Enrichment of provitamin A content in durum wheat grain by suppressing β-carotene hydroxylase 1 genes with a TILLING approach.

The suppression of the HYD-1 gene by a TILLING approach increases the amount of β-carotene in durum wheat kernel. Vitamin A deficiency is a major public health problem that affects numerous countries in the world. As humans are not able to synthesize vitamin A, it must be daily assimilated along with other micro- and macronutrients through the diet. Durum wheat is an important crop for Mediterranean countries and provides a discrete amount of nutrients, such as carbohydrates and proteins, but it is deficient in some essential micronutrients, including provitamin A. In the present work, a targe

The metabolomic profile of red non-V. vinifera genotypes.

Wild American genotypes represent an important part of the Vitis germplasm in relation to grape improvement. Today, these genotypes are currently involved in breeding programmes in order to introgress traits resistant to pests and diseases in V. vinifera cultivars. Nevertheless, the metabolic composition of their grapes has not been widely investigated. This study aimed to explore in detail the metabolomic profile in terms of simple phenolic, proanthocyanidin, anthocyanin and lipid compounds in two hybrids and five American genotypes. The results were compared with those of two V. vinifera cul

New insights into wheat toxicity: Breeding did not seem to contribute to a prevalence of potential celiac disease's immunostimulatory epitopes.

Gluten proteins, namely gliadins, are the primary trigger of the abnormal immune response in celiac disease. It has been hypothesised that modern wheat breeding practices may have contributed to the increase in celiac disease prevalence during the latter half of the 20th century. Our results do not support this hypothesis as Triticum aestivum spp. vulgare landraces, which were not subjected to breeding practices, presented higher amounts of potential celiac disease's immunostimulatory epitopes when compared to modern varieties. Furthermore, high variation between wheat varieties concerning the

Probability of success of breeding strategies for improving pro-vitamin A content in maize.

Biofortification for pro-vitamin A content (pVAC) of modern maize inbreds and hybrids is a feasible way to deal with vitamin A deficiency in rural areas in developing countries. The objective of this study was to evaluate the probability of success of breeding strategies when transferring the high pVAC present in donors to elite modern-adapted lines. For this purpose, a genetic model was built based on previous genetic studies, and different selection schemes including phenotypic selection (PS) and marker-assisted selection (MAS) were simulated and compared. MAS for simultaneously selecting al

Associations between oil- and gas-well sites, processing facilities, flaring, and beef cattle reproduction and calf mortality in western Canada.

From the fall of 1992 through calving 1996, detailed cow breeding outcome records were maintained actively for seven large cow-calf herds in western Canada. The numbers of mature females in the study for the breeding seasons beginning in 1992, 1993, 1994, and 1995 were 1122, 1177, 1251, and 1236, respectively. Outcomes included pregnancy status, calving interval, and the occurrence of twins, abortions, stillbirths, and neonatal mortality. Information also was collected on other risk factors known to influence beef-herd health and productivity. Detailed maps of active and inactive oil and natur

Prospects of microgreens as budding living functional food: Breeding and biofortification through OMICS and other approaches for nutritional security.

Nutrient deficiency has resulted in impaired growth and development of the population globally. Microgreens are considered immature greens (required light for photosynthesis and growing medium) and developed from the seeds of vegetables, legumes, herbs, and cereals. These are considered "living superfood/functional food" due to the presence of chlorophyll, beta carotene, lutein, and minerals like magnesium (Mg), Potassium (K), Phosphorus (P), and Calcium (Ca). Microgreens are rich at the nutritional level and contain several phytoactive compounds (carotenoids, phenols, glucosinolates, polyster

BSA-seq-based identification of candidates associated with stable resistance to Ralstonia pseudosolanacearum in pepper.

Bacterial wilt is a major constraint on pepper production, and the development of resistant cultivars is the most effective and sustainable strategy for disease management. However, pepper germplasm with stable resistance and its genetic basis remain insufficiently characterized. In this study, 63 pepper accessions were screened for bacterial wilt resistance using a leaf-to-whole plant bioassay. Based on disease severity index values, seven accessions were classified as extremely resistant (ER), 25 as resistant, 22 as moderately susceptible, and nine as susceptible. Selected accessions were fu

A ShbHLH34-ShPUR Module Activates ShTPS1-mediated β-caryophyllene Biosynthesis for Insect Resistance in Sugarcane.

Terpenoids represent one of the largest and most structurally diverse classes of plant secondary metabolites, playing essential roles in defense against herbivorous insects. Despite their importance, the mechanisms regulating terpenoid biosynthesis remain largely unexplored. Here, in sugarcane, we identified and characterized a terpene synthase gene from Saccharum spp. Hybrids, ShTPS1, which is strongly induced by feeding from fall armyworm Spodoptera frugiperda and sugarcane stem borer Chilo infuscatellus. Enzymatic assays in vitro revealed that ShTPS1 catalyzes the biosynthesis of volatile t

Natural Variations in GhFLA9_D12 Confer Divergent Fiber Development and Lint Percentage via Cell Wall Plasticity Remodeling in Cotton.

