Vaccination with radiation-attenuated sporozoites (RAS) can provide highly effective protection against malaria in both humans and mice. To extend understanding of malaria immunity and inform the development of future vaccines, we studied the protective response elicited by this vaccine in C57BL/6 mice. We reveal that successive doses of Plasmodium berghei RAS favour the generation of liver CD8+ tissue-resident memory T cells (TRM cells) over circulating memory cells and markedly enhance their longevity. Importantly, RAS immunisation strongly skews the composition of the liver CD8+ TRM compart

Memory T cells play an important role in mediating long-lasting adaptive immune responses to viral infections, such as SARS-CoV-2. In the context of the latter, much of our current knowledge stems from studies in vaccinated individuals or repeatedly infected individuals. However, limited knowledge is available on these responses in fully naive individuals in German communities. We performed immunophenotyping of a previously naive SARS-CoV-2 cohort in convalescent individuals after asymptomatic to moderate COVID-19. The samples were collected median 250 days post infection during the first wave

EBV contributes to around 2% of all tumors worldwide. Simultaneously, more than 90% of healthy human adults persistently carry EBV without clinical symptoms. In most EBV carriers, it is thought that virus-induced tumorigenesis is prevented by cell-mediated immunity. Specifically, memory CD8+ T cells recognize EBV-infected cells during latent and lytic infection. Using a symptomatic primary infection model, similar to infectious mononucleosis (IM), we found EBV-induced CD8+ tissue resident memory T cells (TRMs) in mice with a humanized immune system. These human TRMs were preferentially establi

Despite the rare appearance of potent HIV-neutralizing mAbs in infected individuals requiring prolonged affinity maturation, little is known regarding this process in the majority of viremic individuals. HIV-infected individuals with chronic HIV viremia have elevated numbers of nonconventional tissue-like memory (TLM) B cells that predominate in blood over conventional resting memory (RM) B cells. Accordingly, we investigated affinity maturation in these 2 memory B cell populations. Analysis of IgG-expressing TLM B cells revealed a higher number of cell divisions compared with RM B cells; howe

Tinidazole and metronidazole have been reported beneficial in the treatment of acute anginose infectious mononucleosis. As this disease is caused by the Epstein-Barr virus (EBV), the effect of tinidazole on EBV infection of human B-lymphocytes was investigated. Tinidazole had no effect on induction of EBV determined nuclear antigen (EBNA) or DNA synthesis, while immunoglobulin synthesis was increased in the presence of the drug. Cytotoxicity directed against EBV positive cell lines and long term T-lymphocyte mediated anti-EBV memory, were not affected by tinidazole. In view of these findings w

Atopic dermatitis is a heterogeneous inflammatory skin disease long-defined by skin barrier dysfunction and type 2 (T helper cell [Th]2) immune dysregulation. Dupilumab established interleukin (IL)-4 and IL-13 as key therapeutic targets, yet partial response rates underscore contributions from Th1, Th17, Th22, and other pathways that vary by age, ethnicity, and disease stage. Cytokine signaling convergence through the Janus kinase/signal transducers and activators of transcription axis underlies the broad efficacy of oral Janus kinase inhibitors, while persistence of tissue-resident memory T c

BACKGROUND: Biological sex can be an important source of variation in infection and immunity and sex-dependent differences in immune response to vaccination have been reported in some studies.
METHODS: We conducted an individual participant data meta-analysis of vaccine trials from one research centre, in which vaccines were administered to children under three years of age and immunological parameters measured. Log-transformed antigen-specific antibody and memory B cell results were meta-analysed and differences between girls and boys reported as geometric mean ratios.
RESULTS: Antibody and

BACKGROUND: Pharyngeal colonisation by the commensal bacterium Neisseria lactamica inhibits colonisation by Neisseria meningitidis and has an inverse epidemiological association with meningococcal disease. The mechanisms that underpin this relationship are unclear, but could involve the induction of cross-reactive immunity. In this study, we aimed to evaluate whether colonisation with N lactamica induces N lactamica-specific B-cell responses that are cross-reactive with N meningitidis.

OBJECTIVE: Ovarian cancer remains the most lethal gynaecological malignancy, and the identification of reliable biomarkers for determining treatment responses and prognosis is needed. This study investigated the clinical significance of helper and cytotoxic effector memory T-cell (TEM) subsets, defined by CD27 and CD28 coexpression, circulation and local compartments.
METHODS: Multi-parametric flow cytometry was used to analyse peripheral blood (n = 30), ascites (n = 14) and tumour tissue (n = 31) from patients with high-grade ovarian cancer (FIGO III-C, n = 31) and age-matched healthy contro

Vaccination with the live vaccine strain (LVS) or natural infection with Francisella tularensis confers long-lasting protection against re-infection, and it is almost exclusively dependent on cell-mediated immunity. The extent, if any, to which vaccine-induced and infection-induced immunity differ in quality and persistence remains unclear. In this study, we compared human immune responses resulting from LVS vaccination and tularemia infection. Peripheral blood mononuclear cells from vaccinees and convalescent patients were recall-stimulated and analyzed for proliferation, characterization of

