The role of AI-powered molecular profiling in the diagnosis and management of cancers of unknown primary: a case report and literature review.

BACKGROUND: Molecular profiling (MP) and next-generation sequencing (NGS) have expanded the diagnostics evaluation of cancer by enabling genomic characterization that informs diagnosis and treatment selection. As the second leading cause of global mortality, cancer claimed nearly 10 million lives in 2020. Artificial intelligence (AI)-powered MP is increasingly recognized for its precision in diagnosing complex cancers, particularly cancers of unknown primary (CUP). CASE PRESENTATION: We report a case of a 69-year-old male with a history of treated prostate cancer, initially diagnosed with CUP

A Polygenic Score for Higher Educational Attainment is Associated with Larger Brains.

People who score higher on intelligence tests tend to have larger brains. Twin studies suggest the same genetic factors influence both brain size and intelligence. This has led to the hypothesis that genetics influence intelligence partly by contributing to the development of larger brains. We tested this hypothesis using four large imaging genetics studies (combined N = 7965) with polygenic scores derived from a genome-wide association study (GWAS) of educational attainment, a correlate of intelligence. We conducted meta-analysis to test associations among participants' genetics, total brain

DNA Polymerase Epsilon Deficiency Causes IMAGe Syndrome with Variable Immunodeficiency.

During genome replication, polymerase epsilon (Pol ε) acts as the major leading-strand DNA polymerase. Here we report the identification of biallelic mutations in POLE, encoding the Pol ε catalytic subunit POLE1, in 15 individuals from 12 families. Phenotypically, these individuals had clinical features closely resembling IMAGe syndrome (intrauterine growth restriction [IUGR], metaphyseal dysplasia, adrenal hypoplasia congenita, and genitourinary anomalies in males), a disorder previously associated with gain-of-function mutations in CDKN1C. POLE1-deficient individuals also exhibited distincti

Recurrent Olfactory Neuroblastoma Treated With Cetuximab and Sunitinib: A Case Report.

Olfactory neuroblastoma (ONB) is a rare cancer originating in the olfactory epithelium of the nasal vault. The recurrence rate of ONB is high, as the standard treatment of surgery followed by radiotherapy and/or chemotherapy is usually unsuccessful. The use of targeted therapy based on individual genomic variations after cancer relapse has not been reported. Here, we present the case of a 44-year-old man who was diagnosed with recurrent ONB and treated with a regimen developed using whole exome sequencing. Potential targets were first identified and then matched to appropriate drugs. Gene muta

Rapid effects of aldosterone on sodium-hydrogen exchange in isolated colonic crypts.

Aldosterone plays a central role in the homeostatic regulation of extracellular fluid volume by stimulating transepithelial electrolyte transport. These effects involve binding to an intracellular receptor, modification of genomic events and protein synthesis. Rapid cellular responses to steroid hormones have been observed in a variety of nonepithelial tissues. The term "nongenomic" has been proposed for these fast steroid responses since they are unaffected by inhibitors of protein synthesis. We hypothesized that colonic crypts, recently demonstrated to absorb fluid, would respond rapidly to

MRI-derived habitat radiomics integrated with transformer deep learning for preoperative prediction of high CD9 expression: a novel prognostic marker in adult-type diffuse gliomas.

BACKGROUND: High CD9 expression is significantly associated with shorter overall survival (OS) in patients with adult-type diffuse gliomas (ADGs). Concurrently, drug screening analyses have identified emetine as a potential targeted therapeutic agent for gliomas exhibiting high CD9 expression, indicating that CD9 may serve as a novel biomarker with the potential to transform glioma diagnosis and treatment. This study aimed to develop an artificial intelligence prediction model based on preoperative magnetic resonance imaging (MRI) images to facilitate the early identification of high CD9 expre

The causal relationship between neuroimaging and Parkinson's disease: A two-sample Mendelian randomization study.

Parkinson's disease (PD) is a neurodegenerative disorder whose etiology and progression remain not fully understood, posing significant challenges for both diagnosis and treatment. This study leverages the 2-sample Mendelian Randomization (TSMR) approach to systematically examine the possible causal connections involving neuroimaging characteristics and the risk of developing PD. Making use of genetic variations from extensive genome-wide association studies (GWAS) as instrumental factors, this analysis focuses on neuroimaging traits related to cortical and subcortical gray matter volumes. The

Bidirectional Mendelian randomization study of causal associations between white-matter structural connectivity and multiple sclerosis.

BACKGROUND: Multiple sclerosis (MS) is associated with widespread network disruption, but whether specific white-matter structural connectivity (WMSC) phenotypes contribute causally to MS susceptibility remains unclear. METHODS: We performed bidirectional two-sample Mendelian randomization (MR) using genome-wide association study (GWAS) summary statistics for 206 tractography-derived WMSC phenotypes (UK Biobank; N = 26,333) and MS susceptibility (IMSGC; N = 115,803 of European ancestry). Primary inference used inverse-variance weighted (IVW) MR under a multiplicative random-effects model, com

Temporal changes in brain morphology related to inflammation and schizophrenia: an omnigenic Mendelian randomization study.

BACKGROUND: Over the past several decades, more research focuses have been made on the inflammation/immune hypothesis of schizophrenia. Building upon synaptic plasticity hypothesis, inflammation may contribute the underlying pathophysiology of schizophrenia. Yet, pinpointing the specific inflammatory agents responsible for schizophrenia remains a complex challenge, mainly due to medication and metabolic status. Multiple lines of evidence point to a wide-spread genetic association across genome underlying the phenotypic variations of schizophrenia. METHOD: We collected the latest genome-wide a

Outcome of partial agenesis of corpus callosum.

BACKGROUND: The diagnosis of corpus callosum anomalies by prenatal ultrasound has improved over the last decade because of improved imaging techniques, scanning skills, and the routine implementation of transvaginal neurosonography. OBJECTIVE: Our aim was to investigate all cases of incomplete agenesis of the corpus callosum and to report the sonographic characteristics, the associated anomalies, and the perinatal outcomes. STUDY DESIGN: We performed a retrospective analysis of cases from January 2007 to December 2017 with corpus callosum anomalies, either referred for a second opinion or de

Breast Cancer Tissue Markers, Genomic Profiling, and Other Prognostic Factors: A Primer for Radiologists.

An understanding of prognostic factors in breast cancer is imperative for guiding patient care. Increased tumor size and more advanced nodal status are established independent prognostic factors of poor outcomes and are incorporated into the American Joint Committee on Cancer (AJCC) TNM (primary tumor, regional lymph node, distant metastasis) staging system. However, other factors including imaging findings, histologic evaluation results, and molecular findings can have a direct effect on a patient's prognosis, including risk of recurrence and relative survival. A High-Throughput, Field-Based Phenotyping Technology for Tall Biomass Crops.

Recent advances in omics technologies have not been accompanied by equally efficient, cost-effective, and accurate phenotyping methods required to dissect the genetic architecture of complex traits. Even though high-throughput phenotyping platforms have been developed for controlled environments, field-based aerial and ground technologies have only been designed and deployed for short-stature crops. Therefore, we developed and tested Phenobot 1.0, an auto-steered and self-propelled field-based high-throughput phenotyping platform for tall dense canopy crops, such as sorghum (Sorghum bicolor).
