Sequence and time-dependent deamination of cytosine bases in UVB-induced cyclobutane pyrimidine dimers in vivo.

The mutational specificity of UV-light is characterized by an abundance of C to T transition mutations at dipyrimidines containing cytosine or 5-methylcytosine. A significant percentage of these mutations are CC to TT double transitions. Of the major types of UV-induced DNA lesions, the cis-syn cyclobutane pyrimidine dimers (CPDs) are thought to be the most mutagenic lesions, at least in mammalian cells. It has been proposed that the CPDs become mutagenic perhaps only after cytosine bases within these dimers deaminate to uracil and the resulting U-containing photolesions are correctly bypassed

Prenatal Exposure to Chemotherapy Increases the Mutation Burden in Human Neonatal Hematopoietic Stem Cells.

This study demonstrates that environmental mutagenic exposure during pregnancy can increase somatic mutation accumulation in the fetus. Given that detrimental early life exposures can adversely affect health outcomes later in life, our study highlights the need for further research into the impact of environmentally induced genomic insults during the perinatal period. See related commentary by Furudate and Takahashi, p. 870.

Transposon mutagenesis of group B streptococcus beta-hemolysin biosynthesis.

Beta-hemolysin production by group B streptococci (GBS) is speculated to be a major virulence factor of the organism. A virulent, beta-hemolytic group B streptococcus strain was mutagenized with the self-conjugative transposon Tn916 to derive isogenic strains with mutations only in the gene(s) responsible for beta-hemolysin biosynthesis. There was no significant difference between the virulence of the parent strain and that of the mutant strains in a neonatal rat sepsis model.

[Chromosome instability syndromes].

Chromosome instability syndromes are defined by either an increase of chromosomal breakage or by an increase of sister chromatid exchange number, or by an increase of the two. Bloom's syndrome, Ataxia telangiectasia, Fanconi's Anemia are the main components of this group. The incidence of cancers or malignant blood diseases is high. The finding of DNA repair abnormalities in some of them and their high sensitivity to particular mutagenic agents makes those syndromes an interesting model for oncogenesis.

Fragilitas ossium: a new autosomal recessive mutation in the mouse.

Fragilitas ossium, fro, is an often lethal recessive mutation that was discovered in a randombred stock of mice after treatment with the chemical mutagen tris(1-aziridinyl) phosphine-sulphine. The fro/fro mice have osteoporosis-bending and fracture of the long bones-that is associated with apparently normal cartilage growth. The roentgenological and pathological features are similar to those of the severe, often lethal recessive form of human osteogenesis imperfecta.

Toxicology of daunorubicin in animals and man.

The toxic effects of daunorubicin with specific reference to acute, subacute, and long-term studies in animals are reviewed, and its clinical toxicity is considered. We emphasize the importance of hematologic toxicity and infections when treating with daunorubicin. A summary of the work done on detoxifying activities of certain compounds is presented, and the activities that increase the toxic effects of daunorubicin are discussed. Finally, the immunosuppressive, mutagenic, teratogenic, and carcinogenic activities of daunorubicin are mentioned.

POLYPHARMACY AND CANCER: А NEW VISION FOR SKIN CANCER PATHOGENESIS-PHOTOTOXICITY AND PHOTOCARCINOGENICITY DUE TO NITROSAMINE CONTAMINATION DURING TELMISARTAN/ TAMSULOSIN INTAKE.

The toxicokinetics of nitrosamines remain a mystery to this day, though it appears that the role of nitrosamines in potentiating the generation of mutations required for the onset of skin cancer continues to be a significant concern. Nitrosamines are mutagens, genotoxic substances, and mediators of phototoxicity/carcinogenicity, whose long-term daily usage, in the context of polypharmacy, can result in the parallel appearance of heterogeneous forms of skin cancer: keratinocytic and melanocytic. But a number of clinical observations suggest that it is the nitrosamines that potentiate the multip…

Toxicological effects of prolonged and intense use of mosquito coil emission in rats and its implications on malaria control.

Mosquito coil is a vector control option used to prevent malaria in low income counties, while some studies have addressed this issue, additional reseach is required to increase knowledge on the adverse health effects caused by the prolonged use of coils. In this study we investigated the toxicological effects of fumes from two locally manufactured mosquito coil insecticides (with pyrethroids: transfluthrin and d-allethrin as active ingredients) on male albino rats. For this, we recorded the haematological and biochemical indices, and made histopathology and mutagenicity evaluations in rats ex

Antibiotic-Induced Genotoxicity: Molecular Mechanisms, Cytogenetic Damage, and Implications for Human Health.

Global antibiotic consumption continues to rise across pediatric and adult populations, while the genotoxic consequences of host eukaryotic exposure remain less systematically characterized than the parallel problem of antimicrobial resistance. Several lines of evidence, from molecular cytogenetics, redox biology, and systems toxicology, now permit a more mechanistically resolved synthesis of antibiotic-induced genome stress than was previously possible, although a substantial fraction of this evidence is preclinical and warrants cautious clinical extrapolation. This narrative review evaluates

Evaluation of herbicides action on plant bioindicators by genetic biomarkers: a review.

