The peptidomimetic Vasotide targets two retinal VEGF receptors and reduces pathological angiogenesis in murine and nonhuman primate models of retinal disease.

Blood vessel growth from preexisting vessels (angiogenesis) underlies many severe diseases including major blinding retinal diseases such as retinopathy of prematurity (ROP) and aged macular degeneration (AMD). This observation has driven development of antibody inhibitors that block a central factor in AMD, vascular endothelial growth factor (VEGF), from binding to its receptors VEGFR-1 and mainly VEGFR-2. However, some patients are insensitive to current anti-VEGF drugs or develop resistance, and the required repeated intravitreal injection of these large molecules is costly and clinically p

Retinal vasculitis in rheumatic diseases: an unseen burden.

Retinal vascular inflammation, a potentially blinding condition (herein: retinal vasculitis (RV)) is commonly associated with a heterogeneous group of diseases characterized by systemic inflammatory cell infiltration and/or necrosis of blood vessel walls. RV may arise as an isolated ocular disorder, as part of systemic vasculitis (Wegener's granulomatosis and Adamantiadis-Behcet Disease), or it can be secondary to an underlying connective tissue disease (systemic lupus erythematosus, sarcoidosis, and rheumatoid arthritis), systemic infection, or malignancy. Depending on the type of RV, it can

Improved method for optical coherence tomography angiography: from reconstruction to clinical indicator quantification.

SIGNIFICANCE: Optical coherence tomography angiography (OCTA) represents a significant advance in noninvasive ophthalmic vascular imaging, yet existing reconstruction algorithms face challenges such as spectral leakage and motion artifacts, which can compromise image quality and the accuracy of subsequent clinical quantification. Improving OCTA reconstruction and developing robust, automated methods for clinical indicator extraction are crucial for enhancing diagnostic reliability and facilitating precise disease monitoring. AIM: We aim to propose and validate an improved OCTA reconstruction

Association of Remnant Cholesterol With the Retinal Capillary Plexus.

PURPOSE: Remnant cholesterol (RC) is a well-recognized risk factor for cardiovascular diseases (CVDs) and an important biomarker of cardiovascular risk. However, studies on the effects of RC on the early retinal microvasculature are still scarce. Therefore, this study aimed to investigate the association between RC and the retinal capillary plexus (RCP) using optical coherence tomography angiography (OCTA). METHODS: This cross-sectional study evaluated data from participants of the Jidong Eye Cohort Study. All participants underwent comprehensive ophthalmic examinations and laboratory tests.

Topographic analysis of retinal and choroidal microvasculature according to diabetic retinopathy severity using optical coherence tomography angiography.

PURPOSE: To investigate the topographic changes in the retinal capillary plexus and the choriocapillaris according to the severity of diabetic retinopathy (DR) using optical coherence tomography angiography (OCTA). METHODS: Subjects were recruited and classified into one of the following four groups: normal controls (n = 52), diabetes without DR (n = 49), non-proliferative DR (n = 51) and proliferative DR (n = 38). Using OCTA, the superficial capillary plexus (SCP), deep capillary plexus (DCP) and the choriocapillaris vessel densities were measured and compared in different macular areas: the

Detection of glaucomatous change based on vessel shape analysis.

Glaucoma, a leading cause of blindness worldwide, is a progressive optic neuropathy with characteristic structural changes in the optic nerve head and concomitant visual field defects. Ocular hypertension (i.e. elevated intraocular pressure without glaucoma) is the most important risk factor to develop glaucoma. Even though a number of variables, including various optic disc and visual field parameters, have been used in order to identify early glaucomatous damage, there is a need for computer-based methods that can detect early glaucomatous progression so that treatment to prevent further pro

Autoregulation, a balancing act between supply and demand.

