Microinfusion of corticotropin-releasing factor into the nucleus accumbens shell results in increased behavioral arousal and oral motor activity.

Corticotropin-releasing factor (CRF) is a 41 amino acid peptide postulated to be involved in integrating the physiological and behavioral responses to stress. The purpose of this experiment was to determine the effects of CRF microinfused into the nucleus accumbens core (AcbC) and shell (AcbSh) subregions. Rats were tested for general motor activity, cage crossings, and rearing following CRF (0, 125, 250, or 500 ng). Behavioral observations were also made to determine the profile of activity caused by CRF infusion into the Acb. CRF in the AcbSh but not the AcbC regions elicited an increase in

Behavioral activity of 1S,3R-ACPD, an agonist of metabotropic glutamate receptors.

The influence of intracerebroventricular (i.c.v.) injections of 1-aminocyclopentane-1,3-dicarboxylic acid (1S,3R-ACPD), an agonist of metabotropic glutamate receptors, on the activity of the central nervous system was examined in rats. 1S,3R-ACPD at the i.c.v. doses of 100 and 200 nmole significantly decreased apomorphine-induced stereotypy, and at a dose 200 nmole significantly diminished catalepsy caused by haloperidol, two behavioral symptoms controlled mainly by central dopaminergic system. The tested compound had no influence on the locomotor and exploratory activity of rats estimated in

Estradiol benzoate potentiates neuroactive steroids' effects on pain sensitivity.

Progesterone (P), its metabolites, and other neuroactive steroids alter pain thresholds consistent with their efficacies at modulating gamma-aminobutyric acid (GABAA) receptor complexes. We investigated whether estradiol benzoate (EB) potentiates low dosages of neuroactive steroids' effects on pain. Subcutaneous EB (10 micrograms) or sesame oil vehicle was administered to ovariectomized Long-Evans rats (n = 40) 48 h before intracerebroventricular (ICV) infusion of a neuroactive steroid (0.0, 0.1, 0.3, or 0.5 micrograms) in cyclodextrin vehicle. R-alpha-methylhistamine-induced inhibition of gastric acid secretion in pylorus-ligated rats via central histamine H3 receptors.

1. The effect of central H3 histamine receptor activation on gastric acid and pepsin production has been investigated in pylorus-ligated rats. 2. Intracerebroventricular injections (i.c.v.) of the selective H3 agonist, R-alpha-methylhistamine (0.5-50 nmol per rat) caused a dose-dependent inhibition of gastric acid secretion while intravenous administration (5-500 nmol per rat) was completely ineffective. 3. I.c.v. microinjections of mepyramine, tiotidine and thioperamide (51 nmol per rat), selective antagonists at H1-, H2- and H3-sites respectively, failed to modify the acid secretory response

Centrally mediated ocular hypotension: potential role of imidazoline receptors.

These experiments sought to: (1) determine if alpha 2/I1 agonists that are topically active on the eye have similar effects on intraocular pressure when applied to the CNS and (2) ascertain whether these agents lower IOP, in part, via central alpha 2 receptors and/or imidazoline (I1) receptors. New Zealand White rabbits were fitted with chronic indwelling stainless-steel guide cannulas in several brain regions including the lateral ventricle, third ventricle (3V), or medullary intermediate reticular zone. Animals were allowed 5 days' recovery time prior to experiments measuring the effects of

Neuropeptide-Y stimulates pituitary-adrenal activity in fetal and adult sheep.

Corticotrophin-releasing factor (CRF) and arginine vasopressin (AVP) are the primary neuropeptides regulating the secretion of ACTH from the anterior pituitary gland during fetal and adult life. However, a number of other neuropeptides including neuropeptide-Y (NPY) appear to modulate the activity of this system. The potential role of NPY in the regulation of pituitary-adrenal function was examined in fetal and adult sheep. Administration of NPY (6.5 micrograms) as a bolus injection into the third cerebral ventricle of adult ewes elicited a significant (P < 0.05) increase in plasma concentrati

Opioid-induced stimulation of fetal respiratory activity by [D-Ala2]deltorphin I.

[D-Ala2]deltorphin I effects on fetal respiratory activity was characterized to determine the role delta-opioid receptors play in modulating fetal respiratory activity. [D-Ala2]deltorphin I, infused at 0.3 or 100 micrograms/h, intracerebroventricularly (i.c.v.), stimulated fetal respiratory activity without changing blood pH, PCO2 or PO2. Stimulation by 0.3 micrograms/h, but not 100 micrograms/h, was blocked by i.c.v. infusion of the delta-opioid receptor antagonist, naltrindole. Stimulation by 100 micrograms/h was blocked by the mu 1-opioid receptor antagonist naloxonazine. These data suggest

Aminoglucose-induced feeding suppression is regulated by hypothalamic neuronal histamine in rats.

