Ion Channel Nano-Diagnostics for ER+ Breast Cancer.

Ion channels are pore-forming transmembrane proteins that allow ions to move down an electrochemical gradient and across the channel pore and regulate many cell functions. Among them, are the G-protein-gated inwardly-rectifying K+ channels 1 (GIRK1) that are ubiquitously expressed with major functions in the brain and heart. Interestingly, significantly higher GIRK1 expression has been found in estrogen receptor positive (ER+) breast cancer patients compared to patients with HER2+ tumors or normal patients, and that was statistically correlated with shorter survival times and metastatic potent…

A vacuum-ultraviolet spectropolarimeter for an electron beam ion trap.

We have developed a vacuum-ultraviolet spectropolarimeter for an electron beam ion trap (EBIT) to measure the linear polarization of emission lines from multiply charged ions around the Lyman-α wavelength. The main components for polarimetry are a rotatable MgF2 waveplate and a SiO2/MgF2 multilayer-coated fused silica plate that functions as a reflective polarizer. A grazing-incidence grating is mounted between them to provide wavelength dispersion. The polarization is determined from the intensity modulation of the spectral line as the waveplate is rotated. The performance of the spectropolar…

Zinc Permeation Through Acid-Sensing Ion Channels.

Acid-sensing ion channels (ASICs), activated under acidic conditions, play a critical role in ischemic brain injury, but the detailed mechanisms and signaling pathways remain unclear. Our previous studies have shown that activation of ASIC1a channels contributes to acidosis-induced neuronal injury, partially mediated by increased calcium influx. In this study, we provide evidence that activation of ASIC2a-containing channels induces zinc influx. In cultured mouse cortical neurons, ASIC currents that were insensitive to PcTx1 inhibition were potentiated by extracellular zinc. In Chinese Hamster…

Theory of ion chemical potentials in heterogeneous electrolyte environments.

Electrochemical applications, ranging from energy storage to electrocatalysis and separations, involve ions in heterogeneous environments such as electrode/electrolyte interfaces, material interphases, and confined spaces. These environments influence ion thermodynamics through their effect on chemical potentials and, consequently, on the driving forces relevant to ion transport and electrochemical processes. In addition, features in bulk electrolytes, such as different ion sizes and valence asymmetries, act as intrinsic heterogeneities in an otherwise homogeneous solution. Approaches for mode…

Ion interference and its combined therapies for cancer treatments.

Despite continuous advances in oncology, there remains a pressing need for tumour-specific therapies that can adapt to the unique tumour microenvironment while maintaining high efficacy and low systemic toxicity. Inspired by the critical role of ionic homeostasis in cancer cell survival, ion interference therapy has emerged as a strategy that exploits intracellular ions as cytotoxic effectors to selectively disrupt tumour viability. In particular, nano-engineered platforms enable precise manipulation of ion fluxes through ion-releasing nanomaterials, artificial nanochannels, and transporter-mo…

Legget-Garg inequality for a quantum model of ion channels.

Temporal quantum correlations, revealed by violations of the Leggett-Garg inequalities (LGIs), offer a powerful framework to probe nonclassical dynamics in conceptual models, including those inspired by the living systems. Here, we examine a three-state representation of ionic transport through the selectivity filter, previously presented by Seifi et al. Using this model, we systematically investigate how different scenarios, such as initial state preparation, the specification of measurement basis, hopping dynamics, and environmental fluctuations, affect the emergence of temporal nonclassical…

Collisionless ion-electron energy exchange in magnetized shocks.

Energy partition between ions and electrons in collisionless shocks has been a long-standing unsolved fundamental physical question. We show that kinetic simulations of moderate Alfvénic Mach number, magnetized, collisionless shocks reveal rapid, faster-than-Coulomb, energy exchange between ions and electrons when the plasma is sufficiently magnetized. Using kinetic and multifluid models with counter-streaming ions, we identify resonances between electron whistler and ion magnetohydrodynamic waves that account for this rapid energy exchange.

A DNA-Programmed Potassium Ion Magnetic Resonance Sensor.

Disrupted potassium (K+) homeostasis is an early biochemical hallmark of altered membrane potential, electrolyte imbalance, and malignant transformation, yet in vivo K+ mapping remains unrealized. Optical sensors can report K+ fluctuations but lack the penetration depth required for deep-tissue imaging. Although magnetic resonance imaging (MRI) enables noninvasive, high-resolution visualization of biochemical processes, no contrast agent has achieved K+-dependent modulation of relaxivity, precluding true K+-selective MRI. Here we report a mechanistically defined K+-activated magnetic resonance…

Fluorescence-Based Ion Transport Assays Using Proteoliposomes.

Divalent metal ion transporters are conserved across all domains of life and play essential roles in diverse processes such as manganese acquisition during nutritional immunity in bacteria and iron homeostasis in higher eukaryotes [1-3]. Traditional techniques, such as electrophysiological assays, are often unsuitable due to the slow kinetics of many membrane transporters, electroneutral nature of certain transporter types, and the influence of other proteins with similar activity. To overcome these limitations and to investigate both the activity and ion selectivity of transporters, also incl…

Copper ion-induced MYH9 polymerization executes cuproptosis.

