Role of Rb during Neurogenesis and Axonal Guidance in the Developing Olfactory System.

The Retinoblastoma protein, Rb, was shown to regulate distinct aspects of neurogenesis in the embryonic and adult brain besides its primary role in cell cycle control. It is still unknown, however, whether Rb is required for tissue morphogenesis and the establishment of synaptic connections between adjacent tissues during development. We have investigated here the role of Rb during development of the olfactory system (OS), which heavily relies on reciprocal interactions between the olfactory epithelium (OE) and the olfactory bulb (OB). We show that mice carrying a telencephalic-specific deleti…

Olfactory ensheathing cells: biology in neural development and regeneration.

Olfactory ensheathing cells (OECs) constitute a unique population of glia that accompany and ensheath the primary olfactory axons. They are … ological properties of OECs. Their functions in olfactory neurogenesis, olfactory axonal growth and olfactory bulb formation are sequently discussed. We also describe therapeutic applications of OECs for the treatment of a variety of neural lesions, including spinal cord injury, stroke, degenerative diseases, and PNS injuries. Finally, we address issues that may …

Anosmin-1 immunoreactivity during embryogenesis in a primitive eutherian mammal.

Kallmann syndrome is hypogonadotropic hypogonadism coupled with anosmia. A morphological study found that the endocrine disorder in X-linked Kallmann syndrome is due to failed migration of gonadotropin releasing-hormone (GnRH) neurons from the olfactory placode to the brain during development. Anosmia results from agenesis of the olfactory bulbs and tracts. The gene responsible for the X-linked form of Kallmann syndrome, KAL-1, has been characterized. The orthologues of KAL-1 have been isolated in the chick and the zebrafish, but still await identification in rodents. In the present study, we …

Quantitative MRI Assessment of Olfactory System in Septo-Optic Dysplasia Spectrum and Related Midline Brain Malformations.

BACKGROUND AND PURPOSE: Although olfactory system abnormalities have been described in septo-optic dysplasia (SOD), prior studies have relied on qualitative or semiquantitative assessment, and the reproducibility of continuous MRI measurements remains unknown. We assessed the reproducibility of quantitative MRI measurements of the olfactory sulcus (OS) and olfactory bulb (OB) in patients with the SOD spectrum and related midline brain malformations, and examined their associations with consensus olfactory system abnormality scores. Neurogenesis in olfactory epithelium: loss and recovery of transepithelial voltage transients following olfactory nerve section.

Visual inspection indicated that the olfactory nerve was reconstituted and reconnected to the olfactory bulb between 30-60 days following transection. The fact that physiological activity was recorded in the epithelium prior to this event suggests that molecular recognition and sensory transduction are not dependent on connectivity with the olfactory bulb. …

Diagnosis of Kallmann Syndrome in a Young Adult Male.

Kallmann syndrome is an anosmic variety of GnRH (gonadotropin-releasing hormone) deficiency. A young adult male with anosmia since childhood presented with features of failed sexual maturation and underwent an ultrasound of the genital system and an MRI (magnetic resonance imaging) of the brain. MRI revealed absent olfactory bulbs in bilateral olfactory grooves with hypoplastic olfactory sulci on both sides. Ultrasound showed atrophic testes, while the anterior pituitary appeared normal on MRI. The clinical, hormonal, and imaging findings were characteristic of Kallmann syndrome. MRI serves as…

Olfactory marker protein: turnover and transport in normal and regenerating neurons.

A 19,000-dalton acidic protein designated olfactory marker protein (OMP) is a cell-specific marker of mature olfactory chemosensory neurons. Intranasal irrigation of mouse olfactory epithelium with [35S]methionine labeled OMP to high specific activity. Turnover and transport characteristics of 35S-labeled OMP were compared to those of 35S-labeled global cytosol protein in groups of young, adult, and Triton-treated adult mice. The latter contained primarily large numbers of regenerating olfactory neurons. In olfactory epithelium of young and Triton-treated mice, the specific activity of OMP was

Hydrogen sulfide as an oxygen sensor in trout gill chemoreceptors.

Olfactory information is processed by a diverse group of interconnected forebrain regions. Most efforts to define the cellular mechanisms involved in processing olfactory information have been focused on understanding the function of the olfactory bulb, the primary second-order olfactory region, and its principal target, the piriform cortex. However, the olfactory bulb also projects to other targets, including the rarely studied olfactory tubercle, a ventral brain region recently implicated in regulating cocaine-related reward behavior. We used whole cell patch-clamp recordings from rat tuberc…

Neonatal capsaicin treatment impairs functional properties of primary olfactory afferents in the rat.

Long-lasting influences of neonatal capsaicin treatment on functional properties of the olfactory nerve were studied in 30-day-old rats by determining excitability thresholds of nerve fibres by means of orthodromic field potentials elicited in the main olfactory bulb, as well as by analyzing the pattern of the low-frequency component of the bulbar electroencephalogram. In addition, body, brain and olfactory bulbar weights were measured. Neonatal capsaicin resulted in reduced excitability of primary olfactory afferents and reduced wave amplitude of the bulbar electroencephalogram. Capsaicin tre…

Extracellular matrix protein anosmin-1 regulates Schwann cell-astrocyte interaction for regenerative axon targeting in dorsal root crush injury model.

Schwann cell (SC) transplantation is considered as a promising strategy for spinal cord injury. However, SCs show less capability in assisting the regenerative axons to penetrate through astrocyte (AS)-formed scar barrier. Anosmin-1, an extracellular matrix glycosylated adhesion protein expressed in the olfactory bulb, is involved in olfactory ensheathing cells and reborn olfactory nerve axons continually penetrating the glial barrier and targeting the olfactory bulb. In this study, we employ a dorsal root crush injury model treated with anosmin-1. A vertical climbing test was used for behavio…

Panglial gap junctions between astrocytes and olfactory ensheathing cells mediate transmission of Ca2+ transients and neurovascular coupling.

Astrocytes are arranged in highly organized gap junction-coupled networks, communicating via the propagation of Ca2+ waves. Astrocytes are gap junction-coupled not only to neighboring astrocytes, but also to oligodendrocytes, forming so-called panglial syncytia. It is not known, however, whether glial cells in panglial syncytia transmit information using Ca2+ signaling. We used confocal Ca2+ imaging to study intercellular communication between astrocytes and olfactory ensheathing glial cells (OECs) in in-toto preparations of the mouse olfactory bulb. Our results demonstrate that Ca2+ transient…
