Cardiac aging increases susceptibility to cardiovascular diseases, particularly myocardial infarction. Although evidence suggest association between activation of the AIM2 inflammasome and cardiovascular diseases, its contribution to age-related cardiac complications remains unknown. We investigated the role of the AIM2 inflammasome and the cardiac response to ischemia-reperfusion (I/R) injury in aged mice. Male wildtype (C57BL/6) and AIM2-deficient (AIM2-/-) mice were subjected to natural aging (21 months), using 3-month-old mice as controls, and human cardiomyocytes (AC16) were stressed by h

Caspase-1 is a central mediator of canonical inflammasome signaling and drives neuroinflammatory injury through interleukin-1β (IL-1β) and interleukin-18 (IL-18) maturation, gasdermin D (GSDMD)-mediated pyroptosis, and secondary inflammatory cascades. In spinal cord injury (SCI) and other neurologic disorders, experimental evidence links caspase-1 activation to blood-spinal cord barrier (BSCB) or blood-brain barrier (BBB) disruption, glial activation, neuronal and oligodendroglial injury, and worse functional outcomes. However, despite mechanistic rationale, the clinical role of caspase-1 as a

Obesity promotes chronic low-grade inflammation through metabolic stress and activation of the NOD-like receptor family pyrin domain-containing 3 (NLRP3) inflammasome. The effects of caloric restriction on this pathway in humans are poorly understood; therefore, we evaluated the impact of a 6-month hypocaloric dietary intervention in 45 adults with obesity by assessing the metabolic, inflammatory, oxidative stress, and inflammasome-related markers in peripheral blood mononuclear cells. Weight loss improved metabolic parameters, reduced oxidative stress, and decreased the expression of nuclear

CONCLUSION: EA may protect peripheral nerve tissue morphology and function by suppressing NLRP3 inflammasomes, thereby alleviating inflammatory responses and potentially attenuating the onset and progression of DPN. This process may be associated with autophagy; but further mechanistic studies are needed to establish causality.

BACKGROUND: Canopy FGF signalling regulator 3 (CNPY3) is an endoplasmic reticulum (ER)-resident chaperone essential for toll-like receptor (TLR) folding, trafficking and inflammasome activation. In rare paediatric cases, biallelic CNPY3 variants have been associated with developmental and epileptic encephalopathy (DEE), but available data remain sparse.
METHODS: 50 individuals from 38 unrelated families with biallelic CNPY3 variants and DEE were evaluated for exome and genome findings, standardised clinical phenotype descriptions, EEG results, and neuroimaging data. A selection of variants un

Osteoarthritis (OA) is a highly heterogeneous whole-joint disease. Current treatment strategies, which primarily focus on symptomatic pain relief, are insufficient to address the therapeutic needs of patients with distinct clinical phenotypes. Accumulating evidence has demonstrated that the empirical use of conventional synthetic disease-modifying antirheumatic drugs (csDMARDs) has shown limited efficacy in OA cohorts selected on the basis of inflammatory features, underscoring the need to more precisely define inflammation-driven OA and elucidate its underlying pathogenic mechanisms. Activation of the NLRP3 inflammasome pathway and dysfunction of brain-derived neurotrophic factor (BDNF) are both implicated in the pathogenesis of Alzheimer's disease (AD). While in vitro and in vivo models support these mechanisms, there is little research investigating their relationship in human brain tissue, and whether this relationship may be abnormal in the AD brain. Temporal cortex tissue taken post-mortem from AD patients and control subjects was analysed for expression of genes for the NLRP3 pathway (NLRP3, PYCARD, CASP1), the potassium channel THIK1 (KCNK13), and related pro-inflam

The NOD-like receptor family pyrin domain-containing 3 (NLRP3) inflammasome has emerged as a critical mediator in cardiovascular diseases, connecting cholesterol crystals and metabolic danger signals to atherogenic inflammation, myocardial injury, and adverse remodeling. Using direct inhibitors that bind to NLRP3 has advantages over indirect strategies. These advantages include retaining the function of other inflammatory body sensors and the potential for oral treatment. Several synthetic compounds have progressed to clinical trials. Recently, NT-0796 demonstrated a significant reduction in C

Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by the loss of dopaminergic neurons in the substantia nigra and pathological α-synuclein aggregation. Growing evidence identifies chronic neuroinflammation-particularly NLRP3 inflammasome activation in microglia-as a central driver for PD onset and progression. Misfolded α-synuclein, mitochondrial dysfunction, and environmental toxins act as endogenous danger signals that prime and activate NLRP3 inflammasome, leading to caspase-1-mediated maturation of IL-1β and IL-18 and subsequent pyroptotic cell death. Canonical inflammasome assembly is driven by interactions between sensors and effector procaspase-1, primarily mediated by the adaptor ASC. Homotypic interactions between Death Domains pyrin (PYD) and caspase recruitment and activation domain (CARD) lead to sensor and ASC oligomerization, along with the recruitment of procaspase-1. ASC self-association is essential for inflammasome activation, which initiates inflammatory responses. Therefore, uncontrolled inflammasome activation contributes to chronic inflammatory diseases. ASC-c, an isoform of ASC, acts as a negative regulator of the inflamm

The NLRP3 inflammasome is a cytoplasmic multiprotein complex that is activated under stress or infection conditions. It promotes the maturation of IL-1β by cleaving pro-Caspase-1, thereby playing a critical role in host innate immune responses. Various viruses can differentially activate NLRP3 through distinct mechanisms, including potassium ion efflux and reactive oxygen species (ROS) accumulation. Mitochondrial DNA (mtDNA) has been demonstrated to be involved in virus-induced inflammasome activation and subsequent inflammatory responses. Red-spotted grouper nervous necrosis virus (RGNNV) is

The activation of the NLRP3 inflammasome is a pivotal step in hyperinflammation in sepsis; however, the regulatory mechanisms underlying its activation are not fully understood. In this study, we found that 14-3-3ε facilitates NLRP3 inflammasome activation by enhancing NLRP3 K63 deubiquitination and promoting its translocation to the mitochondria-associated ER membranes (MAMs) for full activation. Mass spectrometry revealed that 14-3-3ε binds to NLRP3 in macrophages during sepsis. Plasma 14-3-3ε levels were elevated in patients with sepsis and were positively associated with disease severity.

Atherosclerosis (AS) is a chronic inflammatory vascular disease driven by endothelial dysfunction, and dysregulated ubiquitination has emerged as an important pathogenic mechanism. Although deubiquitinating enzymes (DUBs) are key regulators of protein homeostasis, the role of USP40 in AS remains unknown. Here, we defined the function and mechanism of USP40 in cellular and animal models of AS. USP40 expression was reduced in the aortic endothelium of atherosclerotic mice and in ox-LDL-treated human umbilical vein endothelial cells (HUVECs). In Western diet-fed ApoE-/- mice, AAV9-mediated USP40

BACKGROUND: Pro-inflammatory caspase-1 is a key player in innate immunity. Physiological activation of the inflammasome/caspase-1 pathway supports host defense against pathogens, whereas dysregulated activation contributes to the pathogenesis of neonatal diseases such as bronchopulmonary dysplasia. Although preterm infants exhibit reduced IL-1β secretion from monocytes compared with term infants, a comprehensive functional characterization of distinct inflammasome pathways is lacking.
OBJECTIVE: To specifically analyze the activity of NLRP3, AIM2 and NLRC4 inflammasomes in cord blood of pre- a…
