Mutations in the vasopressin prohormone involved in diabetes insipidus impair endoplasmic reticulum export but not sorting.

Familial neurohypophysial diabetes insipidus is characterized by vasopressin deficiency caused by heterozygous expression of a mutated vasop … H sensitivity revealed these defects to be due to retention of mutant prohormone in the endoplasmic reticulum. Mutant prohormones that partially passed the endoplasmic reticulum were normally targeted to the regulated secretory pathway. Surprisingly, this also included mutants with mutations in residues involved in binding of vasopressin to neurophysin, a p …

Immunoisolation and characterization of a subdomain of the endoplasmic reticulum that concentrates proteins involved in COPII vesicle biogenesis.

Rubella virus E1 glycoprotein normally complexes with E2 in the endoplasmic reticulum (ER) to form a heterodimer that is transported to and retained in the Golgi complex. In a previous study, we showed that in the absence of E2, unassembled E1 subunits accumulate in a tubular pre-Golgi compartment whose morphology and biochemical properties are distinct from both rough ER and Golgi. We hypothesized that this compartment corresponds to hypertrophied ER exit sites that have expanded in response to overexpression of E1. In the present study we constructed BHK cells stably expressing E1 protein co…

Role of different types of Ca2+ channels and a reticulum-like Ca2+ pump in neurotransmitter release.

The factors controlling the Ca2+ concentration directly responsible for triggering acetylcholine (ACh) release were investigated at an ident … esynaptic action potential by controlling the Ca(2+)-dependent K+ current. tBuBHQ, a blocker of the reticulum Ca2+ pump, induces a potentiation of evoked release without modifying the presynaptic Ca2+ influx. This seems to indicate that a part of the Ca2+ entering the presynaptic terminal through N- and P-type Ca2+ channels is sequestered in a presynaptic r …

Different concentrations of thyroid peroxidase and thyroglobulin in the nuclear envelope and the endoplasmic reticulum throughout the cytoplasm.

Thyroid peroxidase (TPO) and thyroglobulin (TG) represent two major glycoproteins of thyroid follicular cells performing biological function … sections and the immunogold technique. It is documented that TG is transported from the endoplasmic reticulum and the Golgi apparatus to the follicular lumen by transport vesicles; most of it being present in the expanded endoplasmic reticulum throughout the cytoplasm. On the other hand, gold particles indicating TPO are adjacent to the membranes of the exo …

Membrane properties of the sarcolemma and sarcoplasmic reticulum of pigs susceptible to malignant hyperthermia. Action of halothane.

The role of sarcolemma and sarcoplasmic reticulum in malignant hyperthermia was studied by the esr technique using the trapezius muscle membrane of both normal and genetically susceptible pigs. Normal and malignant hyperthermia membranes from sarcolemma as well as from sarcoplasmic reticulum did not show significant differences near the polar heads of the phospholipidic bilayer. In contrast, the fluidity and activation energy of normal membranes differed from those in malignant hyperthermia; in both sarcolemma and sarcoplasmic reticulum the mobility of the label was greater than the controls. …

ATP-dependent interaction of propranolol and local anaesthetic with sarcoplasmic reticulum. Stimulation of Ca2+ efflux.

Preincubation of sarcoplasmic reticulum (SR) with propranolol or tetracaine inhibits Ca2+ accumulation and stimulates ATPase activity by more than 2-fold. This effect is obtained only when the preincubation is carried out in the presence of ATP or other nucleoside triphosphates. The (ATP + drug)-induced inhibition of Ca2+ accumulation is pH-dependent, increasing as the pH rises above 7.5. The presence of micromolar concentrations of Ca2+ or Mg2+ during the preincubation prevents the inhibitory effect of ATP plus drug on Ca2+ accumulation or ATPase activity. The (ATP + drug) modification of SR …

Effects of calcitonin and CGRP alone or in combination on food intake and forestomach (reticulum) motility in sheep.

The effects of intracerebroventricular (ICV) and intravenous (IV) administration of calcitonin and calcitonin gene-related peptide (CGRP) on feeding behavior and reticular motility were investigated in sheep. ICV calcitonin at a dose of 2 to 200 mU/kg reduced, in a dose-related manner, the immediate (0-60 min) food intake. The daily food intake was also significantly (p less than 0.05) decreased for doses up to 20 mU/kg, and the frequency of reticular contractions during the first hour of eating was decreased by 27.9%. Calcitonin at the highest IV dose (200 mU/kg) did not affect feeding behavi

The effects of deoxycholate and trypsin on the cross-linking of rabbit skeletal muscle sarcoplasmic reticulum proteins.

Dithiobis (succinimidyl propionate) has been used to cross-link sarcoplasmic reticulum microsome proteins. Although the 100,000 dalton calcium stimulated ATPase and the 60,000 dalton calcium-binding protein calsequestrin were readily cross-linked to form homopolymers, no heteropolymer formation between these two proteins were detected. The 90,000 dalton protein A1 which is always observed in our preparations appeared to preferrentially form dimers on cross-linking. When calsequestrin was solubilized using 0.1 mg deoxycholate/mg protein, this protein was not cross-linked even at dithiobis(succi…

Endoplasmic reticulum stress in autoimmune skin diseases: immune activation, cutaneous cell damage, and therapeutic advances.

