Leucine-rich repeat-containing proteins are central to host defense in plants and animals. We show that in the mosquito Anopheles gambiae, two such proteins that antagonize malaria parasite infections, LRIM1 and APL1C, circulate in the hemolymph as a high-molecular-weight complex held together by disulfide bridges. The complex interacts with the complement C3-like protein, TEP1, promoting its cleavage or stabilization and its subsequent localization on the surface of midgut-invading Plasmodium berghei parasites, targeting them for destruction. LRIM1 and APL1C are members of a protein family wi

The human helicase gene SKI2W is located between RD and RP1 in the class III region of the major histocompatibility complex. Transcripts of SKI2W are detectable in RNA samples isolated from multiple tissues. The protein product Ski2w shares striking amino acid sequence similarities to the yeast antiviral protein Ski2p that controls the translation of mRNAs, probably based on the mRNA structural integrity. Whether this translational regulation mechanism for cellular and viral RNAs exists in mammals is under investigation. Antisera against human Ski2w were generated using fusion proteins produce

Chemosensory proteins (CSPs) are small soluble proteins that recognize and bind to various small chemical molecules. Although numerous studies have shown that CSPs play an important role in olfactory communication among insects, recent research indicated that CSPs also contribute to insecticide resistance. The antennae-enriched GmolCSP11 from Grapholita molesta Busck exhibits high binding affinities for seven insecticide ligands, as verified by fluorescence competitive binding assays of recombinant protein. GmolCSP11 exhibited the strongest binding affinity for chlorpyrifos (Ki = 1.01 ± 0.00 μ

In Neurofibromatosis 1 (NF1) germ line loss of function mutations result in reduction of cellular neurofibromin content (NF1+/-, NF1 haploinsufficiency). The Ras-GAP neurofibromin is a very large cytoplasmic protein (2818 AA, 319 kDa) involved in the RAS-MAPK pathway. Aside from regulation of proliferation, it is involved in mechanosensoric of cells. We investigated neurofibromin replacement in cultured human fibroblasts showing reduced amount of neurofibromin. Full length neurofibromin was produced recombinantly in insect cells and purified. Protein transduction into cultured fibroblasts was

Chemosensory proteins (CSPs) are small soluble proteins often associated with chemosensory organs in insects but include members involved in other functions, such as pheromone delivery and development. Although the CSPs of the sensory organs have been extensively studied, little is known on their functions in other parts of the body. A first screening of the available databases has identified 70 sequences encoding CSPs in the oriental locust Locusta migratoria manilensis. Applying proteomic analysis, we have identified 17 of them abundantly expressed in the female reproductive organs, but only

Secreted ferritin is the major iron storage and transport protein in insects. Here, we characterize the message and protein expression profiles of yellow fever mosquito (Aedes aegypti) ferritin heavy chain homologue (HCH) and light chain homologue (LCH) subunits in response to iron and bacterial challenge. In vivo experiments demonstrated tissue-specific regulation of HCH and LCH expression over time post-blood meal (PBM). Transcriptional regulation of HCH and LCH was treatment specific, with differences in regulation for naïve versus mosquitoes challenged with heat-killed bacteria (HKB). Tran

Amuc_1100, an outer membrane protein of the mucin-degrading commensal bacterium Akkermansia muciniphila, is known to strengthen intestinal epithelial barrier integrity, with reports suggesting a potential involvement of Toll-like receptor 2 (TLR2). Its role in the vaginal epithelial barrier, however, remains unexplored. In this study, recombinant Amuc_1100 was expressed in a baculovirus-insect cell system and purified by immobilized metal affinity chromatography (IMAC). Purified Amuc_1100 exhibited concentration-dependent binding to recombinant TLR2 in ELISA. In VK2/E6E7 vaginal epithelial cel

BACKGROUND: Chicken infectious anemia virus (CIAV) causes immunosuppression, aplastic anemia, and lymphoid atrophy in young chicks, causing major economic losses to the poultry industry. Vaccination is the primary strategy for disease control. This study aimed to develop a ferritin-based nanoparticle vaccine displaying CIAV VP1-VP2 proteins and evaluate its protective efficacy in chicks.
RESULTS: Using the SpyTag003/SpyCatcher003 system, we expressed ferritin-SpyCatcher003 (SC-FN) in insect cells and SpyTag003-VP2-VP1Nd129 in E. coli, achieving in vitro assembly of FN-VP2-VP1Nd129 nanoparticl

Dopa decarboxylase (DDC) is a crucial enzyme that regulates melanin synthesis and plays an essential role in cuticular pigmentation and insect development. However, the function of DDC in the fall armyworm, Spodoptera frugiperda, is not yet well understood. In this study, we knocked out the DDC gene and found that mosaic mutants displayed an albino phenotype across all life stages, including larval, pupal, and adult stages. Notably, homozygous DDC mutants showed complete larval albinism, exhibited developmental delays, and frequently failed to pupate, ultimately leading to death. Histological

The global food system is under growing strain to deliver sufficient high-quality protein for a rising population, while curbing environmental degradation, tackling climate change, and maintaining long-term food security. Conventional livestock production is a major source of greenhouse gas emissions, excessive land and water use, and biodiversity loss, which has boosted demand for sustainable alternative proteins such as plant-based, microbial, algal, insect, fermented, and cultivated meat products. This multidisciplinary review comprehensively assesses these emerging protein sources across n

