Barley as a production platform for oral vaccines in sustainable fish aquaculture.

Vaccination is the most effective measure to prevent disease outbreaks in fish aquaculture, with oral vaccine administration emerging as the most practical approach. However, oral vaccines face a notable limitation due to insufficient stimulation of the complex gut-associated lymphoid tissue caused by factors such as vaccine degradation, poor absorption, and recognition by the immune cells. An innovative solution to these limitations lies in the plant-based production of recombinant vaccines. Plant cells enable the production and targeted storage of recombinant vaccines in specific cell organe

Sulfur-Induced Resistance against Pseudomonas syringae pv. actinidiae via Triggering Salicylic Acid Signaling Pathway in Kiwifruit.

Sulfur has been previously reported to modulate plant growth and exhibit significant anti-microbial activities. However, the mechanism underlying its diverse effects on plant pathogens has not been elucidated completely. The present study conducted the two-year field experiment of sulfur application to control kiwifruit canker from 2017 to 2018. For the first time, our study uncovered activation of plant disease resistance by salicylic acid after sulfur application in kiwifruit. The results indicated that when the sulfur concentration was 1.5-2.0 kg m-3, the induced effect of kiwifruit canker

Humans have antibodies against a plant virus: evidence from tobacco mosaic virus.

Tobacco mosaic virus (TMV), a widespread plant pathogen, is found in tobacco (including cigarettes and smokeless tobacco) as well as in many other plants. Plant viruses do not replicate or cause infection in humans or other mammals. This study was done to determine whether exposure to tobacco products induces an immune response to TMV in humans. Using a sandwich ELISA assay, we detected serum anti-TMV antibodies (IgG, IgG1, IgG3, IgG4, IgA, and IgM) in all subjects enrolled in the study (20 healthy smokers, 20 smokeless-tobacco users, and 20 non-smokers). Smokers had a higher level of serum an

Advancing Global Hepatitis B Elimination: The Case for Using Maize as a Low-Cost, Heat-Stable, and Scalable Oral Vaccine.

Because hepatitis B virus (HBV) remains a major global health burden, innovative strategies are essential to achieve the World Health Organization's goal of eliminating viral hepatitis and closing persistent coverage gaps for injectable vaccines. While parenteral administration remains the gold standard for immunization, constraints such as cold-chain dependence and needle-associated barriers limit its reach, particularly in resource-constrained environments. This review summarizes work aimed at a plant-produced orally delivered vaccine as a transformative, scalable step towards global hepatit

Immunological monitoring of dry-cleaning shop workers--exposure to tetrachloroethylene.

A panel of immunological parameters has been examined in a group of dry-cleaning workers (n = 21) and in a control group of administrators (n = 16) from the same plant. The results were also compared to long-term laboratory reference values (LRV) (n = 14-311). External exposure to tetrachloroethylene (PER) was represented by TWA (8 h) values in the range 11-752 mg PER/m3. Biological monitoring showed an amount from 9 to 344 mg PER/m3 in exhaled air by the end of workshift. 1. The exposed dry-cleaning workers compared to the controls from the plant had statistically significant changes in metab

CO2 signalling in plants.

Elevated atmospheric CO2 concentrations increase the productivity of plants by enhancing the photosynthesis/photorespiration balance. It has long been recognized that CO2 acts as a signal, for example, in the regulation of stomatal closure, as well as the substrate of photosynthesis. Early concepts of CO2 signalling in plants focused on a mode of action based on alterations in primary metabolism, particularly sugar availability and signalling. However, stomatal guard cells employ a CO2 sensing and signalling mechanism that is independent of sugars. We discuss the possibility that a similar pat

Flaxseed Extracts Impact the Cellular Structure of a Keratinocyte Model for Oral Lichen Planus-A Preliminary Study.

Oral inflammation and the immune response are distinct but related processes where Linum usitatissimum L., fam. Linaceae represents a possible use for localized relief. Oral lichen planus (OLP) is an oral potentially malignant disorder (OPMD) with an inflammatory background that mainly affects post- and peri-menopausal women. The presented methodology was threefold. Firstly, the plant extracts were made from flaxseeds of selected cultivars (Szafir [SZ] and Jantarol [JA]) containing plant lignans. In silico docking affinity was performed to verify the beta and alpha estrogen receptors of kerati

Phytodegradation of neonicotinoids in Cyperus papyrus from enzymatic and transcriptomic perspectives.