BACKGROUND: Monoclonal antibodies targeting CD20-expressing B cells constitute effective therapies for people with multiple sclerosis (pwMS) and can be administered intravenously (i.v.) or subcutaneously (s.c.). However, little is known about the differences in immunomodulatory effects of distinct B-cell depletion therapies.
METHODS: We performed detailed characterizations of the immune cell composition in blood samples of pwMS treated with either i.v. [ocrelizumab (Ocre); n = 25] or s.c. [ofatumumab (Ofa); n = 25 or Ocre n = 25] administered anti-CD20 therapy compared to healthy controls (HC

Atopic dermatitis (AD) in infants is associated with increased risk of developing other allergic diseases, including asthma. This progression is known as atopic march (AM). We established a murine AM model by first inducing AD-like skin inflammation to house dust mite (HDM) allergen. Mice were subsequently challenged with HDM delivered to the lung. Our data show that epicutaneous sensitization with HDM increased serum IgE, ear thickness, and immune cell infiltration into the skin, accompanied by an increase in CD4+ tissue-resident memory T cells (Trm) in the lung. Following pulmonary HDM chall

There are several instances of co-investigation and related discoveries and achievements in bovine and human immunology; perhaps most interesting is the development of the BCG vaccine, the tuberculin skin test and the more recent interferon-gamma test that were developed first in cattle to prevent and diagnosis bovine tuberculosis and then applied to humans. There are also a number of immune-physiological traits that ruminant share with humans including the development of their immune systems in utero which increases the utility of cattle as a model for human immunology. These are reviewed her

T cell-based immunotherapies have demonstrated effectiveness in treating diffuse large B cell lymphoma (DLBCL) and follicular lymphoma (FL) but predicting response and understanding resistance remains a challenge. To address this, we developed syngeneic models reflecting the genetics, epigenetics, and immunology of human FL and DLBCL. We show that EZH2 inhibitors reprogram these models to re-express T cell engagement genes and render them highly immunogenic. EZH2 inhibitors do not harm tumor-controlling T cells or CAR-T cells. Instead, they reduce regulatory T cells, promote memory chimeric an

BACKGROUND: Chimeric antigen receptor (CAR) T cell therapy in solid tumors is hampered by dense stromal barriers, rapid functional exhaustion, and limited persistence. Here, we report that low-dose docetaxel acts as an immunomodulatory primer that reprograms CAR T cells to address these barriers. Retrospective analysis of breast cancer patients identifies host lymphocyte reserve as a key determinant of docetaxel treatment benefit, implying an immunological component beyond direct tumor cell killing.
METHODS: Sub-cytotoxic docetaxel concentrations were evaluated in primary human T cells and CA

Trained Immunity is defined as a biological process normally induced by exogenous or endogenous insults that triggers epigenetic and metabolic reprogramming events associated with long-term adaptation of innate immune cells. This trained phenotype confers enhanced responsiveness to subsequent triggers, resulting in an innate immune "memory" effect. Trained Immunity, in the past decade, has revealed important benefits for host defense and homeostasis, but can also induce potentially harmful outcomes associated with chronic inflammatory disorders or autoimmune diseases. Interestingly, evidence s

BACKGROUND: Rheumatoid arthritis (RA) and osteoarthritis (OA) are prevalent chronic joint disorders with immunological pathogenesis. However, the causal relationships between circulating immune cells and them remain largely unknown. Therefore, we conducted a bidirectional two-sample Mendelian randomization (MR) study to determine their causal relationship.
METHODS: Genome-wide association study summary statistics were extracted from publicly available databases regarding immune cell phenotypes, RA, and OA. MR analysis was conducted using five MR methods, with inverse-variance-weighted (IVW) a

An attenuated West Nile virus (WNV), a nonstructural (NS) 4B-P38G mutant, induced higher innate cytokine and T cell responses than the wild-type WNV in mice. Recently, myeloid differentiation factor 88 (MyD88) signaling was shown to be important for initial T cell priming and memory T cell development during WNV NS4B-P38G mutant infection. In this study, two flow cytometry-based methods - an in vitro T cell priming assay and an intracellular cytokine staining (ICS) - were utilized to assess dendritic cells (DCs) and T cell functions. In the T cell priming assay, cell proliferation was analyzed

Glioblastoma is a lethal brain tumor that is resistant to conventional therapies. Here we present a non-lytic replicating retrovirus (RRV) that delivers an IL-15-receptor-linked fusion protein (RLI) superagonist directly into glioblastoma cells, creating localized immunotherapy biofactories. In orthotopic mouse models, RRV-RLI dramatically suppresses tumor growth, prolongs survival, and induces lasting remission with immunologic memory. Mechanistically, we observe increased CD8⁺ T cell and natural killer cell infiltration and activation, alongside elevated antigen presentation pathways. Combin

BACKGROUND: Vγ9Vδ2 T cells constitute the predominant human circulating γδ T cell subset and serve as key mediators of anti-mycobacterial immunity through major histocompatibility complex (MHC)-unrestricted phosphoantigen recognition and rapid effector activation.
SUMMARY: This review evaluates Vγ9Vδ2 T cell biology in the context of tuberculosis (TB). We delineate the three-stage developmental pathway of Vγ9Vδ2 T cells in the postnatal thymus and the butyrophilin 3A1/2A1 (BTN3A1/BTN2A1)-mediated "inside-out" antigen sensing mechanism, noting the limitations of extrapolating in vitro findings
