Sex differences in sleep fragmentation in 5xFAD mice.

Sleep alterations have long been associated with Alzheimer's disease (AD), but whether it is an early symptom or only develops later in the pathological progression remains unknown. To study this, 5xFAD heterozygous (Het) mice, a transgenic model of amyloid overexpression, and wild-type (WT) littermates at 1, 2, 3, 4 and 6 months of age were assessed within instrumented home cages to noninvasively score 3-state sleep using respirations and gross body movements during the dark cycle. Progressive alterations in dark cycle sleep architecture were identified in Het animals as early as 1 month of a

Diagnostic value of sleep stage dissociation as visualized on a 2-dimensional sleep state space in human narcolepsy.

BACKGROUND: Type 1 narcolepsy (NT1) is characterized by symptoms believed to represent Rapid Eye Movement (REM) sleep stage dissociations, occurrences where features of wake and REM sleep are intermingled, resulting in a mixed state. We hypothesized that sleep stage dissociations can be objectively detected through the analysis of nocturnal Polysomnography (PSG) data, and that those affecting REM sleep can be used as a diagnostic feature for narcolepsy. NEW METHOD: A Linear Discriminant Analysis (LDA) model using 38 features extracted from EOG, EMG and EEG was used in control subjects to sele

Short-term use of fluoxetine in asymptomatic obese subjects with sleep-related hypoventilation.

Disordered nocturnal breathing with significant arterial oxygen desaturation and sleep apnoea is a feature of extreme obesity which is often difficult to manage in the short term. We have evaluated the effect of fluoxetine, a centrally acting 5-HT re-uptake inhibitor, on sleep-breathing patterns in asymptomatic extremely obese subjects. A double-blind cross-over study was used to compare fluoxetine (60 mg for three days) to placebo. Eleven obese subjects (ten males, one female, mean weight +/- s.d. 131 +/- 2 kg) slept overnight in a sleep laboratory with the polysomnographic study recorded aft

Sleeping brain oscillates with intensity-induced auditory rhythm.

The rhythmic patterns embedded in auditory stimuli hold considerable significance, as individuals often exhibit a subconscious tendency to synchronize bodily movements with perceived rhythms during wakefulness. However, the extent to which the sleeping brain can discern specific acoustic attributes, such as sound intensity, and entrain to intensity-induced rhythms remains largely unexplored. Here, we employed a frequency-tagging paradigm with electroencephalographic (EEG) and magnetoencephalographic (MEG) simultaneous recording to investigate whether the sleeping brain could perceive the inten

Movements and behaviors during spontaneous arousals in healthy young adults: an intermediary stage between wakefulness and sleep?

BACKGROUND: Arousals are common, sudden and transient elevations of the vigilance level during normal sleep, but arousal-associated behaviors have not yet been studied. OBJECTIVE: We aimed to describe the duration as well as motor and autonomic patterns associated with arousals across sleep stages in normal subjects. METHODS: The spontaneous arousals of 25 healthy young adults were randomly analyzed on polysomnography with body- and face-oriented video cameras. The duration of the heart rate response as well as the frequency, amplitude, speed, body segment and semiology of associated movemen

Posttraumatic stress disorder diagnosis is associated with reduced parasympathetic activity during sleep in US veterans and military service members of the Iraq and Afghanistan wars.

STUDY OBJECTIVES: To determine whether high-frequency heart rate variability (HF-HRV) during sleep differs between those with and without posttraumatic stress disorder (PTSD) as a function of sleep type (non-rapid eye movement [NREM] vs. rapid eye movement [REM]), and to explore this relationship across successive sleep cycles. Participants with PTSD were hypothesized to have lower HF-HRV across both REM and NREM sleep. METHODS: Sixty-two post-9/11 military veterans and service members completed self-report measures of sleep quality, insomnia severity, and disruptive nocturnal behaviors. Part

Sleep architecture, cocaine and visual learning.

