Hepatic and duodenal drug metabolism in the rat during fat deficiency.

1. The effect of dietary fat deficiency on the composition of hepatic microsomes and postmitochondrial supernatant fraction of duodenal mucosa and on the activities of drug-metabolizing enzymes was studied in the rat. 2. There were only little changes in the measured protein, cholesterol and phospholipid fractions in the liver microsomes. In the postmitochondrial supernatant fraction of the duodenal mucosa, however, protein content was increased during fat deficiency. 3. In the fatty acid composition of hepatic microsomal phospholipids marked differences took place during fat deficiency. The a

Reduction in bovine milk fat globule size is characterized by remodeling of membrane-associated lipids in milk.

Milk fat globule (MFG) size is closely linked to milk lipid interfacial organization, but its association with membrane-associated lipid remodeling remains unclear. Here, milk composition, MFG size distribution, fatty acid profiles, and whole-milk lipidomics were analyzed in conjugated linoleic acid (CLA)-supplemented cows. CLA supplementation reduced milk fat content and shifted MFGs toward smaller sizes. Whole-milk lipidomics showed that the small-MFG phenotype was accompanied by remodeling of membrane-associated lipid classes, particularly sphingolipids. Among major sphingolipids, hexosylce

The UHPLC-Orbitrap MS/MS and Network Pharmacology Strategies Reveal the Active Antioxidants of Bleeding Sap from Sponge Gourd in Treating Tuberculosis.

The bleeding sap of Luffa cylindrica (L.) Roem has been used for the treatment of tuberculosis since the record of Supplements to Compendium of Materia Medica. The active components and possible mechanism of it are yet ambiguous. Hence, this study is focused on investigating the possible mechanism underpinning this effect on the perspective of the antioxidant ingredients from the bleeding sap. Through organic solvents extraction, HPLC fractionation, DPPH trials evaluation, and UHPLC-Orbitrap tandem MS identification, a total of 37 compounds were identified from the bleeding sap with the strong

Integrative transcriptomics and metabolomics reveal Cry1-associated hepatic alterations and plasma essential fatty acids as biomarkers of residual circadian disruption post-sepsis.

BACKGROUND: Sepsis disrupts circadian rhythm, leaving lasting molecular traces and circadian insensitivity to jet lag even after acute phase recovery. However, the core mechanism and biomarkers of residual circadian dysfunction post-sepsis remain unclear, which hinders targeted interventions. METHODS: Male mice were subjected to non-lethal LPS injection, and after 7 days of recovery, liver transcriptomics and plasma metabolomics were integrated. Behavioral, physiological, and molecular analyses were performed, with statistical tests including DESeq2, Student's t-test, MetaCycle, and ROC curve

Instant controlled pressure drop processing modulates phenolic composition and fatty acid profile in Amaranthus hypochondriacus grain.

Instant Controlled Pressure Drop (DIC) is a high-pressure/short-time thermo-mechanical pretreatment that can expand grain structure and impr … re the predominant compounds across treatments. GC-MS showed oils dominated by palmitic, oleic, and linoleic acids and squalene, with balanced saturated and unsaturated fractions. In both varieties, pressure-time interactions indicated that mild-to-moderate treatments were optimal, maximizing phenolic recovery without markedly altering the fatty-acid signature.

Omics insights into the mechanisms of flavor changes in Eriocheir sinensis under different restraint methods during live transportation.

Eriocheir sinensis frequently experiences stress from inappropriate restraint during live transportation, impairing animal welfare and edible quality. This study employed flavoromics and metabolomics to investigate the mechanisms linking physiological metabolism with edible quality changes in deteriorated tissues of E. sinensis under different restraint methods. The hepatopancreas was identified as the key tissue of deterioration. The degree of deterioration ranked as follows: net-bag binding (NB) group > plastic-box binding (PB) group > cotton-rope binding (RB) group. Oxidative stress reduced

Fatty Acid and Micronutrient Profile of Longissimus Lumborum from Red Angus and Red Angus x Akaushi Cattle Finished on Grass or Grain.

