Pesticides are extensively used to control pests, diseases, and weeds in order to increase agricultural production. Usage of indiscriminate doses and persistent pesticides has not only caused resistance issues in insect pests but has also had deleterious effects on non-target organisms (beneficial insects, fish, and wildlife) and caused environmental contamination (soil, water, and air) through leaching, overflow, and insecticide spray drift. Exposure from eating food and drinking water contaminated to pesticide residues is also affecting human health. This study was conducted to obtain inform

BACKGROUND: Leaf-cutting ants (LCAs) of the genera Atta and Acromyrmex (Hymenoptera: Formicidae) are important pests of forest plantations, agriculture and livestock. Toxic baits containing the active ingredients fipronil or sulfluramid are the main method used to control LCAs. Insecticide dispersion among members of an LCA colony during control with toxic bait is not well understood. The objective of the study was to determine whether self-grooming, allogrooming or touching behavior among Atta sexdens (Hymenoptera: Formicidae) workers disperses the insecticides fipronil and sulfluramid among

INTRODUCTION: Global rise in obesity and metabolic syndrome has increased incidence of metabolic dysfunction-associated steatotic liver disease (MASLD). Owing to its high prevalence and the emergence of new therapies, noninvasive tests (NITs) for liver fibrosis are becoming more widely used.
OBJECTIVES: The aim of this study was to analyze and compare NIT performance in identifying fibrosis of grade F2 or greater in patients with MASLD confirmed on liver biopsy.
PATIENTS AND METHODS: This study was a retrospective analysis of 134 patients. The accuracy of 9 NITs in identifying significant fi

WHAT IS KNOWN AND OBJECTIVE: Primary myelofibrosis (PMF) is characterized by myeloid cell proliferation and prominent bone marrow fibrosis. Ruxolitinib, a selective inhibitor of JAK 1 and 2, significantly reduces constitutional symptoms and spleen size compared with placebo, and has significant clinical benefits in patients with myelofibrosis. The most common haematological side effects are thrombocytopenia and anaemia, and the most common non-haematological side effects are grade 1-2 diarrhoea and pyrexia. Leukocytoclastic vasculitis is small vessel vasculitis, characterized histopathological

Ruxolitinib (RUX) is a kinase inhibitor used in the treatment of various medical conditions and its mechanism of action involves suppression of the immune system. While beneficial in treatment of polycythemia vera, myelofibrosis and other indications, it can also increase a patient's susceptibility to various infections, including bacterial, viral and fungal. We present a case of a patient being treated with RUX who presented with a disseminated fungal infection. This case emphasises the need for vigilance of endemic fungal infections in individuals who are on RUX therapy.

In western North America, sylvatic plague (a flea-borne disease) poses a significant risk to endangered black-footed ferrets (Mustela nigripes) and their primary prey, prairie dogs (Cynomys spp.). Pulicides (flea-killing agents) can be used to suppress fleas and thereby manage plague. In South Dakota, US, we tested edible "FipBit" pellets, each containing 0.84 mg fipronil, on free-living black-tailed prairie dogs (Cynomys ludivicianus). FipBits were applied along transects at 125 per ha and nearly eliminated fleas for 2 mo. From 9-14 mo post-treatment, we found only 10 fleas on FipBit sites ve

Fipronil and emamectin benzoate (EMB) are effective insecticides for controlling cotton leafworm, Spodoptera littoralis. Fipronil works by blocking gamma-amino butyric acid (GABA) gated chloride. In contrast, EMB is activating GABA transporters. The objectives of our study were to assess relative toxicity of the technical fipronil and EMB alone and in mixture against S. littoralis. In addition, the GABA content was simultaneously determined using HPLC. Technical fipronil and EMB and their mixtures were applied topically to the fourth-instar larvae, and their LD50 values were estimated after 48

The sonidegib and ruxolitinib combination was assessed in an open-label study in JAK inhibitor-naive patients with myelofibrosis (MF). The primary objective of phase 1b was to establish the maximum tolerated dose (MTD) and/or recommended phase 2 dose (RP2D) and phase 2 was to assess spleen volume reduction at weeks 24 and 48. Fifty patients were enrolled. In the dose-escalation phase (n = 23), doses for sonidegib once daily/ruxolitinib twice daily were 400/10 mg (level 1, n = 8), 400/15 mg (level 2, n = 10), and 400/20 mg (level 3, n = 5). Two patients had dose-limiting toxicity at level 2: in

This study was conducted to investigate the toxic effects of fipronil on common carp (Cyprinus carpio). The trial was performed in laboratory of Department of Life Sciences at the Islamia University of Bahawalpur, Punjab, Pakistan. Total of 72 fishes, with apparently no clinical signs were purchased from local fish hatchery near Bahawalpur, Punjab, Pakistan. After 2 weeks of acclimatization fish were divided into six groups in glass aquaria (each having 100 liters of water capacity) and treated with different concentrations of fipronil for 12 days: 0 mg/L-control group (A),0.02 mg/L (B),0.04 m

In this review, we present a summary of how computer modeling has been used in the development of covalent-modifier drugs. Covalent-modifier drugs bind by forming a chemical bond with their target. This covalent binding can improve the selectivity of the drug for a target with complementary reactivity and result in increased binding affinities due to the strength of the covalent bond formed. In some cases, this results in irreversible inhibition of the target, but some targeted covalent inhibitor (TCI) drugs bind covalently but reversibly. Computer modeling is widely used in drug discovery, bu