The use of pesticides has increased worldwide, owing to the demand for products of good quality and to satisfy a growing population. Herbicides represent almost half of the total amount of pesticides used. Although important to the reduction of costs and an increase of productivity, their indiscriminate use, as well as that of the other pesticides, is a global environmental problem, since they affect the living organisms. To evaluate the damage caused by herbicides to the environment, different organisms have been used as bioindicators, especially higher plants, due to several advantages. This

The antimicrobial activity, toxicity and antimicrobial mechanism of a new type of tris(alkylphenyl)sulfonium.

The antimicrobial activity, toxicity and antimicrobial mechanism of a new type of tris(4-alkylphenyl)sulfonium which has sterically bulky alkyl substituents (bTAPS), were estimated and compared with those of other sulfoniums which we reported previously. Concerning tris {4-(iso-propyl)phenyl}sulfonium (bTAPS-iso3) and tris{4-(tert-butyl)phenyl}sulfonium (bTAPS-tert4), the antimicrobial activity of these compounds tended to be lower than both tri(n-alkyl)sulfoniums (TASs) and tris{4-(n-alkylphenyl)}sulfoniums (TAPSs) at similar ClogP values. However, the activities of tris{4-(cyclohexyl)phenyl}

Mechanisms of arsenic carcinogenicity: genetic or epigenetic mechanisms?

Environmental and occupational exposure to arsenic is associated with increased risk of skin, urinary bladder, and respiratory tract cancers. The mechanisms responsible for arsenic carcinogenesis have not been established. Arsenic does not act through classic genotoxic and mutagenic mechanisms, as do other metals such as cadmium or chromium. Increasing evidence indicates that arsenic acts at the level of tumor promotion by modulating the signaling pathways responsible for cell growth.

[Chromosome aberrations in a lymphocyte culture from persons in contact with chromium].

A detailed analysis was carried out of chromosome aberrations induced by chromium compounds in a lymphocyte culture of persons which were in contact with the compounds. It is shown that the frequency of cells with chromosome aberrations depends on the age and duration of contact with chromium and does not depend on the sex. It was marked that hexavalent chromium compounds have a higher cytogenetic activity. Mechanisms of mutagenic action of heavy metal ions are discussed (taking chromium as an example).

The Discovery of Novel Antimalarial Aminoxadiazoles as a Promising Nonendoperoxide Scaffold.

Since the appearance of resistance to the current front-line antimalarial treatments, ACTs (artemisinin combination therapies), the discovery of novel chemical entities to treat the disease is recognized as a major global health priority. From the GSK antimalarial set, we identified an aminoxadiazole with an antiparasitic profile comparable with artemisinin (1), with no cross-resistance in a resistant strains panel and a potential new mode of action. A medicinal chemistry program allowed delivery of compounds such as 19 with high solubility in aqueous media, an acceptable toxicological profile

A population study using the human erythrocyte PIG-A assay.

Erythrocyte-based PIG-A assay is sensitive and reliable in detecting exposure to mutagenetic agents in animal studies, but there are few data from human populations. In this study, we employed a method for detecting CD59 phenotypic variants, resulting from mutation in the PIG-A gene, in human red blood cells (RBCs), and determined the CD59-deficient RBC (RBCCD59- ) frequencies in 217 subjects from general population. The majority of subjects had a relatively low mutant frequencies (MFs) (average, 5.25 ± 3.6 × 10-6 , median, 4.38 × 10-6 , for all subjects), but with males having a significantly

Genotoxicity profile of fexinidazole--a drug candidate in clinical development for human African trypanomiasis (sleeping sickness).

The parasitic disease human African trypanomiasis (HAT), also known as sleeping sickness, is a highly neglected fatal condition endemic in sub-Saharan Africa, which is poorly treated with medicines that are toxic, no longer effective or very difficult to administer. New, safe, effective and easy-to-use treatments are urgently needed. Many nitroimidazoles possess antibacterial and antiprotozoal activity and examples such as tinidazole are used to treat trichomoniasis and guardiasis, but concerns about toxicity including genotoxicity limit their usefulness. Fexinidazole, a 2-substituted 5-nitroi

Effects of toxic chemicals on the reproductive system. Council on Scientific Affairs.

In an effort to make physicians more aware of the hazards of the workplace to pregnant workers, the Council on Scientific Affairs' Advisory Panel on Reproductive Hazards in the Workplace prepared this third and final report reviewing the effects of chemical exposure. A total of 120 chemicals were considered for reviews based on an estimation of their imminent hazard, ie, widespread use and/or inherent toxicity. Following a brief introduction, which sets out general principles, clinical applications, and aids to the recognition of a human teratogen, the report presents reviews and opinions for
