Hypouricemic and renal protective effects of the Chinese functional tea Ligustrum robustum on hyperuricemic mice.

Hyperuricemia (HUA) is a prevalent metabolic disorder requiring safe, effective interventions. Ligustrum robustum (LR), a Chinese functional tea approved as a general food by the National Health Commission of the People's Republic of China, was studied for its anti-HUA and renoprotective effects in potassium oxonate and hypoxanthine-induced HUA mice. Results showed that LR significantly reduced serum uric acid and improved renal function in a dose-responsive manner without organ toxicity. Western blot analysis indicated that LR bidirectionally regulated uric acid metabolism by downregulating t

Nonsteroidal Anti-Inflammatory Drugs as Modulators of Cation Channels: Fenamates Repurposing in Channelopathies.

Cationic ion channels are transmembrane proteins that regulate the flux of cations (potassium, sodium, and calcium) across cell membrane, playing a pivotal role in many cellular functions. Disruptions of their activity can lead to the so-called genetic or acquired channelopathies, a heterogeneous group of diseases that affect multiple human systems. Fenamates, a class of nonsteroidal anti-inflammatory drugs, has recently emerged as modulators of cationic ion channels highlighting the possibility of their repurposing for the treatment ion channel-related disorders, such as channelopathies, chro

Reactivity of Cyclopropenylaluminates.

Reactions of the potassium cyclopropenylaluminates, [{SiNDipp}-Al-η2-(C2Ph2)-K] and [{SiNDipp}-Al-η2-(PhCCSiMe3)-K] with terminal alkynes provide alkynylvinylaluminate derivatives with the silyl-substituted analog providing a level of kinetic discrimination. While this latter behavior results in the regiochemical protonation at Al-C-(Ph) and retention of the more sterically congested Al-C-(SiMe3) bond, reactions with CO2 and phenyl-substituted ketones are complicated by a reduced level of discrimination and a likely tendency toward multiple CO insertion or loss of coordinated alkyne. This la

Copper difluorocarbene-involved catalytic gem-difluoropropargylation.

The use of metal for catalytic difluorocarbene transfer reactions has long been hindered by the lack of understanding of metal difluorocarbene chemistry, despite the potential implications for medicinal chemistry and advanced materials science. Here, we report a copper-catalyzed difluorocarbene transfer reaction via 1,1-migration of copper difluorocarbene, in contrast to the previous nucleophilic addition of copper difluorocarbene pathway. This reaction enables the development of a modular catalytic gem-difluoropropargylation reaction using a variety of simple and widely available potassium pr

Role of Environmental Factors in Legume-Rhizobium Symbiosis: A Review.

Legumes play a pivotal role in addressing global challenges of food and nutrition security by offering a sustainable source of protein and bioactive compounds. The capacity of legumes to establish symbiotic relationships with rhizobia bacteria enables biological nitrogen fixation (BNF), reducing the dependence on chemical fertilizers while enhancing soil health. However, the efficiency of this symbiosis is significantly influenced by environmental factors, such as soil acidity, salinity, temperature, moisture content, light intensity, and nutrient availability. These factors affect key process

Chemotaxonomy and Bioactive Potential of High-Mountain Plantago atrata.

Plantago atrata Hoppe is a high-altitude mountain plant exposed to harsh environmental factors. This study aims to elucidate the ecological, phytochemical and pharmacological characteristics of this lesser-known plantain. Despite nutrient-poor peat soil, the leaves of P. atrata contained increased levels of phosphorus, potassium and calcium, and the anatomy revealed an isobilateral mesophyll. Molecular taxonomic identification and phylogenetic analyses confirmed the classification of the studied plant as P. atrata and its clustering with narrow-leaved plantains growing in extreme conditions. Potassium/sodium cation carriers robustly upregulate CD20 antigen by targeting MYC, and synergize with anti- CD20 immunotherapies to eliminate malignant B cells.

Our investigation uncovers that nanomolar concentrations of salinomycin, monensin, nigericin, and narasin (a group of potassium/ sodium cation carriers) robustly enhance surface expression of CD20 antigen in B-cell-derived tumor cells, including primary malignant cells of chronic lymphocytic leukemia and diffuse large B-cell lymphoma. Experiments in vitro, ex vivo, and animal model reveal a novel approach of combining salinomycin or monensin with therapeutic anti-CD20 monoclonal antibodies or anti-CD20 chimeric antigen receptor T cells, significantly improving non-Hodgkin lymphoma therapy. hERG stereoselective modulation by mexiletine-derived ureas: Molecular docking study, synthesis, and biological evaluation.

Long QT syndrome (LQTS) is a disorder of cardiac electrophysiology resulting in life-threatening arrhythmias; nowadays, only a few drugs are available for the management of LQTS. Focusing our attention on LQT2, one of the most common subtypes of LQTS caused by mutations in the human ether-à-go-go-related gene (hERG), in the present work, the stereoselectivity of the recently discovered mexiletine-derived urea 8 was investigated on the hERG potassium channel. According to preliminary in silico predictions, in vitro studies revealed a stereoselective behavior, with the meso form showing the grea

Snake Venom: A Promising Source of Neurotoxins Targeting Voltage-Gated Potassium Channels.

