What is the role of PFTs in monitoring adverse effects of surgery, drug treatments, radiation therapy, and during hospitalization?

Measurement of various aspects of pulmonary function is a relatively easy, noninvasive, and inexpensive way to gauge the status of the respiratory system. Interest in using these tests to determine risk from medical and surgical interventions stems from their presumed ability to be more sensitive than history or physical examination in detecting underlying lung disease. When coupled with the assumption that early detection of pulmonary abnormalities will lead to alterations in patient management, the ultimate goal of improved patient outcomes becomes very attractive. However, despite advances

Protective effects of metallothionein I and II against metal- and ultraviolet radiation-induced damage in cultured lens epithelial cells.

PURPOSE: Few quantitative analyses of the induction of metallothioneins (MTs) have been conducted, and there are no reports on the contribution of MTs to the protective mechanism against ultraviolet (UV) radiation in the lens. In this study, we quantitatively analyzed the induction of MTs and analyzed the resulting protective effects against both metal- and UV radiation-induced damage in the cultured lens epithelial cell line, alphaTN4-1. METHODS: The induction profiles of MTs by ZnCl(2) treatment in alphaTN4-1 cells were analyzed by quantitative real-time reverse transcription polymerase cha

Cover-up of the effects of internal exposure by residual radiation from the atomic bombing of Hiroshima and Nagasaki.

The criteria certifying atomic bomb disease adopted by the Japanese government are very different from the actual state of the survivors. The criteria are based on epidemiological research by the Radiation Effects Research Foundation, the successor to the Atomic Bomb Casualty Commission (ABCC). The ABCC studied only the effects of primary radiation from the atomic bombing on the survivors of Hiroshima and Nagasaki, and ignored the damage from residual radiation. Analysis of the incidence of acute radiation disease, the rate of chromosomal aberrations, and the relative risks of chronic disease …

Effects of cranial radiation in children with high risk T cell acute lymphoblastic leukemia: a Pediatric Oncology Group report.

Contemporary chemotherapy has significantly improved event-free survival among patients with T cell-lineage acute lymphoblastic leukemia (T-ALL). Unlike B-precursor ALL, most investigators are still using cranial radiation (CRT) and are hesitant to rely solely on intrathecal therapy for T-ALL. In this study we assessed the effects of CRT upon event-free survival and central nervous system (CNS) relapses in a cohort of children with high risk features of T cell leukemia. In a series of six consecutive studies (1987-1995) patients were non-randomly assigned their CNS prophylaxis per individual p

Nuclear and cell membrane effects contribute independently to the induction of apoptosis in human cells exposed to UVB radiation.

UVB-induced DNA damage is a crucial event in UVB-mediated apoptosis. On the other hand, UVB directly activates death receptors on the cell surface including CD95, implying that UVB-induced apoptosis can be initiated at the cell membrane through death receptor clustering. This study was performed to measure the relative contribution of nuclear and membrane effects in UVB-induced apoptosis of the human epithelial cell line HeLa. UVB-mediated DNA damage can be reduced by treating cells with liposomes containing the repair enzyme photolyase followed by exposure to photoreactivating light. Addition

Effects of ionizing radiation and beta-adrenergic stimulation on the expression of early response genes in rat parotid glands.

There is little known about the regulation of gene expression in rat parotid glands after exposure to ionizing radiation. The present studies investigate the effects of in vivo ionizing radiation, with subsequent stimulation of beta-adrenergic receptors by isoproterenol, on parotid gland function and on the expression of the early response genes, c-fos, c-jun, and jun B. Ionizing radiation diminished parotid gland weight and saliva output. Treatment of irradiated rats with isoproterenol increased the gland weight to levels similar to those in nonirradiated rats. However, such treatment had no

Comparison of cellular and whole-animal bioassays for estimation of radiation effects in the polychaete worm Neanthes arenaceodentata (Polychaeta).

