Rapid Classification of Sarcomas Using Methylation Fingerprint: A Pilot Study.

Sarcoma classification is challenging and can lead to treatment delays. Previous studies used DNA aberrations and machine-learning classifiers based on methylation profiles for diagnosis. We aimed to classify sarcomas by analyzing methylation signatures obtained from low-coverage whole-genome sequencing, which also identifies copy-number alterations. DNA was extracted from 23 suspected sarcoma samples and sequenced on an Oxford Nanopore sequencer. The methylation-based classifier, applied in the nanoDx pipeline, was customized using a reference set based on processed Illumina-based methylation

Microwave imaging for monitoring breast cancer treatment: A pilot study.

BACKGROUND: Microwave imaging has been proposed for medical applications, creating maps related to water content of tissues. Breast imaging has emerged as a key application because the signals can be coupled directly into the breast and experience limited attenuation in fatty tissues. While the literature contains reports of tumor detection with microwave approaches, there is limited exploration of treatment monitoring. PURPOSE: This study aims to detect treatment-related changes in breast tissue with a low-resolution microwave scanner. METHODS: Microwave scans of 15 patients undergoing trea

Effects of Monacolin K in Nondiabetic Patients with NAFLD: A Pilot Study.

Non-alcoholic fatty liver disease (NAFLD) is a common chronic liver condition with significant risk of progression to steatohepatitis and cirrhosis. Therapeutic strategies in NAFLD include lifestyle changes mainly related to dietary interventions and use of drugs or nutritional components that could improve plasma lipid profiles and insulin sensitivity and decrease the local inflammatory response. In this study, we tested the effects of monacolin K, an inhibitor of HMCoA reductase. In a prospective, uncontrolled, open study, we treated 24 patients with NAFLD and mild hypercholesterolemia with

Periodontitis Risk Factors in Adults with Cystic Fibrosis: A pilot study.

Purpose Individuals with cystic fibrosis (CF) present with multiple condition-specific risk factors for periodontitis including CF-related diabetes, chronic inhaled treatments that induce xerostomia, and increased systemic inflammation because of frequent lung infections. General factors like age, oral hygiene, and diet may also contribute to the risk of periodontitis. However the relative importance of these specific risk factors and periodontitis in individuals with CF has not yet been evaluated. The purpose of this pilot study was to assess the associations between CF condition-specific and

TRPS1 immunohistochemical expression in salivary gland tumors: A pilot study.

OBJECTIVES: TRPS1 is a new, sensitive marker for breast carcinoma (BC). Salivary glands and breasts are both exocrine glands; thus, their tumors may share similar morphology and immunophenotype. Among salivary gland-type BC, TRPS1 is reported to be positive in secretory carcinomas (SCs) but negative in acinic cell carcinomas (AciCCs) and most adenoid cystic carcinomas (AdCCs). A subset of salivary duct carcinomas (SDCs) is positive for TRPS1. Herein, we investigate TRPS1 immunohistochemical expression in salivary gland tumors (SGTs). METHODS: A retrospective search yielded 110 SGTs (97 primar

Efficiency of portable air purification on public buses: A pilot study.

High concentrations of fine particulate matter (PM2.5) have been frequently reported in public transit systems and can cause adverse health effect. The portable air purifier is an inexpensive solution that could potentially clean in-cabin PM2.5. This study aims to find the PM2.5 removal efficiency of portable air purifiers in a public transit bus. In various scenarios, after artificially preloading the in-cabin PM2.5 concentration to 400 μg/m3, the concentrations were measured every 10 s, with and without the intervention of air purifiers. In a test bus with a volume of approximately 62.5 m3,

Low cycle fatigue properties of porcine aorta - Pilot study.

INTRODUCTION: Biomechanical rupture risk assessment of aortic tissues is commonly based on computed stress to measured uniaxial static strength comparison. Loading of the arterial wall, however, is cyclic; thus, the static strength may not be a proper limit value. This study investigates the low cycle fatigue of porcine aortic samples tested in a circumferential direction. METHODS: 7 porcine descending aorta (both thoracic and abdominal) were harvested and 56 dogbone-shaped samples were prepared. Static strength was measured, the limit of engineering stress was chosen and then force controlle

Pilot study of newborn screening for six lysosomal diseases in Brazil.

BACKGROUND: Lysosomal diseases (LDs) are progressive life-threatening disorders that are usually asymptomatic at birth. Specific treatments are available for several LDs, and early intervention improves patient's outcomes. Thus, these diseases benefit from newborn screening (NBS). We have performed a pilot study for six LDs in Brazil by tandem mass spectrometry. METHODS: Dried blood spot (DBS) samples of unselected newborns were analyzed by the Neo-LSD™ kit (Perkin-Elmer) by MS/MS. Samples with low enzyme activity were submitted to the evaluation of specific biomarkers by ultra-performance li
