Isonicotinic acid hydrazide (INH), a well-known and promising first-line anti-tubercular agent, is enlisted in the World Health Organization's (WHO) list of vital medicines. Herein, the binding characteristics of more efficacious INH-derivatives with human serum albumin (HSA) were investigated using multiple spectroscopic techniques combined with molecular modelling approaches. The findings suggested that all the derivatives quench the intrinsic fluorescence of HSA via the static mode of quenching, validated by UV-visible differential spectroscopy. The stoichiometry (n) and binding constant (K

Mass spectrometry combined with stable isotope labeling is a powerful technique for detecting disease-related changes in glycosylation patterns and identifying potential biomarkers. However, stable isotope labeling reagents that simultaneously offer high sensitivity, low cost, and stable sialic acid modifications remain scarce. In this study, we developed a convenient and cost-effective microwave-assisted method for synthesizing a stable isotopic quaternary phosphonium hydrazide labeling reagent pair, 14N/15N-P4HZD, for the quantitation difference analysis of N-glycans using HPLC-ESI-HRMS with

Bacterial biofilms represent a major clinical challenge due to their high tolerance to conventional antibiotics and their implication in persistent infections. Staphylococcus aureus is particularly known for its ability to form biofilms that significantly reduce antibiotic efficacy. In this context, the development of new molecules capable of eradicating mature biofilms is of great interest. Herein, we report the synthesis of 20 novel hydroxamic acid and hydrazide containing-imidazole compounds and the evaluation of their antibacterial effect on a panel of bacteria from the ESKAPE group includ

A 76-year-old woman with stage IIIB squamous cell lung carcinoma developed pancytopenia following maintenance therapy with durvalumab. She was diagnosed with squamous cell lung carcinoma (T4N2M0, stageIIIB) in December 2023. Following a favorable response to chemoradiotherapy, maintenance therapy with durvalumab was initiated in June 2024. Thrombocytopenia was observed starting in August, and platelet count had decreased to 22,000/µl in October, leading to the discontinuation of durvalumab. However, the condition progressed to pancytopenia. Bone marrow biopsy revealed hypocellular marrow witho

Aplastic anemia (AA) is an inherited, idiopathic, or acquired syndrome of bone marrow failure characterized by pancytopenia and ineffective hematopoiesis. Diagnosis, while crucial, is often difficult due to required exclusion of numerous inherited or acquired diseases with similar phenotypes. Mortality from severe AA without treatment approaches 70% within 2 years. The diagnostic algorithm for AA has increased in complexity, now incorporating molecular and genetic testing, and AA treatment guidelines have evolved to optimize patient outcomes. For individuals younger than age 50 years, a matche

BACKGROUND/AIM: Anamorelin, a ghrelin receptor agonist, increases body weight and lean body mass (LBM); however, its effects on the gut microbiota remain unclear. Furthermore, the predictive value of nutritional scores, such as the Controlling Nutritional Status (CONUT) score, for anamorelin response is not established. This study aimed to evaluate the effects of anamorelin on body weight, quality of life (QOL), and gut microbiota in patients with advanced cancer and cachexia, and to clarify the predictive role of nutritional indicators.

BACKGROUND: Pediatric primary immune thrombocytopenia (ITP) is an autoimmune disorder characterized by isolated thrombocytopenia and an increased risk of bleeding. Conventional therapies, while effective in some cases, are often limited by suboptimal response rates and significant adverse effects with prolonged use. Thrombopoietin receptor agonists (TPO-RAs), including recombinant human thrombopoietin (rhTPO), romiplostim, and eltrombopag, have emerged as promising therapeutic alternatives for pediatric ITP. However, a comprehensive comparison of their efficacy and safety profiles remains lack

INTRODUCTION: Thrombocytopenia is a common clinical problem in cancer patients undergoing high-dose chemotherapy and autologous hematopoietic stem cell transplantation (HSCT). It can occur as prolonged isolated thrombocytopenia (PIT) or secondary failure of platelet recovery (SFPR) and may cause potentially fatal bleeding. However, data on the treatment of post-transplant thrombocytopenia is still lacking.
METHODS: We reported our practices involving 15 pediatric patients who received eltrombopag (ELT) treatment for PIT and SFPR after autologous HSCT.
RESULTS: The overall response was 78.5%

Paediatric immune thrombocytopenia treatment was revolutionized with the advent of the thrombopoietin mimetics romiplostim and eltrombopag. For eltrombopag, dosing recommendations have been based on patients' ages (with dose reductions for those with hepatic impairment and for some of East Asian ethnicity) and titrated based on platelet counts and adverse drug reactions. However, it is clear that identical eltrombopag dosing for paediatric patients can result in variable side effects and platelet counts, raising the question as to whether variable eltrombopag plasma concentrations are responsi