The polychaete worm Neanthes arenaceodentata was used in experiments to determine possible relationships between short-term genotoxicity tests and reproductive and lethal consequences of exposure to ionizing radiation. Groups of juvenile N. arenaceodentata received one of four different radiation doses (2, 4, 8, and 16 Gy) to determine dose-effect estimates for chromosomal aberration induction, and groups of both adult and juveniles received one of seven different radiation doses (1, 4, 8.4, 46, 102, 500, and 1000 Gy) to determine dose-effect estimates for reproduction, mortality, and life spa

Alleviation of the Genotoxic Effect of Experimental Circulatory Brain Hypoxia in Rat Cornea under the Effect of Infrared Low-Level Laser Radiation.

Experimental modeling of oxidative stress was conducted by creating circulatory brain hypoxia, accompanied by development of free radical processes leading to genome instability through an example of anterior epithelial layer of rat cornea. We studied the influence of infrared low-level laser radiation (ILLLR) in the therapeutic dose and mode (pulse power 8 W, λ=0.89 μm). Exposure of animals with circulatory brain hypoxia (ischemia) to ILLLR reduced the level of chromosome aberrations in corneal epitheliocytes and led to change in the mitotic index. Correction of the damaging effects of ischem

Re-Examining the Applicability of the Linear No-Threshold (LNT) Theory in Radiation Protection for Very Low Doses: Does a Threshold Exist? The Carcinogenic Effect of Radiation and the Possibility of a Dose Threshold.

Integrating recent biological and epidemiological insights, this analysis re-examines the Linear No-Threshold (LNT) model-the current basis for radiological protection-which assumes that cancer risk increases proportionally with any dose of ionizing radiation (IR). Empirical data from a cross-sectional study of 5156 radiation workers indicated a lower frequency of micronuclei in lymphocytes at very low cumulative doses (< 1 mSv) compared to a pre-employment control group. This observation is suggestive of, but does not directly prove, an adaptive cellular response; importantly, micronuclei are

Evofosfamide Is Effective against Pediatric Aggressive Glioma Cell Lines in Hypoxic Conditions and Potentiates the Effect of Cytotoxic Chemotherapy and Ionizing Radiations.

Hypoxia is a hallmark of many solid tumors and is associated with resistance to anticancer treatments. Hypoxia-activated prodrugs (HAPs) were developed to target the hypoxic regions of these tumors. Among 2nd generation HAPs, Evofosfamide (Evo, also known as TH-302) exhibits preclinical and clinical activities against adult glioblastoma. In this study, we evaluated its potential in the field of pediatric neuro-oncology. We assessed the efficacy of Evo in vitro as a single drug, or in combination with SN38, doxorubicin, and etoposide, against three pediatric high-grade glioma (pHGG) and three d

Effect of HfO2 incorporation on the radiation shielding performance of cement mortars under 6 MV photon irradiation.

Improving the radiation shielding performance of cement-based materials is of increasing importance for medical and nuclear facilities exposed to high-energy photon radiation. In this study, the effect of hafnium oxide (HfO2) incorporation on the radiation shielding performance of cement mortars was experimentally and theoretically investigated under clinically relevant 6 MV photon irradiation. Mortar specimens were prepared by partially replacing standard sand with HfO2 at replacement levels of 5 and 10 wt% and irradiated using a medical linear accelerator (LINAC). The linear attenuation coef

Electron Beam Crosslinking as a Strategy for the Synthesis of Pectin/PVA Aerogels: Effect of Concentration and Radiation Dose.

Radiation techniques represent an innovative and alternative method for polymer gelation or crosslinking; their application in aerogel research has not been extensively investigated. The irradiation of polymer solutions, either with or without a crosslinking agent, may result in stable polymer gels with a highly developed mesoporous structure. In this study, the effect of the pectin polysaccharide concentration, the presence of PVA, and the dose of ionizing radiation (27-58 kGy) were systematically investigated, particularly with respect to the structural, textural, and mechanical properties.