Cuproptosis is a recently identified form of copper-dependent cell death implicated in various physiological and pathological processes. However, the executive proteins involved and the role of physiological intracellular copper in cuproptosis remain unclear. Herein, we identified myosin heavy-chain 9 (MYH9) as a cuproptosis executioner. Copper binding induced MYH9 polymerization, which disrupted the actin cytoskeleton and led to cuproptotic cell death. Hexyl 2-(3,4,5-trihydroxy-phenyl) acetate (HThPA) was shown to induce cuproptosis by binding to the nuclear receptor Nur77, leading to depleti

Secondary-Ion Mass Spectrometry Analysis of Pentosan Polysulfate.

Pentosan polysulfate sodium (PPS) is a glycosaminoglycan mimic that is the only current active pharmaceutical ingredient (API) for the oral treatment of interstitial cystitis. PPS is an inherently complex API due to variations in chain length, backbone structure, sulfation pattern, and end group, making discriminating between batch-to-batch as well as manufacturer-to-manufacturer differences difficult. In this study, high-throughput liquid handling and time-of-flight secondary-ion mass spectrometry (ToF-SIMS) are used to enable the rapid chemical analysis (450 samples in 6 h) of small amounts …

The role of ion channels in the regulation of dendritic cell function.

Ion channels, membrane proteins that facilitate the transport of various inorganic ions across hydrophobic cellular lipid membranes, are ubiquitous in a wide variety of cell and tissue types. They are involved in establishing the cell membrane potential and play a role in various physiological activities by regulating ion concentrations within the cell. Dendritic cells (DCs) are specialised antigen-presenting cells found mainly on the surface of the body (skin and mucous membranes), in the mesenchyme of most organs, in the T-cell compartment of the spleen and in lymph nodes. DCs exert an impor…

Restructuring of Hydrogel Polymer Networks via Ion Storming.

Hydrogel is a 3D network gel with high hydrophilicity, and its mechanical properties are weakened by the disordered polymer network. Although traditional techniques such as directional freezing and salting-out improve the mechanical properties of the hydrogel, the biomedical and chemical engineering applications are limited by the complex processing procedures. In view of this situation, an urgent demand for the non-intrusive in situ hydrogel processing technique is required, and the disordered polymer network resembles a tangled yarn that can be unraveled through the external electric field. …

Ion concentrations in CSF and serum are differentially and precisely regulated.

Fluctuations of extracellular brain ion concentrations have been associated with transitions between brain states such as sleep and wakefulness, and disturbances have been implicated in a variety of neurological conditions, including dementia, epilepsy and migraine. This study aims to define the normal CSF ion profile and identify key factors influencing its regulation. In this cross-sectional study, we analysed samples from 42 individuals (16 men), including 28 healthy participants and 14 patients with medically unexplained neurological symptoms. Age spanned between 20 and 55 years. Using val…

A Quick Reference on Anion Gap and Strong Ion Gap.

Metabolic acid-base disorders frequently occur in emergency and critically ill patients. Recognizing these disturbances can be challenging, particularly when multiple acid-base imbalances coexist within a single patient. The anion gap (AG) and strong ion gap (SIG) are valuable calculations that help identify complex metabolic acid-base disturbances associated with unmeasured anions. This review provides definitions, reference values, indications, limitations, and guidelines for interpreting AG and SIG changes in clinical settings.

Crystal structures of three salts of the tri-phenylsulfonium ion.

The reactions of tri-phenyl-sulfonium chloride ([TPS][Cl]) with various acids in methanol yield the corresponding salts tri-phenyl-sulfonium triiodide, C18H15S+·I3 - or [TPS][I3] (I), tri-phenyl-sulfonium perchlorate, C18H15S+·ClO4 - or [TPS][ClO4] (II), and tri-phenyl-sulfonium hexa-fluoro-phosphate, C18H15S+·PF6 - or [TPS][PF6] (III), as crystalline products. These crystals were structurally characterized by single-crystal X-ray diffraction. In all three compounds, the sulfur atom in the tri-phenyl-sulfonium cation adopts a distorted trigonal-pyramidal geometry. [TPS][I3] (I) and [TPS][PF6] …

Calcium ion nanomodulators induce pyroptosis for tumor immunotherapy.

Pyroptosis is a promising strategy for tumor immunotherapy, but nanomaterial-based pyroptosis inducers remain underexplored. Here, we developed a pH-responsive calcium nanomodulator by loading curcumin onto ultrathin Ca/Al layered double hydroxide nanosheets (LDH-CUR). LDH-CUR induces pyroptosis via mitochondrial calcium overload. Upon internalization by tumor cells, it degrades in acidic environments, releasing calcium ions while curcumin blocks calcium efflux, leading to calcium surges. This triggers reactive oxygen species production, cytochrome C release, and caspase-3 activation. Caspase-…
