Helminth parasites of the Neotropical treefrog Itapotihyla langsdorffii in southeastern Brazil.

Itapotihyla langsdorffii is a large-bodied hylid frog endemic to the Atlantic Forest, occurring in Brazil, Paraguay and northern Argentina. … . On the other hand, host size did not seem to influence the infection intensity of the most common parasites. The present survey indicated that Itapotihyla langsdorffii has one of the richest endoparasite faunas among South American hylids, including both heteroxenous and monoxenous species (although they seemingly serve as final hosts only for the latter) …

Cost-Effectiveness of Treatment for Canine Parasites in Remote Indigenous Communities.

Zoonotic canine parasites and the vector-borne diseases they may carry can cause high morbidity and mortality in dogs and people. Many remote Indigenous communities in northern Australia have numerous free-roaming dogs, tropical climates favouring parasite development, and limited access to veterinary care, which can promote high prevalence of zoonotic parasites. To successfully combat parasites, treatment programs are needed, which are effective in reducing parasite burden and prevalence as well as being cost-effective and feasible. We compared canine parasite treatments in a Torres Strait Is…

Supersaturation mutagenesis reveals adaptive rewiring of essential genes among malaria parasites.

Malaria parasites are highly divergent from model eukaryotes. Large-scale genome engineering methods effective in model organisms are frequently inapplicable, and systematic studies of gene function are few. We generated more than 175,000 transposon insertions in the Plasmodium knowlesi genome, averaging an insertion every 138 base pairs, and used this "supersaturation" mutagenesis to score essentiality for 98% of genes. The density of mutations allowed mapping of putative essential domains within genes, providing a completely new level of genome annotation for any Plasmodium species. Although…

Gastrointestinal helminth parasites of urban and rural foxes around Melbourne, Australia.

The red fox (Vulpes vulpes) is an introduced species to Australia whose population and spatial distribution have grown irreversibly. Due to their opportunistic feeding habits, extensive populations of foxes now inhabit urban and rural environments, where they coexist with humans and domesticated animals. The proximity of these predators presents public and animal health concerns as they harbour diseases that can cross between species. Accordingly, monitoring potential disease risk and prevalence in urban foxes is warranted. This study investigated the occurrence of gastrointestinal parasites i…

Independent nuclear and organellar mechanisms determine apicoplast fate in malaria parasites.

The apicoplast organelle of the malaria parasite, Plasmodium falciparum, is essential for parasite replication, though its cell cycle regulation remains poorly understood. We developed a dynamic live-imaging platform with analytical capabilities to track subcellular structures throughout the parasite's 48-h intraerythrocytic life cycle. Our analysis revealed four distinct morphological stages in apicoplast development that correlate with nuclear replication. We identified a critical "Crown" morphology stage required for nucleus-apicoplast attachment, where the apicoplast stretches across multi

Liver parasites: A global endemic and journey from infestation to intervention.

Parasites have coexisted with humans throughout history, forming either symbiotic relationships or causing significant morbidity and mortality. The liver is particularly vulnerable to parasitic infections, which can reside in, pass through, or be transported to the liver, leading to severe damage. This editorial explores various parasites that infect the liver, their clinical implications, and diagnostic considerations, as discussed in the article "Parasites of the liver: A global problem?". Parasites reach the liver primarily through oral ingestion, mucosal penetration, or the bloodstream, wi…

Parasites of Stray Cats in Iran: A Parasitological and Histopathological Study.

Many zoonotic parasitic diseases, including Toxocara cati, may be spread by stray cat populations. This study aimed to determine the prevalence of parasites by performing parasitological and histopathological examinations on stray cats in Shiraz, Iran. A total of 106 stray cats from different geographical areas of Shiraz, southern Iran, were examined for the presence of parasites. The overall prevalence was found to be 83.02% (88/106), and eight parasites were found. The parasites included three genera of cestodes [Joyeuxiella echinorhynoides (52.83%), Taenia taeniaeformis (21.70%), and Dipyli…

Detection of Parasites in the Field: The Ever-Innovating CRISPR/Cas12a.

The rapid and accurate identification of parasites is crucial for prompt therapeutic intervention in parasitosis and effective epidemiological surveillance. For accurate and effective clinical diagnosis, it is imperative to develop a nucleic-acid-based diagnostic tool that combines the sensitivity and specificity of nucleic acid amplification tests (NAATs) with the speed, cost-effectiveness, and convenience of isothermal amplification methods. A new nucleic acid detection method, utilizing the clustered regularly interspaced short palindromic repeats (CRISPR)-associated (Cas) nuclease, holds p…

Monitoring of Lipid Fluxes Between Host and Plastid-Bearing Apicomplexan Parasites.

