Herpes simplex virus type 1 replicates in the lens and induces cataracts in mice.

In a mouse model, cataracts were produced after corneal inoculation with herpes simplex virus type 1 (HSV-1) strain F. Viral antigen was detected in vivo by the immunoperoxidase technique in lens fibers and occasionally in lens epithelial cell nuclei at 4 and 5 days postinoculation. HSV-1 replication within the lens was further demonstrated by virus isolation from lenses that had been inoculated in vitro. Lens lesions in mice were present as early as 4 days postinoculation and by later times (23 to 60 days postinoculation), severe degenerative changes typical of cataracts were present in many …

The effect of flurbiprofen on herpes simplex virus type 1 stromal keratitis in mice.

The use of steroidal compounds to reduce the inflammation and scarring associated with herpes simplex virus type 1 (HSV-1) stromal keratitis can result in severe exacerbation of the corneal disease. We compared the nonsteroidal anti-inflammatory drug (NSAID) flurbiprofen sodium with dexamethasone for the treatment of HSV-1 induced corneal stromal disease in an inbred mouse model. Stromal disease was induced by the direct intrastromal injection of HSV-1. A stromal opacity and corneal neovascularization (CNV) developed in 100% of the injected eyes, with no epithelial involvement until late in th…

Herpes simplex virus stromal keratitis is not titer-dependent and does not correlate with neurovirulence.

We developed a murine model of ocular herpes simplex virus (HSV) disease which is particularly suited for testing stromal keratitis because most animals show some evidence of infection. Using this model, we characterized the ocular disease patterns caused by ten recent low-passage clinical isolates of HSV-1, as well as those caused by the established laboratory strains HSV-1 KOS and HSV-2 333. Viral strains were evaluated for their ability to cause stromal keratitis, blepharitis, vascularization of the cornea, and mortality. The model was not useful for scoring epithelial keratitis. The ocular…

Induction of cervical neoplasia in the mouse by herpes simplex virus type 2 DNA.

Induction of cervical neoplasia in the mouse cervix by herpes simplex virus types 1 (HSV-1) and 2 (HSV-2) has been reported. The present study was done to determine if transfection with DNA of HSV-2 can induce carcinogenesis in this animal model. Genomic HSV-2 DNA was isolated from infected HEp-2 cells and separated from host cell DNA by cesium chloride density gradient centrifugation. The DNA was applied to mouse cervix for periods of 80-100 weeks. Experimental controls were treated with uninfected genomic HEp-2 cell DNA or with calf thymus DNA. Vaginal cytological preparations from all anima…

Shedding of herpes simplex virus type 1 into saliva in patients with orofacial fracture.

Shedding of herpes simplex virus type 1 (HSV-1) into saliva was studied in 83 patients with orofacial fractures. Infectious virus was isolated from 14 of 83 patients (16.8%) with no detectable herpetic lesion during hospitalization. Of the 83 patients, 44 (53%) had HSV-1 specific antibody. Virus shedding into saliva was observed only in the patients with antibody to HSV-1; thus, the frequency of virus shedding patients among those with antibody was 31% (14 of 44 patients). The frequency was obviously higher than that of a healthy population. The period of HSV-1 shedding had a mean of 3.7 days …

Usefulness of clinical features and liver biopsy in diagnosis of disseminated herpes simplex infection.

With the advent of effective antiviral treatment for disseminated herpes infection it has become important to be able to make a firm clinical diagnosis of this condition. We undertook a retrospective analysis of 20 patients with disseminated herpes simplex infection diagnosed at necropsy to derive useful clinical pointers to this severe form of the disease. The most useful clinical criteria were malnutrition, post measles state, and local herpes infection in addition to encephalopathy, enlarging hepatomegaly, and deteriorating pneumonia. The liver showed characteristic histological changes in …

Leptomeningitis and polycaryocyte formation in the CNS of rats inoculated subcutaneously with herpes simplex virus.

Wistar rats were inoculated subcutaneously with either type 1 (HSV1) or type 2 (HSV2) Herpes simplex virus at 5 days of age. Animals were killed in extremis or at the end of the 14-day observation period postinoculation. Acute destructive meningoencephalitis with hemorrhage and leukocytic infiltration was observed in both groups. Polycaryocytes comprised of cells of the internal granular layer of the cerebellum were observed in some animals inoculated with HSV1. These multinucleated cells appeared to be formed by fusion of virus-infected cells, and intranuclear inclusion bodies were observed. …

ELISA for the detection of herpes simplex virus antigens in the cerebrospinal fluid of patients with encephalitis.

An inhibition enzyme-linked immunosorbent assay (ELISA) for the detection of herpes simplex virus antigens in cerebrospinal fluid (CSF) has been developed. A Triton X-100 extract of herpes simplex virus type 1 (HSV-1) infected HEp-2 cells was used to coat wells of polyvinyl chloride plates. Rabbit anti-HSV-1 globulin served as the reference antibody and the CSF specimens were tested at a final dilution of 1:4. Positive results were obtained in CSF specimens from 11/18 (61%) neonates with HSV infection, 15/23 (65%) older individuals with HSV culture positive brain biopsies, and in 4/29 (14%) pa…

Herpes simplex encephalitis. An immunohistological study of the distribution of viral antigen within the brain.

