies and the performance of polygenic risk score. It also highlights the role of admixture in elucidating the genetic basis of complex diseases, and addresses key methodological challenges, including population stratification and limited representation in genomic databases. Furthermore, it evaluates the transferability of risk models developed in other populations. T

Enhanced lymphotoxin alpha DNA hypermethylation and correlation with genetic polymorphism in rheumatoid arthritis among the Chinese population.

BACKGROUND: Rheumatoid arthritis (RA) is a multifactorial autoimmune disorder characterized by persistent inflammation and joint destruction. Despite advances in understanding RA's immunopathology, its epigenetic regulation remains underexplored, particularly in diverse populations. This study aims to bridge this gap by examining lymphotoxin alpha (LTA) DNA methylation in a Chinese cohort and exploring the relationship with gene polymorphism. METHODS: This study involved 145 RA patients and 140 age-matched healthy controls from Qingdao, China. We analyzed DNA methylation in the LTA gene using

Determinants of Vitamin D in Indian Populations: Understanding the Interplay of Skin Pigmentation, Sun Exposure, and Genetic Variants.

OBJECTIVES: The present study evaluates the effects and interaction of skin pigmentation, sun exposure, and genetics on 25(OH)D3 levels among ostensibly healthy Indians. METHODS: Vitamin D3 was estimated (N = 427) and means were compared between the sexes and study populations. Vitamin D3 was correlated with Melanin Index (MI) and Weekly Sun Exposure Scores (WSES) using Kendall's tau-b (τb). The effect of age, sex, MI, WSES, and 12 SNPs from vitamin D and skin pigmentation genes were tested on 25(OH)D3 levels using univariate and multivariate linear models. RESULTS: A total of 76.6% of the s

Identifying causal genetic variants for high-altitude adaptation through blood eQTL analysis in plateau populations.

A substantial number of genetic variants have been associated with high-altitude adaptation (HAA), yet most of them are located in non-coding genomic regions, leaving their specific functions and underlying mechanisms largely unknown. In this study, we analyze whole-genome and transcriptome sequencing data from a self-established cohort comprising 61 native highlanders (NHs) and 164 acclimatized newcomers (ANs), identifying 6,586 cis- and 34,203 trans-expression quantitative trait loci (eQTLs), along with 130 cell type-specific eQTLs. By further combining these data with a large East Asia (~30

Social Disconnection, Genetic Risk, and the Incidence of Degenerative Valvular Heart Disease: A Population-Based Cohort Study.

BACKGROUND: Degenerative valvular heart disease (VHD) is increasingly recognized as a major public health concern in aging populations. Although traditional cardiovascular risk factors have been extensively investigated, the role of loneliness and social isolation has remained largely overlooked in the context of valvular pathology. METHODS: We conducted a prospective cohort study involving 462 917 participants from the UK Biobank, free of VHDs at baseline. Loneliness and social isolation were assessed using validated questionnaires. RESULTS: During a median follow-up of 13.9 years, 11 003 c

Genetic overlap between schizophrenia and constipation: insights from a genome-wide association study in a European population.

BACKGROUND: Patients with schizophrenia (SCZ) experience constipation at significantly higher rates compared with the general population. This relationship suggests a potential genetic overlap between these two conditions. METHODS: We analyzed genome-wide association study (GWAS) data for both SCZ and constipation using a five-part approach. The first and second parts assessed the overall and local genetic correlations using methods such as linkage disequilibrium score regression (LDSC) and heritability estimation from summary statistics (HESS). The third part investigated the causal associat

Population genetic structure and historical demography of Saccostrea echinata in the Northern South China sea and Beibu Gulf.

Marine invertebrates, such as oysters, were once thought to form large, panmictic populations with little genetic differentiation due to their high reproductive capacity and dual life stages. However, recent studies have shown significant genetic structuring and moderate gene flow across populations, influenced by factors like ocean currents, historical climate events, and environmental changes. The black-lip oyster (Saccostrea echinata), with its extensive dispersal potential, is ideal for population genetics studies. In current study, mitochondrial DNA (COI gene) was utilized to investigate

Genetic variants associated with chronic fatigue syndrome predict population-level fatigue severity and actigraphic measurements.

STUDY OBJECTIVES: The diagnosis of myalgic encephalomyelitis/chronic fatigue syndrome (CFS) is based on a constellation of symptoms which center around fatigue. However, fatigue is commonly reported in the general population by people without CFS. Does the biology underlying fatigue in patients with CFS also drive fatigue experienced by individuals without CFS? METHODS: We used UK Biobank actigraphy data to characterize differences in physical activity patterns and daily temperature rhythms between participants diagnosed with CFS compared to controls. We then tested if single nucleotide varia
