Rapid Forming, Robust Adhesive Fungal-Sourced Chitosan Hydrogels Loaded with Deferoxamine for Sutureless Short-Gap Peripheral Nerve Repair.

Clinically, conventional sutures for repair of short-distance nerve injuries (< 5 mm) may contribute to uncontrolled inflammation and scar formation, thus negatively impacting nerve regeneration. To repair transected peripheral nerves with short distances, a rapid-forming, robust adhesive chitosan hydrogel is prepared by synthesizing maleic and dopamine bi-functionalized fungal-sourced chitosan (DM) and subsequently photopolymerizing DM precursor solution. The hydrogel rapidly polymerized under UV light irradiation (≈2 s) and possessed a strong adhesive strength (273.33 ± 55.07 kPa), facilitat

3D-engineering of Cellularized Conduits for Peripheral Nerve Regeneration.

Tissue engineered conduits have great promise for bridging peripheral nerve defects by providing physical guiding and biological cues. A flexible method for integrating support cells into a conduit with desired architectures is wanted. Here, a 3D-printing technology is adopted to prepare a bio-conduit with designer structures for peripheral nerve regeneration. This bio-conduit is consisted of a cryopolymerized gelatin methacryloyl (cryoGelMA) gel cellularized with adipose-derived stem cells (ASCs). By modeling using 3D-printed "lock and key" moulds, the cryoGelMA gel is structured into conduit

Transplantation into the central nervous system: a therapy of the future.

Grafts in the central nervous system probably work by a number of mechanisms. Our understanding of these mechanisms and how they might be employed to treat neurological disease is still elementary. As the sophistication in the functioning of the grafts increases, so does the sophistication required in the preparation of the tissues for grafting. Similarly, new techniques will be required to place the grafts in a manner advantageous to both graft survival and function and that carries minimal risk to the host. There is still much basic science investigation that must be performed, but basic sci

Recurrent carpal tunnel syndrome.

Persistent or recurrent symptoms following carpal tunnel release surgery are an infrequent but challenging clinical problem. A thorough evaluation of these patients is mandatory and must confirm the accuracy of the original diagnosis and rule out the presence of concurrent conditions or disorders that may cause persistent symptoms that mimic carpal tunnel syndrome. If an alternative explanation of the patients' symptoms cannot be identified, and if conservative care is ineffective, then surgical treatment should be considered. Adequate exposure of the median nerve and carpal tunnel are mandato

Demonstration of therapeutic effect of plasma-synthesized polypyrrole/iodine biopolymer in rhesus monkey with complete spinal cord section.

Spinal cord injury (SCI) can cause paralysis, and although multiple therapeutic proposals have been developed in murine models, results have hardly been replicated in humans. As non-human primates (NHP) are more similar to humans than rodents, the current study investigated whether it was possible to reproduce in a NHP, the previously obtained beneficial results by using a plasma-synthesized polypyrrole/iodine (PPy/I) biopolymer, which reduce glial scar formation and inflammatory response and promotes nerve tissue preservation, regenerative processes and functional recovery in rats. In NHPs (R…

[Effects of wheat-grain moxibustion on autophagy and endoplasmic reticulum stress in rats with cervical spondylotic radiculopathy].

OBJECTIVE: To observe the effects of wheat-grain moxibustion on autophagy and endoplasmic reticulum stress (ERS) in the spinal cord and nerve root tissues of rats with cervical spondylotic radiculopathy (CSR), and to explore the potential mechanisms by which wheat-grain moxibustion alleviates neuropathic pain in CSR. METHODS: Forty-eight SPF-grade SD rats were randomly divided into a sham operation group, a model group, a wheat-grain moxibustion group, and a wheat-grain moxibustion + 3-methyladenine (3MA) group, with 12 rats in each group. The CSR model was established in the model group, the

Neuro-Instructive Hydrogels - A Key to Sensory Innervation in Oral Tissues: A Comprehensive Review.

Current treatments for oral tissue defects restore structure but rarely restore sensory nerves. Tissues without innervation lose protective reflexes and long-term stability. This review examines neuro-instructive hydrogels designed to guide nerve regeneration in oral tissues. We discuss four design mechanisms: physical cues that direct axon growth, spatiotemporal release of neurotrophic signals, regulation of oral stem cells and Schwann cells, and immunomodulation that supports a pro-regenerative niche. We then map these mechanisms onto three clinical scenarios: regeneration of the dentin-pulp

Preparation of decellularized optic nerve grafts.

BACKGROUND: Decellularized tissues based on well-conserved extracellular matrices (ECMs) are a common area of research in tissue engineering. Although several decellularization protocols have been suggested for several types of tissues, studies on the optic nerve have been limited. METHODS: We report decellularization protocol with different detergent for the preparation of acellular optic nerve and tissues were examined. DNA, glycosaminoglycan (GAG), and collagen content of the groups were evaluated with biochemical analyses and examined with histological staining. Mechanical properties, che

Oxygen-releasing scaffolds in tissue engineering: design strategies, fabrication and regenerative applications.