Apicomplexan parasites are unicellular eukaryotes responsible for major human diseases such as malaria and toxoplasmosis, which cause massive social and economic burden. Toxoplasmosis, caused by Toxoplasma gondii, is a global chronic infectious disease affecting ~1/3 of the world population and is a major threat for any immunocompromised patient. To date, there is no efficient vaccine against these parasites and existing treatments are threatened by rapid emergence of parasite resistance. Throughout their life cycle, Apicomplexa require large amount of nutrients, especially lipids for propagat…

Imaging Assays to Detect DNA Damage in Trypanosome Parasites Using γH2A.

Diseases caused by trypanosomatid parasites remain a significant unmet medical need for millions of people globally. Trypanosomatid parasites such as Trypanosoma cruzi and subspecies of Trypanosoma brucei cause Chagas disease and human African trypanosomiasis (HAT), respectively. Although efforts to find novel treatments have been successful for HAT, Chagas disease is still treated with decades-old therapies that suffer from long treatment durations and severe safety concerns. We recently described the identification and characterization of the cyanotriazole compound class that kills trypanoso…

The Dominance of Coinfecting Parasites' Indirect Genetic Effects on Host Traits.

AbstractIndirect genetic effects (IGEs) exist when there is heritable variation in one organism's ability to alter a second organism's traits. For example, parasites have antigens that can induce a host immune response, as well as disparate strategies to evade or suppress host immunity; among-parasite genetic variation in these antigens generates among-host variation in immune traits. Here, we experimentally show that the cestode parasite Schistocephalus solidus exerts an IGE on an immune trait (peritoneal fibrosis) in its threespine stickleback host: stickleback developed strong fibrosis afte…

The interdependence of isoprenoid synthesis and apicoplast biogenesis in malaria parasites.

Isoprenoid precursor synthesis is an ancient and fundamental function of plastid organelles and a critical metabolic activity of the apicoplast in Plasmodium malaria parasites [1-3]. Over the past decade, our understanding of apicoplast properties and functions has increased enormously [4], due in large part to our ability to rescue blood-stage parasites from apicoplast-specific dysfunctions by supplementing cultures with isopentenyl pyrophosphate (IPP), a key output of this organelle [5,6]. In this Pearl, we explore the interdependence between isoprenoid metabolism and apicoplast biogenesis i…

Zoonotic Parasites in Playgrounds in Southern Spain: A One Health Approach.

Zoonotic parasitic diseases are considered a global threat to public health. In this sense, canines and felines may be infected by different cosmopolitan parasites, with playgrounds serving as an important focus of infection for humans, as well as domestic or wild animals. Knowledge of the epidemiological situation of parasites in animal reservoirs integrated into the environment, identifying the spread pathways, is a key element for an effective response to this threat. Thus, the aim of this study was to assess the frequency of intestinal parasites with zoonotic potential in 120 playgrounds i…

Parasites in ancient Egypt and Nubia: Malaria, schistosomiasis and the pharaohs.

The civilizations of ancient Egypt and Nubia played a key role in the cultural development of Africa, the Near East, and the Mediterranean world. This study explores how their location along the River Nile, agricultural practices, the climate, endemic insects and aquatic snails impacted the type of parasites that were most successful in their populations. A meta-analysis approach finds that up to 65% of mummies were positive for schistosomiasis, 40% for headlice, 22% for falciparum malaria, and 10% for visceral leishmaniasis. Such a disease burden must have had major consequences upon the phys…

Progeny counter mechanism in malaria parasites is linked to extracellular resources.

Malaria is caused by the rapid proliferation of Plasmodium parasites in patients and disease severity correlates with the number of infected red blood cells in circulation. Parasite multiplication within red blood cells is called schizogony and occurs through an atypical multinucleated cell division mode. The mechanisms regulating the number of daughter cells produced by a single progenitor are poorly understood. We investigated underlying regulatory principles by quantifying nuclear multiplication dynamics in Plasmodium falciparum and knowlesi using super-resolution time-lapse microscopy. Thi…

Demographic, environmental and physiological predictors of gastrointestinal parasites in urban raccoons.

Raccoons are host to diverse gastrointestinal parasites, but little is known about the ecology of these parasites in terms of their interactions with each other during coinfections, their interactions with host physiology and environmental factors, and their impact on raccoon health and survival. As a first step, we investigated the patterns of parasite infection and their demographic distribution in an urban-suburban population of raccoons trapped in the summers and autumns of 2018 and 2019. We collected faecal samples, demographic data, morphometric measurements, and blood smears, and used G…