Avatrombopag is the newest thrombopoietin receptor agonist (TPO-RA) approved to treat immune thrombocytopenia (ITP). Real-world evidence regarding effectiveness/safety is limited. The Spanish ITP Group (GEPTI) performed a retrospective study with patients starting avatrombopag for the first time. A total of 268 ITP patients were recruited. The median (interquartile range [IQR]) follow-up time was 47.5 (30.4-58.9) weeks. Among the 193 patients with baseline platelet counts <50 × 109/L, 174 (90.1%) of them achieved response (PC ≥50 × 109/L), and 113 (87.6%) of the 129 who persisted on avatrombop

BACKGROUND: The significant increase in patients suffering from different types of cancer, guides scientists to take prompt measures in the development of novel and effective antiproliferative agents, where the intercalation of heterocyclic fragments in the designed molecules has proven to be a useful practice.
OBJECTIVE: The newly synthesized compounds were obtained from the corresponding 1,4-dicarbonyl derivative through multicomponent reactions to produce biologically active target molecules and assessed by in silico and in vitro assays for their possible antitumor activity.

ABSTRACTThrombocytopenia is one of the most common hematological adverse reactions in chronic myeloid leukemia (CML) patients receiving tyrosine kinase inhibitors (TKI) therapy, causing life-threatening bleeding cases. However, there are fewer therapeutic drugs for TKI-induced thrombocytopenia. Eltrombopag is a non-peptide thrombopoietin receptor agonist used for the treatment of immune thrombocytopenia, aplastic anemia, and hepatitis C-associated thrombocytopenia. Nevertheless, studies of eltrombopag for TKI-induced thrombocytopenia are still lacking. This study retrospectively analyzed the c

Approximately 20-50% patients with chronic phase chronic myeloid leukemia (CML-CP) treated with tyrosine kinase inhibitors (TKIs) or with myelofibrosis (MF) treated with ruxolitinib develop grade ≥3 thrombocytopenia needing treatment interruptions and dose reductions. We conducted a non-randomized, phase II, single-arm study to determine the efficacy of eltrombopag for patients with CML or MF with persistent thrombocytopenia while on TKI or ruxolitinib. Eltrombopag was initiated at 50 mg/day, with dose escalation up to 300 mg daily allowed every 2 weeks. Twenty-one patients were enrolled (CML=

Despite a moderate prevalence in low-risk myelodysplastic syndromes (MDS) and chronic myelomonocytic leukaemia (CMML), thrombocytopenia remains a risk of severe bleeding and therapeutic options are still limited. There are only a few studies with eltrombopag (ELT), a thrombopoietin receptor agonist, in those patients. In this retrospective multicentre study, ELT was used in 50 patients with MDS and 11 with CMML, with no excess of marrow blasts and platelet counts of <50 × 109 /l in a 'real-life' situation. Platelet response occurred in 47 (77%) patients. The median (range) duration of response

Post-transplantation thrombocytopenia (PT) is a common and severe complication which usually leads to poor prognosis. Eltrombopag (EPAG), a novel oral thrombopoietin (TPO) receptor agonist, has shown promising effects in thrombocytopenia due to immune thrombocytopenic purpura (ITP) and refractory severe aplastic anemia (rSAA), while the effectiveness of EPAG for PT patients still needs to be evaluated. A total of 32 PT patients receiving EPAG were retrospectively analyzed between September 2017 and July 2019, including 15 patients with poor graft function (PGF) and 17 patients with secondary f

OBJECTIVE: The bioavailability of the non-hormonal male contraceptive adjudin is low in rats due to the blood-testis barrier (BTB). This study was designed to examine if F5-peptide, an endogenously produced reversible BTB modifier, could enhance the bioavailability of adjudin to affect spermatogenesis and provide a contraceptive effect in rats while reducing systemic toxicity.
STUDY DESIGN: We overexpressed F5-peptide in adult male rats (n=10 rats; with 3 or 4 rats for each of the three different experiments noted in the three regimens) by intratesticular injection of a mammalian expression v

Pure RBC aplasia (PRCS) is a well-recognized complication after allogeneic hematopoietic stem cell transplantation (HSCT). Many therapeutic options are available to treat this condition, including erythropoietin, rituximab, bortezomib, plasma exchange, immunoadsorption, donor lymphocyte infusion, mesenchymal stem cells, antithymocyte globulin, and high-dose steroids; however, treatment outcomes are often variable and can sometimes lead to disappointing results. In this brief article we report our experience with 2 patients with PRCA after major ABO-incompatible HSCT who were resistant to multi

Cardiovascular diseases (CVDs) account for over 17 million deaths globally each year, with atherosclerosis as the underlying cause of most CVDs. Herein we describe the synthesis and in vitro mechanistic evaluation of novel N'-benzylidene-carbohydrazide-1H-pyrazolo[3,4-b]pyridines (3-22) designed as non-anionic antiplatelet agents and presenting a 30-fold increase in potency compared to aspirin. The mechanism underlying their antiplatelet activity was elucidated by eliminating potential targets through a series of in vitro assays including light transmission aggregometry, clot retraction, and q