Central mechanisms involved in feeding suppression produced by 1-deoxy-D-glucosamine (1-DGlcN) and 1-deoxy-N-acetylglucosamine (1-DGlcNAc) are unclear. To clarify the mechanisms, we investigated the role of hypothalamic neuronal histamine (HA) in feeding suppression induced by 1-DGlcN and 1-DGlcNAc in rats. Food intake was suppressed for 3 days after a single infusion of 24 mumol 1-DGlcN into the third cerebroventricle (i.c.v.). Depletion of presynaptic HA due to intraperitoneal infusion (i.p.) of alpha-fluoromethylhistidine (FMH), a specific inhibitor of the HA synthesizing enzyme histidine d

Disparate effects of intracisternal RX 77368 and ODT8-SS on gastric acid and serotonin release: role of adrenal catecholamines.

Intracisternal injection of the thyrotropin releasing hormone (TRH) analogue RX 77368 (100 ng) or the somatostatin analogue ODT8-SS (1 microgram) produced an 82% and 101% increase in gastric acid secretion in 2 h pylorus-ligated rats. In contrast, dissimilar effects were produced by intracisternal injection of these peptides on the secretion of serotonin into the gastric lumen. Intracisternal RX 77368 (100 ng) produced a 496% increase in intraluminal serotonin release, while in contrast, intracisternal ODT8-SS (1 microgram) produced a 78% inhibition in intraluminal serotonin release. Indomethacin (i.c.v.) reverses the inhibitory action of peptides on gastric secretion.

Central administration of diverse peptides, such as dermorphin (100 ng), salmon calcitonin (250 ng), human-corticotropin-releasing-factor (10 micrograms) and human-calcitonin-gene-related-peptide (10 micrograms) inhibited gastric acid secretion in pylorus-ligated rats. With the exception of salmon calcitonin this suppressive effect was significantly reversed by the central administration of 400 micrograms indomethacin. These data suggest that the gastric inhibitory effect of dermorphin, human-corticotropin-releasing-factor and human-calcitonin-gene-related-peptide might be mediated by the cent

Interaction of intracerebroventricular insulin and glucose in the regulation of the activity of sympathetic efferent nerves to brown adipose tissue in lean and obese Zucker rats.

The effects of injection of insulin and glucose into the third cerebral ventricle on the firing rate of the sympathetic efferent nerves to intercapsular brown adipose tissue was investigated in anaesthetized lean and obese Zucker rats. Injection of insulin resulted in a dose-dependent (70-480 pmol) inhibition of nerve firing rate, whereas in combination with glucose (140 pmol of insulin and 139 nmol of glucose), insulin strongly potentiated the increase in firing rate seen with glucose alone. Although basal levels of nerve firing rates were lower in the obese rat, responses to insulin, glucose

Role of the hypothalamus in the control of atrial natriuretic peptide release.

Stimulation of the region antero-ventral to the third cerebral ventricle (AV3V) by a cholinergic drug, carbachol, and lesions of the AV3V have been demonstrated in previous studies to either augment or decrease sodium excretion, respectively. Atrial natriuretic peptide (ANP) dramatically increases renal sodium excretion and has been localized to brain areas previously shown to be involved in control of sodium excretion. Consequently, to evaluate a possible role of brain ANP in evoking the changes in renal sodium excretion that follow stimulations or lesions of the AV3V, we determined the effec

Calcitonin and CGRP inhibit gastrointestinal transit via distinct neuronal pathways.

This study compares the central nervous system effects of rat calcitonin and rat calcitonin gene-related peptide (CGRP) on gastrointestinal transit in freely moving rats. Calcitonin and CGRP (0.01-1.0 nmol) injected into the lateral cerebral ventricle significantly inhibited gastric emptying and small bowel transit but did not affect large bowel transit. These biological actions were abolished by ganglionic blockade with chlorisondamine but not by noradrenergic blockade with bretylium. Truncal vagotomy abolished the inhibitory effects of CGRP but not those of calcitonin. Neither adrenalectomy

Low-dose fourth ventricular bombesin selectively suppresses food intake.

Bombesin (BBS) administered into the lateral cerebral ventricles suppresses food intake. Suppression of food intake, however, is accompanied by arousal of competing behaviors such as grooming and increased locomotion, which are atypical of normal satiety. We have found that BBS infused into the fourth cerebral ventricle in rats reduces food intake at doses one-tenth to one-hundredth of those required via the lateral ventricle. In addition, the lowest fourth ventricular dose that significantly reduced food intake did not increase locomotor activity. These data suggest that the hindbrain may be