Neonicotinoids are widely used but environmentally hazardous insecticides. Constructed wetlands offer potential for neonicotinoid removal, but the corresponding metabolic pathways and mechanisms in wetland plants are incompletely understood. This study investigated the fate of six neonicotinoids and their metabolites in Cyperus papyrus, a common wetland plant, and the underlying metabolic mechanisms through enzymatic and transcriptomic analyses. Neonicotinoids were absorbed by roots and translocated upward, causing high levels in shoots. Concentrations of neonicotinoids and their metabolites d

A novel plant-made monoclonal antibody enhances the synergetic potency of an antibody cocktail against the SARS-CoV-2 Omicron variant.

This study describes a novel, neutralizing monoclonal antibody (mAb), 11D7, discovered by mouse immunization and hybridoma generation, against the parental Wuhan-Hu-1 RBD of SARS-CoV-2. We further developed this mAb into a chimeric human IgG and recombinantly expressed it in plants to produce a mAb with human-like, highly homogenous N-linked glycans that has potential to impart greater potency and safety as a therapeutic. The epitope of 11D7 was mapped by competitive binding with well-characterized mAbs, suggesting that it is a Class 4 RBD-binding mAb that binds to the RBD outside the ACE2 bin

In vitro Inhibition of HIV-1 by Cyclotide-Enriched Extracts of Viola tricolor.

Since viral infectious diseases continue to be a global health threat, new antiviral drugs are urgently needed. A unique class of therapeutic compounds are antimicrobial peptides (AMPs). They can be found in humans, bacteria and plants. Plants express a wide variety of such defense peptides as part of their innate immune system to protect from invading pathogens. Cyclotides are non-classical AMPs that share a similar structure. Their unique topology consists of a circular peptide backbone and disulfide bonds. In previous studies they have been attributed to a wide range of biological activitie

[Application of artificial DNA-binding proteins and artificial nucleases to prevention of virus infection: development of virus-resistant plants and protein-based anti-viral drugs].

Various DNA viruses are known to cause severe infectious diseases in both plants and mammals, including humans. For many of these infectious diseases, we have yet to find an effective prevention or treatment. Therefore, new methodologies for the prevention of virus infections in both agricultural crops and humans have been vigorously sought for a long time. One attractive approach to the prevention is inhibition of virus replication. We first inhibited virus replication by blocking binding of a viral replication protein, which initiates virus replication, to its replication origin, with using

An alternative mechanism for gluten toxicity in coeliac disease.

The pathogenesis of gluten-sensitive enteropathies is unknown, although a peptidase deficiency and an immune defect have been postulated. The effect of plant-derived lectins on cells has led to an alternative concept in which a defect of the cell surface membrane allows gluten to act as a lectin and this reaction initiates cell toxicity. The proposed abnormality is viewed as a structural change produced by incomplete oligosaccharide chains in surface-membrane glycoproteins.

Global S-sulfenylation proteome analysis reveals a role of Fd-GOGAT in regulating GABA accumulation during Magnaporthe oryzae infection in rice.

Blast disease, caused by Magnaporthe oryzae, triggers complex redox signaling networks in rice involving reactive oxygen species (ROS) and reactive nitrogen species (RNS). These signaling molecules not only function as key regulators of plant immunity but also induce distinct cysteine-based post-translational modifications, namely S-sulfenylation and S-nitrosation, respectively, thereby modulating the activity, stability, and function of target proteins. While the role of S-nitrosation in rice immunity has been increasingly recognized, the contribution of S-sulfenylation remains largely unexpl…

GhNAC043 and GhBPM2 interaction enhances Verticillium wilt resistance in cotton through jasmonic acid and abscisic acid pathways.

The soil-borne fungal pathogen Verticillium dahliae (V. dahliae) is the causal agent of Verticillium wilt (VW), a vascular disease that severely threatens global cotton production. Although cell wall lignification represents a cornerstone of plant immunity, the precise regulatory circuits that bridge this structural reinforcement with Verticillium dahliae resistance in cotton have yet to be fully elucidated. Here, we demonstrate that the NAC transcription factor GhNAC043 is a key positive regulator of this defense. GhNAC043 expression was rapidly induced upon V. dahliae infection. Silencing Gh…