Oxygen is indispensable for sustaining tissue and cell physiological functions, yet a hypoxic microenvironment often arises in tissue injury and regeneration, impairing graft scaffold performance and hindering repair processes. Thus, oxygen-releasing scaffolds have become a pivotal research frontier in tissue engineering. Via elaborate spatial constructions (e.g. electrospun fibers, hydrogels, microspheres) and diverse modification strategies, these scaffolds enable sustained, spatiotemporally controllable oxygen release at injury sites, alleviating local hypoxia and boosting tissue regenerati

Obturator neuropathy after laparoscopic pelvic lymphadenectomy.

A case of obturator nerve injury with delayed manifestation following laparoscopic pelvic lymphadenectomy is presented. While the incidence of obturator nerve injury does not appear to differ between open surgical and laparoscopic approaches, the laparoscopic approach demands unequivocal visualization of the nerve when utilizing electrocautery because of the risk of nerve injury from thermal conduction through apposed tissues.

[Protective effect of Sini Decoction in attenuating cryopreservation-induced injury of rats' sciatic nerves based on apoptosis and oxidative stress].

Cryopreservation is the primary technique for in vitro preservation of allogeneic tissue. However, its success is often hindered by factors such as low temperature, ischemia, and hypoxia. This study investigated the potential of Sini Decoction, known for its antioxidant and anti-apoptotic properties, to reduce cryopreservation-induced injury in rats' sciatic nerves. Sini Decoction was prepared according to the Chinese Pharmacopoeia, and its cytotoxicity on Rsc96 cells was assessed by using the CCK-8 method. Sini Decoction at concentrations of 4, 8, and 16 mg·mL~(-1), termed as low-(SL), medium

A throwback on 25 years in pannexin research.

Cellular channels consisting of pannexins facilitate signaling between the intracellular and extracellular environment. In this respect, pannexin channel opening serves as a pivotal trigger for both inflammatory processes and cell death. While pannexin channels also contribute to normal physiological functions, their roles in pathological mechanisms have been more extensively studied. Indeed, aberrant pannexin channel activity contributes to the onset and progression of a plethora of diseases. Not surprisingly, pannexin channels have attracted considerable attention as a biomarker and therapeu

The amyloidogenic C-terminal region of TMEM106B modulates lipid membrane biophysical properties: Functional and pathological insights.

The lysosomal transmembrane protein 106B (TMEM106B) forms amyloid filaments in the human brain in an age-dependent manner, observed both in neurologically healthy individuals and in patients with neurodegenerative diseases also containing tau, α-synuclein, or TDP-43 inclusions. Despite its pathological and physiological relevance, the biochemical mechanisms governing TMEM106B structural stability and its functional interactions with membranes remain largely unknown. Here, we examined the luminal C-terminal fragment of TMEM106B (called TST, residues 120-254), corresponding to the amyloid fibril

Leveraging Biomaterial Platforms to Study Aging-Related Neural and Muscular Degeneration.

Aging is a complex multifactorial process that results in tissue function impairment across the whole organism. One of the common consequences of this process is the loss of muscle mass and the associated decline in muscle function, known as sarcopenia. Aging also presents with an increased risk of developing other pathological conditions such as neurodegeneration. Muscular and neuronal degeneration cause mobility issues and cognitive impairment, hence having a major impact on the quality of life of the older population. The development of novel therapies that can ameliorate the effects of agi

Investigating the effects of a single ASPM variant (c.8508_8509) on brain architecture among siblings in a consanguineous Pakistani family.

BACKGROUND: Autosomal Recessive Primary Microcephaly (MCPH) is a rare, neurodevelopmental disorder associated with mild to severe mental retardation. It is characterized by reduced cerebral cortex that ultimately leads to reduction in skull size less than - 3 S.D below the mean for normal individuals having same age and sex. Till date, 30 known loci have been reported for MCPH. METHODS: In the present study, Sanger sequencing was performed followed by linkage analysis to validate the mutation in ASPM gene of the consanguineous Pakistani clans. Bioinformatics tools were also used to confirm th

The Accessory Limb Model Regenerative Assay and Its Derivatives.

When the Accessory Limb Model (ALM) regenerative assay was first published by Endo, Bryant, and Gardiner in 2004, it provided a robust system for testing the cellular and molecular contributions during each of the basic steps of regeneration: the formation of the wound epithelium, neural induction of the apical epithelial cap, and the formation of a positional disparity between blastema cells. The basic ALM procedure was developed in the axolotl and involves deviating a limb nerve into a lateral wound and grafting skin from the opposing side of the limb axis into the site of injury. In this ch

Biological Impact on the Stability and Reliability of Acute and Chronic Platinum based Thin Film Neural Interfaces in Vivo.

Reliability, stability and biocompatibility of an im-plant are the keys to transferring a preclinical study into the re-ality of clinical applications for diagnostics and therapy of pa-tients. Amongst the smallest and most critical components of neuroprostheses are the neural interfaces to the tissue. These could be seen as the most functional and yet most sensitive parts to connect to and interact with the nervous system. Thin film systems in the submicro- to nanometers range with a high num-ber of channels record biological signals and excite nerves aspiring high spatial sensitivity at the s