Among the methods tested for termite eradication, termiticides derived from plants are particularly noteworthy. These termiticides represent an effective strategy for insect management, causing minimal environmental harm and posing less risk to human health than widely used chemical controls. The present study aims to obtain crude protein extracts from termites and to characterize their cellulolytic activities for their use in the bioprospecting of natural termiticides. Adult individuals were collected in wood fragments, identified, selected, and sectioned to obtain two protein extracts: one s

Ectoparasitoid wasps inject venom into their hosts during the parasitism to paralyze the hosts and inhibit their immune response and development. The venom of the parasitoid wasp, Habrobracon hebetor, contains proteins that facilitate its parasitization of various lepidopteran pests, and paralyze the 3rd to 5th instar larvae of Spodoptera litura. We comparatively analyzed the function of venom protein neprilysin (NEP) from H. hebetor in two different hosts. NEP is a highly conserved endopeptidase present in various insect tissues. H. hebetor contains 17 NEPs, but only one venom-specific NEP wa

Trypanosoma brucei are protozoan parasites that cause sleeping sickness in humans and nagana in cattle. Inside the mammalian host, a quorum sensing-like mechanism coordinates its differentiation from a slender replicative form into a quiescent stumpy form, limiting growth and activating metabolic pathways that are beneficial to the parasite in the insect host. The post-translational modification of proteins with the Small Ubiquitin-like MOdifier (SUMO) enables dynamic regulation of cellular metabolism. SUMO can be conjugated to its targets as a monomer but can also form oligomeric chains. Here

Cuticular proteins (CPs) are a large family and perform a variety of functions. However, the physiological roles of cuticle protein 21-like (Cpr21L) in the brown planthopper (Nilaparvata lugens, BPH), one of the most destructive insect pests of rice, are largely unclear. In this study, Cpr21L was revealed to be expressed in both BPH nymphs and adults, and the mRNA expression level was much higher in male adults than female adults. Spatially, the expression of Cpr21L in the testis was higher than in the ovary. The RNA interference (RNAi) of Cpr21L seriously decreased nymph survival, and no indi

Insecticides have made great strides in reducing the global burden of vector-borne disease. Nonetheless, serious public health concerns remain because insecticide-resistant vector populations continue to spread globally. To circumvent insecticide resistance, it is essential to understand all contributing mechanisms. Contact-based insecticides are absorbed through the insect cuticle, which is comprised mainly of chitin polysaccharides, cuticular proteins, hydrocarbons, and phenolic biopolymers sclerotin and melanin. Cuticle interface alterations can slow or prevent insecticide penetration in a

Aureolysin, a secreted metallopeptidase (MP) from the thermolysin family, functions as a major virulence factor in Staphylococcus aureus. No specific aureolysin inhibitors have yet been described, making this an important target for the development of novel antimicrobial drugs in times of rampant antibiotic resistance. Although small-molecule inhibitors are currently more common in the clinic, therapeutic proteins and peptides (TPs) are favourable due to their high selectivity, which reduces off-target toxicity and allows dosage tuning. The greater wax moth Galleria mellonella produces a uniqu

Pathogens can manipulate the phenotypic traits of their hosts and vectors, maximizing their own fitness. Among the phenotypic traits that can be modified, manipulating vector behavior represents one of the most fascinating facets. How pathogens infection affects behavioral traits of key insect vectors has been extensively investigated. Major examples include Plasmodium, Leishmania and Trypanosoma spp. manipulating the behavior of mosquitoes, sand flies and kissing bugs, respectively. However, research on how pathogens can modify tick behavior is patchy. This review focuses on current knowledge

Loxosceles intermedia venom comprises a complex mixture of proteins, glycoproteins and low molecular mass peptides that act synergistically to immobilize envenomed prey. Analysis of a venom-gland transcriptome from L. intermedia revealed that knottins, also known as inhibitor cystine knot peptides, are the most abundant class of toxins expressed in this species. Knottin peptides contain a particular arrangement of intramolecular disulphide bonds, and these peptides typically act upon ion channels or receptors in the insect nervous system, triggering paralysis or other lethal effects. Outbreaks of avian influenza A virus infection, particularly the H5N1 strains that have affected birds and some humans for the past 15 years, have highlighted the need for increased surveillance and disease control. Such measures require diagnostic tests to detect and characterize the different subtypes of influenza virus. In the current study, a simple method for producing reference avian influenza virus antisera to be used in diagnostic tests was developed. Antisera of nine avian influenza A virus neuraminidases (NA) used for NA subtyping were produced using a recombinant baculovirus. The re

BACKGROUND: Yersinia pestis is a gram-negative bacterium that causes plague, a disease linked historically to the Black Death in Europe during the Middle Ages and to several outbreaks during the modern era. Metabolism in Y. pestis displays remarkable flexibility and robustness, allowing the bacterium to proliferate in both warm-blooded mammalian hosts and cold-blooded insect vectors such as fleas.
RESULTS: Here we report a genome-scale reconstruction and mathematical model of metabolism for Y. pestis CO92 and supporting experimental growth and metabolite measurements. The model contains 815 g