Cattle diet and breed modify the nutritional profile of beef. The objective of this study was to compare the fatty acid (FA) and micronutrient profiles of Red Angus (RA) and RA x Akaushi (AK) crossbreed steers fed either a grass or grain diet. This two-year study randomly assigned steers to the diets using a 2 × 2 factorial experiment. FAs and micronutrients were analyzed. Diet effect was the strongest with grass-finished beef being higher in n-3 polyunsaturated FAs (p < 0.001), conjugated linoleic acid (p < 0.05), vaccenic acid (p < 0.05), iron (p < 0.001), and vitamin E (p < 0.001) compared

NAFLD causes selective CD4(+) T lymphocyte loss and promotes hepatocarcinogenesis.

Hepatocellular carcinoma (HCC) is the second most common cause of cancer-related death. Non-alcoholic fatty liver disease (NAFLD) affects a large proportion of the US population and is considered to be a metabolic predisposition to liver cancer. However, the role of adaptive immune responses in NAFLD-promoted HCC is largely unknown. Here we show, in mouse models and human samples, that dysregulation of lipid metabolism in NAFLD causes a selective loss of intrahepatic CD4(+) but not CD8(+) T lymphocytes, leading to accelerated hepatocarcinogenesis. We also demonstrate that CD4(+) T lymphocytes

Newborns with lower levels of circulating polyunsaturated fatty acids (PUFA) are abdominally more adipose.

BACKGROUND: Maternal nutrition is the main source of Poly-Unsaturated Fatty Acids (PUFA) for the fetus. PUFA may influence the accumulation of fat in early life. OBJECTIVES & METHODS: In 33 breastfed infants born appropriate-for-gestational-age, we studied whether body composition (judged by absorptiometry at 2 wk and 4 mo) relates to PUFA levels (assessed by gas chromatography) in the maternal or fetal circulation at birth. RESULTS: Abdominal fat at 2 wk associated negatively to umbilical-cord levels of separate PUFA (linoleic, arachidonic, eicosapentanoic and docosahexaenoic acid; all P be

Isolation of sterols from the marine fungus Corollospora lacera.

Several marine fungi collected from the waters of Prince Edward Island, Canada, were screened for the presence of natural products exhibiting antibacterial activity. Both broths and mycelia of these fungi were studied using the bioassay-guided chromatographic separation. The 4 fractions from the extract of mycelia of Corollospora lacera exhibited weak antibacterial activity and were analyzed further. From these fractions, 2 sterols (5 alpha,8 alpha-epidioxyergosterol and 22E,24R-ergosta-7,22-diene-3beta,5 alpha,6 beta-triol) and a 3:1 mixture of linoleic and oleic acids were isolated. The pres

In vitro studies of the phototoxic potential of the antidepressant drugs amitriptyline and imipramine.

Amitriptyline and imipramine, two tricyclic antidepressant drugs, have been studied to evaluate their phototoxic potential using various models. Reactive oxygen species production was investigated. A negligible production of singlet oxygen was observed for both compounds whereas a significant production of superoxide anion was noted for amitriptyline in particular. Moderate red blood cell lysis under UVA light (365 nm) was induced in the presence of the two drugs at a concentration of 50 microM. Cellular phototoxicity was investigated on a murine fibroblast cell line (3T3). The two drugs were

Magnesium deprivation or short-term essential fatty acid deficiency in rats: effects on serum lipids, platelet fatty acid composition and arachidonic acid incorporation into platelet phospholipids.