Regulation of blood flow is necessary to adapt to different conditions. Regulation of ocular blood flow (OBF) compensates for varying perfusion pressures (autoregulation), adapts to the retinal activity (neurovascular coupling), and keeps the back of the eye at constant temperature (thermoregulation). While all vessels are under the control of the vascular endothelial cells, the retinal vessels are additionally under the control of the neural and glial cells, and the choroidal vessels are influenced by the autonomic nervous system. The optic nerve head is additionally controlled by circulating…

A novel method of quantifying retinal vascular tortuosity in retinopathy of prematurity.

This study evaluated the diagnostic performance of a novel vessel tracing software that quantifies retinal vessel tortuosity in premature infants for assessing retinopathy of prematurity (ROP). Seventy-three retinal images of premature infants were subjectively classified as having "Normal" or "Abnormal" tortuosity based on arterial tortuosity of the posterior retina by seven clinicians. The software then semi-automatically traced the same arteries within circular regions of interest centered at the optic disc, with radii ranging from 2 to 20x the disc diameters. Vessel tortuosity was defined

The role of microglia in the development of diabetic retinopathy and its potential clinical application.

Lately, research on the function of microglia in diabetic retinopathy (DR) is becoming increasingly focused. Microglia are immune cells that dwell in the central nervous system and are crucial to the pathophysiology of DR. According to studies, a hyperglycemic environment can activate microglia, bringing them out of a resting state to an active state. This allows them to release a variety of inflammatory factors and chemokines, which can then cause retinal inflammatory reactions. When it comes to angiogenesis in DR, activated microglia release a variety of angiogenic substances, such as vascul

The Natural History of Retinal Vascular Changes from Infancy to Adulthood in Wyburn-Mason Syndrome.

Wyburn-Mason syndrome is a rare, non-hereditary congenital neurocutaneous disorder leading to arteriovenous malformations. Malformations are characterized by an artery that is directly connected to veins without a capillary system and forms a fragile mass of abnormal vessels. It can be found in the midbrain, in the eyes, orbit, and rarely in cutaneous nevi. Neurological and ocular symptoms are the most common. Ocular signs and symptoms include abnormally dilatated vessels of conjunctiva, nystagmus, strabismus, vitreous hemorrhage, vein occlusions, retinal detachment, etc. Neurological symptoms

Hydranencephaly in an infant with vascular malformations.

Hydranencephaly is a condition in which cerebral hemispheres are absent and reduced to fluid-filled sacs in a normal skull. Numerous causes have been proposed. We report a male infant with hydranencephaly and congenital vascular malformations (port wine stains, generalized nevus flammeus, anomalous retinal vessels, and absent internal carotid flow). Magnetic resonance imaging of the brain showed absence of most of the cerebrum except for small portions of the occipital cortex and thalami. Magnetic resonance angiography showed flow within the vertebral and basilar arteries without internal caro…

Amaurosis fugax: a clinical comparison.

Amaurosis fugax (transient monocular blindness) is a symptom of retinal ischemia just as contralateral hemiparesis and sensory loss are symptoms of cerebral ischemia. These symptoms are produced by atherosclerotic stenosis of the carotid vessels at the ipsilateral carotid bifurcation and emboli from these areas causing focal, repetitive, retinal ischemia. A study of 31 endarterectomy patients was undertaken to see if eight patients with amaurosis fugax (25%) could be differentiated from 22 patients with transient cerebral ischemia. The patients with amaurosis fugax were found to be younger. Th

Retinal lesions experimentally produced by intravitreal ultrasound.

Currently, vitreous surgical techniques combined with phakoemulsification are an established procedure for the removal of luxated natural lenses or dislocated lenticular material. This has proven to be a safe procedure, but occasionally retinal lesions have been inadvertently created. We describe the ophthalmoscopic and morphologic features of ultrasonically induced retinal lesions in rabbit eyes using a similar technique. The retina was treated directly using a conventional ultrasound tip for pars plana lensectomy (Fragmatome). Ophthalmoscopically slight lesions corresponded to an area of ret