An immunoperoxidase technique was used to map the sites of herpes simplex virus antigen (VA) within the brain in 29 autopsied cases of herpes simplex encephalitis. Attention was directed particularly to those parts of the brain that are known from pathological studies to be involved in the disease. Material was studied from cases surviving for varying periods from a few days to a few years after the onset of neurological disease. VA was found within the brain in all cases dying within 3 weeks of onset, but in none dying thereafter. VA was already most abundant in patients dying within the firs…

A great nursing challenge--recognition and effective management of the patient with herpes simplex encephalitis.

There are many difficulties associated with management of persons suspected or diagnosed with HSE. Excellent coordinated care is critical and the health care team must devote particular attention to these patients' complex clinical problems, including nutrition. While Vira-A therapy as the treatment for diagnosed HSE is significant, it may be associated with difficulties in fluid overload. Other problems are likely to evolve around a serious illness that progresses rapidly and is associated with fever, seizures, and increased intracranial pressure.

Fulminant herpes simplex hepatitis in an adult: report of a case in renal transplant recipient.

A case of disseminated herpes simplex infection is reported in a 31-year-old renal transplant recipient. The patient presented with a unique clinical syndrome: high fever, severe sore throat with buccal and pharyngeal ulcerations, fulminant hepatitis, thrombocytopenia, and leukopenia. The patient died from hepatic failure, disseminated intravascular coagulopathy, and upper gastrointestinal bleeding. The diagnosis was made by positive herpes simplex virus culture from the throat, and was confirmed at autopsy by typical Cowdry's type A intranuclear inclusions in hepatocytes with positive herpes …

Antiviral Treatment of Herpes Simplex Virus Decreases the Risk of Alzheimer's Disease and Dementia.

BACKGROUND: Alzheimer's disease (AD) and dementia create major global health and economic burdens. Herpes simplex virus (HSV) infects over 3 billion people, and chronic infection is increasingly linked to neurodegeneration. OBJECTIVES: To evaluate whether antiviral therapy for oral, mucocutaneous, or anogenital HSV lowers the subsequent risk of AD and dementia. METHODS: A retrospective cohort study with propensity-score matching was performed in the TriNetX Research Network. On 24 May 2024, 615,324 individuals with HSV were identified; those with prior AD, intracranial injury, or cerebral in…

Development of a HiFi-LAMP assay for the detection of herpes simplex virus type 1.

Herpes simplex virus type 1 (HSV-1) has a significant impact on global public health. Developing a rapid, sensitive, and specific diagnostic assay for the detection of HSV-1 is of critical clinical significance for guiding individualized therapy and optimizing outcomes. A high-fidelity loop-mediated isothermal amplification (HiFi-LAMP) assay was developed by targeting the highly conserved region of the HSV-1 US4 gene. The sensitivity of the assay was determined using serial dilutions of standard templates, and specificity was assessed against ten common pathogens. Intra- and inter-assay reprod…

Herpes simplex virus esophagitis in an immunocompromised patient: case report of an atypical presentation.

We present the case of a 28-year-old male with diarrhea, headache, and significant weight loss. He was diagnosed with HIV in the AIDS stage (CD4: 3 cells/μL), Cryptococcus neoformans meningoencephalitis, and multisystemic tuberculosis. Given the persistence of diarrhea and negative coproparasitological studies, endoscopic studies were performed. Upper digestive endoscopy incidentally showed esophageal erosions coated with fibrin and punched-out ulcers; these findings were identified as herpetic esophagitis by histopathological and immunohistochemical analysis. Treatment with intravenous acyclo

A Rare Finding of Herpes Simplex Virus Esophagitis Concomitant With Duodenitis in an Immunocompetent Host.

Herpes simplex virus (HSV) esophagitis and duodenitis are well-known features of herpes virus reactivation in immunosuppressed patients. We discuss the case of HSV esophagitis concomitant with duodenitis in an immunocompetent individual. A 77-year-old male patient, previously healthy, was found to have on upper endoscopy several ulcers in the esophagus and second portion of the duodenum. A diagnosis of HSV esophagitis concomitant with duodenitis was made. The patient was started on intravenous acyclovir, and 48 hours after treatment, his symptoms significantly improved. He was then discharged …

Seroepidemiology of Herpes Simplex Viruses Type 1 and 2 in Pregnant Women in Croatia.

Background and Objectives: Herpes simplex viruses (HSV-1 and HSV-2) are one of the most widespread causes of human viral infections. In Croatia, only two published studies have analyzed the seroprevalence of HSV infections in childbearing-aged and pregnant women (2005-2010), while more recent data are lacking. This study aimed to analyze the prevalence and risk factors for HSV-1 and HSV-2 infections among pregnant women in Croatia in the period from 2011 to 2021. Materials and Methods: This study included 667 pregnant women aged 16-45 years submitted for HSV-1 and HSV-2 serology testing. Serum…