The aim of this study was to evaluate the effect of short-term magnesium or essential fatty acid (EFA) deficiencies on plasma lipids, platelet fatty acid composition and [1-14C] arachidonic acid incorporation into platelet phospholipids. Weanling rats were fed purified diets (casein 20%, sucrose 70.5%, lipid 5%) for two weeks. The control and magnesium-deficient diets included corn oil as lipid source. The EFA-deficient diet included hydrogenated coconut oil. The fatty acid composition of serum lipids confirmed the linoleic acid deprivation in the EFA-deficient group. Significant changes in pl

DHA and EPA reverse cystic fibrosis-related FA abnormalities by suppressing FA desaturase expression and activity.

Patients and models of cystic fibrosis (CF) exhibit consistent abnormalities of polyunsaturated fatty acid composition, including decreased linoleate (LA) and docosahexaenoate (DHA) and variably increased arachidonate (AA), related in part to increased expression and activity of fatty acid desaturases. These abnormalities and the consequent CF-related pathologic manifestations can be reversed in CF mouse models by dietary supplementation with DHA. However, the mechanism is unknown. This study investigates this mechanism by measuring the effect of exogenous DHA and eicosapentaenoate (EPA) suppl

Characteristics of lipid substances activating the ileal brake in the rat.

Studies were carried out in 36 adult male rats to determine the characteristics of lipid substances which, after infusion into the ileum, slow the stomach to caecum transit time of the head of a bean meal in the rat. Stomach to caecum transit time was measured by environmental hydrogen analysis. Ileal infusion of a range of free fatty acids including petroselinic, oleic, myristoleic, erucic, linoleic, and linolenic all significantly slowed stomach to caecum transit time, as did the detergents (sodium bis (2-ethyl hexyl) sulphosuccinate and sodium linoleyl sulphate), the triglyceride corn oil,

Unveiling the Mechanism of Action of Palmitic Acid, a Human Topoisomerase 1B Inhibitor from the Antarctic Sponge Artemisina plumosa.

Cancer remains a leading cause of death worldwide, highlighting the urgent need for novel and more effective treatments. Natural products, with their structural diversity, represent a valuable source for the discovery of anticancer compounds. In this study, we screened 750 Antarctic extracts to identify potential inhibitors of human topoisomerase 1 (hTOP1), a key enzyme in DNA replication and repair, and a target of cancer therapies. Bioassay-guided fractionation led to the identification of palmitic acid (PA) as the active compound from the Antarctic sponge Artemisina plumosa, selectively inh

Investigating the association between polyunsaturated fatty acids and osteomyelitis by Mendelian randomization.

Osteomyelitis, characterized by bone inflammation and infection, poses a significant global health burden. This Mendelian randomization (MR) study investigates the causal relationship between polyunsaturated fatty acids (PUFAs) and osteomyelitis risk. By using GWAS data from 114,999 individuals, we explore specific PUFAs and their genetic variations using Inverse variance weighted (IVW), MR-Egger and weighted median methods. The results reveal a suggestive association between genetically predicted higher docosahexaenoic acid (DHA) and omega-6 levels with increased osteomyelitis risk. Conversel

Effect of vegetable oils on the experimental infection of mice with Trypanosoma congolense.

Vegetable oils are frequently used as solvents for lipophilic materials; accordingly, the effects of their components should be considered in animal experiments. In this study, the effects of various vegetable oils on the course of Trypanosoma congolense infection were examined in mice. C57BL/6J mice were orally administered four kinds of oils (i.e., coconut oil, olive oil, high oleic safflower oil, and high linoleic safflower oil) with different fatty acid compositions and infected with T. congolense IL-3000. Curcumin improves alcoholic fatty liver by inhibiting fatty acid biosynthesis.

Alcoholic fatty liver is a threat to human health. It has been long known that abstinence from alcohol is the most effective therapy, other effective therapies are not available for the treatment in humans. Curcumin has a great potential for anti-oxidation and anti-inflammation, but the effect on metabolic reconstruction remains little known. Here we performed metabolomic analysis by gas chromatography/mass spectrometry and explored ethanol pathogenic insight as well as curcumin action pattern. We identified seventy-one metabolites in mouse liver. Carbohydrates and lipids were characteristic c
