The use of self-tracking of bio-behavioral states along with prescription dosing information is increasingly popular in the care and study of many human diseases. Parkinson's Disease is particularly amenable to such tracking, as patients live with the progressive disease for many years, increasing motivation to pursue quality of life changes through careful monitoring of symptoms and self-guided management of their medications and lifestyle choices. Through the use of digital self-tracking technologies, patients independently or in conjunction with professional medical advice are modulating th

Preclinical drug testing in primary human cell models that recapitulate disease can significantly reduce animal experimentation and time-to-the-clinic. We used intestinal organoids to quantitatively study the potency of multivalent cholera toxin inhibitors. The method enabled the determination of IC50 values over a wide range of potencies (15 pM to 9 mM). The results indicate for the first time that an organoid-based swelling assay is a useful preclinical method to evaluate inhibitor potencies of drugs that target pathogen-derived toxins.

The Nuremberg Code is a foundational document in the ethics of medical research and human experimentation; the principle its authors espoused in 1946 have provided the framework for modern codes that address the same issues, and have received little challenge and only slight modification in decades since. By analyzing the Code's tragic genesis and its normative implications, it is possible to understand some of the essence of modern experimental ethics, as well as certain outstanding controversies that still plague medical science.

Primary human fallopian tube secretory epithelial cell (FTSEC) cultures are useful for studying normal fallopian tube epithelial biology, as well as for developing models of fallopian tube disease, such as cancer. Because of the limited ability of primary human FTSECs to proliferate in vitro, it is necessary to immortalize them in order to establish a cell line that is suitable for long-term culture and large-scale in vitro experimentation. This protocol describes the isolation of FTSECs from human fallopian tube tissue, conditions for primary FTSEC culture and techniques for establishing immo

EMBRYONIC DEVELOPMENT IS INFLUENCED BY THREE ENVIRONMENTS: the intra-uterine, the maternal body and the maternal surroundings. Factors present in one or other may cause abnormal development.Usually environmental factors act in association with genetic factors but they can be the dominant or sole cause of birth defects. Many malformation-producing agents exist in the maternal environment and some cause abnormalities in man. Use of such agents in experimental animals yields valuable information on how malformations develop. Different species and strains of animals often react differently to the

INTRODUCTION: Up to 40% of major complications during airway management result from human factors. Through human factors such as planning, effective communication, strong leadership and clear role allocation within the team, smooth airway securement can be facilitated and the risk of complications reduced. Simulation-based teaching provides real-time situations and allows healthcare workers and learners to identify, refine and improve their technical and non-technical skills. This study aims to evaluate the effect of simulation-based human factors training on airway management among healthcare

OBJECTIVE: Toll-like receptor 4 (TLR4) is one of the key players in the development of many autoimmune diseases. To determine the possible role of TLR4 in polymyositis (PM) development, we collected muscle samples from PM patients and mice subjected to an experimental autoimmune myositis (EAM) model.
METHODS: We measured TLR4-MyD88 pathway-related factors, interferon-γ (IFN-γ), and interleukin-17A (IL-17A) in EAM mice and PM patients. Then, we observed the changes of above factors and the inflammatory development of EAM mice with TLR4 antagonist TAK-242, IFN-γ, or IL-17A antibody treatment.

BACKGROUND: The use of transanal laparoscopic access to completely avoid abdominal wall incisions represents the most current evolution in minimally invasive surgery. The combination of single-site surgery and natural orifice transluminal endoscopic surgery (NOTES™) can be used for totally transanal laparoendoscopic pull-through colectomy with J-pouch creation (TLPC-J). The aim of the present study was to provide evidence for the feasibility of TLPC-J in adult human cadavers.
METHODS: TLPC-J was performed in six fresh adult human cadavers. The procedure involved endorectal submucosal dissecti

In vitro models are currently being used to study head and neck squamous cell carcinoma (HNSCC). Several hundred HNSCC cell lines have been established by various investigators and used to study a broad spectrum of questions related to head and neck cancer. The head and neck model with respect to multistage carcinogenesis is now complete. Several techniques exist for the culture of normal epithelial cells from the upper aerodigestive tract (UADT). The biology of these UADT cells (oral cavity, oropharynx, hypopharynx and larynx) is being studied. Successful culture of premalignant lesions (dysp

We described here the characterization of Schistosoma mansoni egg antigens recognized by human monoclonal antibodies B10 (HmAb-B10) and D5 (HmAb-D5). SDS-PAGE and Western blot analysis revealed that these monoclonals recognized two antigens of M W 44.7/56.8 kDa, with pI of 7.0 and 7.8, respectively. The passive transfer of B10 and D5 induced a significant protection of 48% and 54% in Balbic mice. Results of in vitro cytotoxicity assay showed that both monoclonals were able to kill schistosomula in the presence of rabbit complement. These monoclonals mediated 48% and 74% of schistosomula cytoto

The pancreas is a vital gland of exocrine and endocrine function. It is the target of two main affections: diabetes and pancreatic cancer. We describe the tissue interactions, signaling pathways and intracellular targets that are involved in the emergence of the pancreas primordium and its proliferation, morphogenesis and differentiation. It appears that several genes of developmental relevance have an adult function and are involved in pancreas affections. Embryological experimentation in animals contributed to provide candidate genes for human disease and holds promise for future treatments.