Lint percentage (LP) is a core determinant of cotton yield. Here, we systematically characterized the evolutionary impacts and molecular mechanisms of GhFLA9_D12 on fiber development. Population genomics revealed a significant selective sweep at the locus, where the superior haplotype HapA (T/INS) correlates with higher LP and underwent continuous selection during modern breeding. This favorable variation was likely introgressed from G. barbadense into G. hirsutum. Mechanistically, GhFLA9_D12 regulates fiber development via divergent expression and protein functions. Transcriptionally, a 162-b

Identification of genes encoding GRF transcription factors in Taraxacum kok-saghyz Rodin and their potential role in response to salt stress.

GRF transcription factors (GRF) are pivotal regulators of plant development and stress tolerance, but their functional repertoire in the rubber crop Taraxacum kok-saghyz (Russian dandelion) remains entirely uncharacterized. Genome-wide analysis of the T. kok-saghyz genome identified 10 TkGRF members, unevenly distributed across eight chromosomes and phylogenetically resolved into five clades. All TkGRFs contained the canonical QLQ and WRC domains, confirming their identity as bona fide GRF transcription factors. Synteny analysis further established close evolutionary conservation of these 10 T

Virulence Profiles and Fungicide-Target-Gene Variability in Puccinia graminis f. sp. tritici Populations from Russian Regions.

Wheat stem rust, caused by Puccinia graminis f. sp. tritici (Pgt), threatens food security owing to the pathogen's high virulence and evolutionary variability, as well as the risk of selecting fungicide-resistant forms. For the first time in Russia, we performed a comparative analysis of virulence and genetic diversity in fungicide-target genes (SdhA-SdhD, Cyp51) in 35 monopustular Pgt isolates collected from regions with contrasting fungicide pressure (Saratov, n = 12; Chelyabinsk, n = 12; Moscow, n = 11). iPheno: A Novel Dual-Aware Vision-Language Model for Multi-Task Fine-Scale Crop Phenotyping.

Crop phenotyping is crucial for advancing plant breeding, yet remains a significant challenge. Manual approaches are labor-intensive and do not scale to the analysis of large datasets, while computational methods like Vision-Language Models (VLMs) lack the adaptability for fine-scale spatial reasoning and diverse phenotyping scenarios. To bridge the gaps, we present iPheno, a fully open-source, domain-specialized multimodal VLM for fine-scale crop phenotyping. A key innovation of iPheno is its dual-aware architecture. First, a spatial-aware feature extractor samples mask regions into local K-N…

Genome-Wide Characterization of Calmodulin-Binding Transcription Activators Genes in Aegilops tauschii.

Calcium signaling plays a central role in plant adaptation to abiotic stresses and is primarily mediated by calmodulin and its associated transcription factors. Calmodulin-binding transcription activators (CAMTAs) regulate stress-responsive gene expression, but their characteristics and functions remain largely unexplored in Aegilops tauschii Coss., the D-genome progenitor of bread wheat. In this study, a genome-wide identification and characterization of the CAMTA gene family was performed, followed by phylogenetic, structural, conserved domain, promoter cis-element, and expression analyses u

Receptor-Like Kinase ZmRLK9 Functions as an Upstream ABA Signaling Factor Conferring Drought Resilience in Maize.

The phytohormone abscisic acid (ABA) plays a pivotal role in plant drought responses. Although a substantial amount of research has concentrated on intracellular ABA receptors and SnRK2 protein kinases as the central signaling components, how the extracellular ABA signal is transduced to this core pathway remains largely unknown. Here, through a genome-wide association study mapping drought resistance in maize at the flowering stage, we identify a 4,052-bp hAT transposon insertion upstream of a leucine-rich repeat receptor-like kinase gene, named ZmRLK9, that suppresses gene expression and com

A novel CENH3-based immunostaining method for ploidy estimation in potato.

Accurate and reliable ploidy estimation is essential for potato genetic improvement programmes, yet current methods face limitations in cost, throughput, or technical complexity. Here, an immunofluorescence assay was developed that employs a species-specific antibody to label the centromeric histone CENH3 in Solanum leaf cell nuclei. The method was validated on genotypes of known ploidy (2x, 3x, 4x) and compared against flow cytometry and root-tip chromosome counting. The antibody specificity was examined across a range of both related and distant plant species. The results demonstrated that C

Ramu stunt virus genome reveals previously unreported segments and nucleocapsid domain duplication in Mechlorovirus.

Ramu stunt virus (RmSV), a member of the genus Mechlorovirus within the family Phenuiviridae, was previously described as a six-segmented RNA virus infecting sugarcane. In this study, we re-examined type material and additional isolates using high-throughput sequencing and RT-PCR validation, revealing that RmSV possesses a nine-segmented genome, making it the largest reported in the Phenuiviridae. This expanded architecture includes duplicated RNA segments (RNA 2a and RNA 2b) encoding nucleocapsid-like proteins and two novel segments (RNA 7 and RNA 8). Comparative analysis showed that RNA 2a a

Integrative metabolomic and transcriptomic profiling deciphers flavonoid biosynthesis of bulbs in Lilium brownii var. Viridulum.

Lilium brownii var. viridulum is rich in flavonoids, which offer antioxidant, anti-inflammatory, antitumor, and immunomodulatory effects. Recently, a novel, more resistant variant called L. brownii var. viridulum (Xuefeng, xf) was identified through individual plant selection. However, the metabolic and transcriptional profiles of xf, particularly concerning flavonoid metabolism, have not yet been compared to those of the traditional variant (Longya, ly). To address this gap, we performed an integrated metabolomic and transcriptomic analysis to systematically compare flavonoid metabolism betwe