The venom derived from various sources of snakes represents a vast collection of predominantly protein-based toxins that exhibit a wide range of biological actions, including but not limited to inflammation, pain, cytotoxicity, cardiotoxicity, and neurotoxicity. The venom of a particular snake species is composed of several toxins, while the venoms of around 600 venomous snake species collectively encompass a substantial reservoir of pharmacologically intriguing compounds. Despite extensive research efforts, a significant portion of snake venoms remains uncharacterized. Recent findings have de

Discovery of agonist-antagonist pairs for the modulation of Ca [2]+ and voltage-gated K+ channels of large conductance that contain beta1 subunits.

Large conductance, calcium/voltage-gated potassium channels (BK) regulate critical body processes, including neuronal, secretory and smooth muscle (SM) function. While BK-forming alpha subunits are ubiquitous, accessory beta1 subunits are highly expressed in SM. This makes beta1 an attractive target for pharmaceutical development to treat SM disorders, such as hypertension or cerebrovascular spasm. Compounds activating BK via beta1 have been identified, yet they exhibit low potency and off-target effects while antagonists that limit agonist activity via beta 1 remain unexplored. Beta1-dependen

Antiarrhythmic Hit to Lead Refinement in a Dish Using Patient-Derived iPSC Cardiomyocytes.

Ventricular cardiac arrhythmia (VA) arises in acquired or congenital heart disease. Long QT syndrome type-3 (LQT3) is a congenital form of VA caused by cardiac sodium channel (INaL) SCN5A mutations that prolongs cardiac action potential (AP) and enhances INaL current. Mexiletine inhibits INaL and shortens the QT interval in LQT3 patients. Above therapeutic doses, mexiletine prolongs the cardiac AP. We explored structure-activity relationships (SAR) for AP shortening and prolongation using dynamic medicinal chemistry and AP kinetics in human-induced pluripotent stem cell-derived cardiomyocytes

Dichloromethylation of enones by carbon nitride photocatalysis.

Small organic radicals are ubiquitous intermediates in photocatalysis and are used in organic synthesis to install functional groups and to tune electronic properties and pharmacokinetic parameters of the final molecule. Development of new methods to generate small organic radicals with added functionality can further extend the utility of photocatalysis for synthetic needs. Herein, we present a method to generate dichloromethyl radicals from chloroform using a heterogeneous potassium poly(heptazine imide) (K-PHI) photocatalyst under visible light irradiation for C1-extension of the enone back

ZC88, a novel N-type calcium channel blocker from 4-amino-piperidine derivatives state-dependent inhibits Cav2.2 calcium channels.

Small molecular inhibitors of Cav2.2 have been reported for the treatment of neuropathic pain; however, low selectivity and side effects limit their further development. In our study, a series of new compounds were designed and synthesized by optimizing the 4-amino-piperidine template. The results show that ZC88 inhibits transiently expressed Cav2.2 in state-dependent manner in oocytes with an IC50 of 0.45 ± 0.09 μM. The steady-state inactivation relationship curve is shifted to more negative potentials for the calcium channels, suggesting that ZC88 blocks inactivated state of the channel. ZC8

The application of bioactive compounds from the food industry to control mold growth in indoor waterborne coatings.

Microbial growth in indoor environments creates health problems, especially in people with asthma; approximately 80% of these patients are allergic to mold. Antimicrobial coatings are formulated to generate surfaces that are easy to clean and may also incorporate active agents, commonly called biocides, which inhibit microbial colonization, subsequent growth and bio-deterioration of the substrates. Some research lines seek to replace traditional organometallic and organochlorines biocides with environmentally acceptable ones. The aim of this research was, primarily, to explore the possible app

The CN7(-) anion.

Several salts containing the CN7(-) anion were prepared by deprotonation of 5-azido-1H-tetrazole (1). The highly explosive compounds hydrazinium (2), ammonium (3), aminoguanidinium (4), guanidinium (5), lithium (6), sodium (7), potassium (8), cesium (9), and calcium (10) azidotetrazolate were characterized by low temperature single X-ray diffraction. Other analytic methods, such as IR and Raman spectroscopy, multinuclear NMR spectroscopy, mass spectrometry, and differential scanning calorimetry were performed when possible since some of the metal salts explode spontaneously. 2 contains the hig

Fate of major compounds in source-separated urine.

Urine separation is a promising alternative to present-day waste water management. It can help to manage our nutrient flows in a sustainable way. Currently, techniques are being developed to recycle and treat source-separated urine. These techniques, however, must consider the spontaneous processes that change the separated urine. The initial cause of changes is the contamination with microorganisms, which can hardly be avoided in urine-collecting systems. The most important transformation processes are microbial urea hydrolysis, mineral precipitation and ammonia volatilisation. Additionally,

Clinical dentin hypersensitivity: understanding the causes and prescribing a treatment.

Dentin hypersensitivity is a common condition of transient tooth pain associated with a variety of exogenous stimuli. There is substantial variation in the response to such stimuli from one person to another. Except for sensitivity associated with tooth bleaching or other tooth pathology, the clinical cause of dentin hypersensitivity is exposed dentinal tubules as a result of gingival recession and subsequent loss of cementum on root surfaces. The most widely accepted theory of how the pain occurs is Brännström's hydrodynamic theory of dentin hypersensitivity. Dentinal hypersensitivity must be