Systemic anthrax infection is usually fatal even with optimal medical care. Further insights into anthrax pathogenesis are therefore urgently needed to develop more effective therapies. Animal models that reproduce human disease will facilitate this research. Here, we describe the detailed histopathology of systemic anthrax infection in A/J mice infected with Bacillus anthracis Sterne, a strain with reduced virulence for humans. Subcutaneous infection leads to systemic disease with multiple pathologies including oedema, haemorrhage, secondary pneumonia and lymphocytolysis. These pathologies be

Research involving whitening agents requires several steps of experimentation, and the initial step is to test whitening agents with human melanocytes and those with human tyrosinase. Unfortunately, it takes a long time to gather human melanocytes, and these cells have some limitations when it comes to performing experiments, such as their passage difficulties and their cost. In this study, we suggest that the TXM13 human melanoma cells could be a useful cell candidate for studying human tyrosinase inhibition and depigmentation. We applied a tyrosinase inhibitor, such as dithioglycerine (DTGC)

The objective of this study was to describe a novel minimally invasive robotic video-assisted approach for lung transplantation, utilizing a minimally invasive technique with a subxiphoid incision, in an animal experimentation model. Two left robotic-assisted single lung transplants were performed in sheep using a robotic surgical system. A subxiphoid incision was made, and robotic ports were inserted into the thoracic cavity for dissection and anastomoses of the bronchus, artery, and pulmonary veins. The integrity of anastomoses was evaluated, and procedural details were recorded. Both animal

We present a historical account of the story behind the famous hawk/goose experiments of Lorenz and Tinbergen in a wider context of cognitive ethology. We discuss their significance, for ethological experimentation in general, and specifically for understanding innate constraints on cognition. As examples of the continuing significance of the hawk/goose paradigm of selective habituation, we discuss its relation to "exposure therapy" of human phobias and the use of hawk silhouettes as deterrents for songbirds. Finally we rephrase Uexküll's thesis of taxon-specific worlds ("Umwelten") as a "Theo

The isolation and purification of insulin nearly 100 years ago has been one of the most ground-breaking discoveries in the history of medicine. Subsequent determination of the specific structure of human insulin has permitted further developments and modifications of the formulations of insulin to allow improved quality of care and quality of life for patients with diabetes. In this paper, we will review insulin structure and biosynthesis, treatment and prognosis of type 1 diabetes prior to insulin therapy, experimentation leading to the discovery of insulin, and the first patients to be treat

The purpose of this study is to evaluate the activities of the I.G. Farben laboratory at the former "Heil- und Pflegeanstalt" Günzburg. This laboratory was established to test the newly developed epilepsy drug "Citrullamon" and its derivatives. Specifically, the type and manner of the various experiments were examined to determine whether the suspicions of unethical human experimentation could be identified. The commercial and medical activities between I.G. Farben and the Heil- und Pflegeanstalt, including the specific roles of the senior physician Wilhelm Leinisch and the I.G. Farben chemist…

Induced human extended pluripotent stem cells (hEPSC) have been injected into monkey blastocysts to create chimeric embryos. Chimera research is a scientific technique which seeks to develop human organs for transplantation, but it has raised concerns regarding human cells contributing to the brain formation of nonhuman animals. This article considered whether brain development should guide ethical considerations on chimera research by investigating the morality of terminating live anencephalic infants to procure their organs. It found that identifying the presence or absence of a human biolog

Genetic sensorineural hearing loss and Meniere disease have been associated with rare variations in the coding and non-coding region of the human genome. Most of these variants were classified as likely pathogenic or variants of unknown significance and require functional validation in cellular or animal models. Given the difficulties to obtain human samples and the raising concerns about animal experimentation, human-induced pluripotent stem cells emerged as cellular models to investigate the interaction of genetic and environmental factors in the pathogenesis of inner ear disorders. The gene

With the increasing number of patients affected with metabolic diseases such as type 2 diabetes, obesity, atherosclerosis and insulin resistance, academic researchers and pharmaceutical companies are eager to better understand metabolic syndrome and develop new drugs for its treatment. Many studies have focused on the nuclear receptor peroxisome proliferator-activated receptor gamma (PPARγ), which plays a crucial role in adipogenesis and lipid metabolism. These studies have been able to connect this transcription factor to several human metabolic diseases. Due to obvious limitations concerning