Endoplasmic reticulum stress (ERS) arises when the protein-folding load, or perturbations in calcium and redox homeostasis, outpace the adaptive capacity of the endoplasmic reticulum (ER). The unfolded protein response (UPR) initially reduces secretory load and restores proteostasis. Persistent stress can redirect the same pathways toward inflammatory signaling, altered antigen processing, and potentially immunogenic cell death. We term this context-dependent transition ERS-immune tolerance breaking, and propose it as a unifying framework for understanding how unresolved ERS may contribute to …

Endoplasmic Reticulum Stress Drives Carotid Atherosclerotic Plaque Instability: Integrative Insights into Biomarkers and Therapeutic Targets.

OBJECTIVE: Carotid atherosclerosis is a leading cause of stroke, with plaque rupture closely linked to endoplasmic reticulum stress (ERS). However, the precise mechanisms of ERS in plaque instability remain incompletely understood. This study aims to investigate the role of ERS in carotid plaque instability, identify potential biomarkers, and screen candidate therapeutic drugs. METHODS: Transcriptomic data from the Gene Expression Omnibus (GEO) database (GSE43292 and GSE163154) were analyzed using differential expression analysis, weighted gene co-expression network analysis (WGCNA), and mach…

ERX 208 induces endoplasmic reticulum stress and suppresses DNA damage repair to enhance chemotherapy sensitivity.

Ovarian cancer (OCa) remains the most lethal gynecologic malignancy, driven in part by intrinsic and acquired therapeutic resistance to DNA damaging chemotherapies. We previously identified that ERX-208 targets the protein encoded by Lysosomal acid lipase (LIPA) to induce catastrophic endoplasmic reticulum (ER) stress in OCa models. As ERX-class compounds advance to clinical development, rational combination strategies are needed to maximize therapeutic efficacy. To address this question, we conducted an unbiased in vitro screen of the combination of ERX 208 with 147 FDA approved agents. We id…

Mitochondria-associated endoplasmic reticulum membranes facilitate gastric epithelial injury in hypoxia-induced gastric mucosal lesions.

INTRODUCTION: Hypoxia following mucosal congestion in clinical practice causes a wide range of gastrointestinal symptoms and induces profound structural and functional perturbations in mitochondria-associated ER membranes (MAMs), which are subcellular compartments whose integrity and stability are regulated by FUN14 domain-containing protein 1 (FUNDC1). OBJECTIVES: To investigate the effect of hypoxia-induced MAMs formation on gastric mucosal lesions in patients with severe hepatic disorders. METHODS: Liver cirrhosis (LC) and hepatic failure (HF) were selected as representative severe hepati

Mechanisms and therapeutic strategies targeting the reversible phosphorylation-endoplasmic reticulum stress network in diabetic cardiomyopathy.

The pathological progression of diabetic cardiomyopathy (DCM) involves multiple interconnected mechanisms, including metabolic reprogramming, oxidative stress, Ca2+ dysregulation, and programmed cell death. Recent studies have shown that reversible protein phosphorylation, endoplasmic reticulum stress (ERS) and the unfolded protein response (UPR) are not independent events, but rather form an interactive network through the dynamic balance mediated by kinases and phosphatases, thereby coordinating cardiomyocyte fate. This review focuses on the reciprocal regulation of reversible phosphorylatio…

Endoplasmic Reticulum Stress-Related Immune Signature in Atrial Fibrillation: Machine Learning and Single-Cell Transcriptomics.

This study describes a reproducible computational workflow for identifying endoplasmic reticulum stress (ERS)-related gene signatures in atrial fibrillation (AF) by integrating bulk transcriptomics, machine learning, immune infiltration analysis, and single-cell transcriptomics. Public bulk transcriptomic datasets were retrieved from the Gene Expression Omnibus (GEO), followed by phenotype harmonization, normalization, batch-effect correction, and differential expression analysis. Weighted gene co-expression network analysis (WGCNA) was combined with ERS-related gene sets to identify candidate…

Regulation of Endoplasmic Reticulum Stress Suppresses Early Pro-Tumorigenic Events During N-Nitrosodiethylamine-Induced Hepatocarcinogenesis.

Carcinogenesis is a dynamic, multistep process in which the initiation stage represents a critical and irreversible event that determines cellular fate. Although genetic alterations play a central role in tumor development, dysregulation of intracellular organelles such as the endoplasmic reticulum (ER) has emerged as an important contributor to cancer initiation. In the present study, we aimed to evaluate how modulation of ER stress affects the initiation phase of hepatocarcinogenesis. The initiation-stage hepatocarcinogenic model was developed by administering a single dose of N-nitrosodieth…