BACKGROUND: Disturbances in sleep associated with chronic cocaine use may underlie abstinence-related cognitive dysfunction. We hypothesized that sleep-related cognitive function would be impaired in chronic cocaine users, and that this impairment would be associated with abstinence-related changes in sleep architecture. METHODS: Twelve chronic cocaine users completed a 23-day in-patient study that included randomized, placebo-controlled, cocaine self-administration sessions. We report polysomnographic measurement of rapid eye-movement (REM) sleep and slow-wave activity, and performance on a

Cholinergic REM induction test with arecoline in depressed children.

Children with major depressive disorder often fail to exhibit electroencephalographic (EEG) sleep abnormalities similar to those reported in depressed adults. It was hypothesized that a cholinergic rapid eye movement (REM) induction test would contribute to the identification of EEG sleep abnormalities in depressed children. To test this hypothesis, prepubertal children meeting research diagnostic criteria for major depressive disorder (n = 33) and carefully screened healthy control children (n = 15) were enrolled in a 4-day psychobiologic protocol that included 1 night with infusion of arecol

Polysomnographic Parameters in Schizoaffective Disorder: A Systematic Review and Meta-Analysis.

BACKGROUND: Schizoaffective disorder (SZA) is a severe psychiatric condition characterized by overlapping affective and psychotic features; however, its sleep architecture remains poorly characterized. This systematic review and meta-analysis compared polysomnographic findings in drug-free patients with SZA with those of healthy controls (HC), patients with schizophrenia (SCZ), and patients with major depressive disorder (MDD). METHODS: An extensive database search identified 40 studies. Nine case-control studies including 79 patients with SZA, 88 patients with SCZ, 79 HC, and 131 patients wi

The effect of placebo administration on the first-night effect in healthy young volunteers.

The first-night effect is a well-known phenomenon that is considered to result from a subject's lack of adaptation to the unfamiliar environment of a sleep laboratory and to the technical equipment used for polysomnography. The effect has been explored as a laboratory model for transient insomnia. The main characteristics of this effect are short total sleep time (TST) and rapid eye movement (REM) sleep, a lower sleep efficiency index, and longer REM sleep latency. Previous studies have reported that personality traits (such as trait anxiety) are a potential cause of the first-night effect and

Sleep in cluster headache - beyond a temporal rapid eye movement relationship?

BACKGROUND AND PURPOSE: Cluster headache (CH) is a primary headache disorder characterized by severe attacks of unilateral pain following a chronobiological pattern. There is a close connection with sleep as most attacks occur during sleep. Hypothalamic involvement and a particular association with rapid eye movement (REM) sleep have been suggested. Sleep in a large, well-characterized population of CH patients was investigated. METHODS: Polysomnography (PSG) was performed on two nights in 40 CH patients during active bout and one night in 25 age, sex and body mass index matched controls in h

Advance taper of antidepressants prior to multiple sleep latency testing increases the number of sleep-onset rapid eye movement periods and reduces mean sleep latency.

STUDY OBJECTIVES: Patients presenting with excessive sleepiness are frequently using antidepressant medication(s). While practice parameters recommend discontinuation of antidepressants prior to multiple sleep latency testing (MSLT), data examining the impact of tapering these medications on MSLT results are limited. METHODS: Adult patients who underwent MSLT at Mayo Clinic Rochester, Minnesota, between 2014 and 2018 were included. Clinical and demographic characteristics, medications, including use of rapid eye movement-suppressing antidepressants (REMS-ADs) at assessment and during testing,

Clinical significance of periodic leg movements during sleep in rapid eye movement sleep behavior disorder.

The aim of the study was to explore the clinical significance of periodic leg movements during sleep (PLMS) in rapid eye movement sleep behavior disorder (RBD) and the pathological relation between these two disorders. Eighty-one consecutive idiopathic RBD (iRBD) patients, classified into two groups-27 patients with PLMS (iRBD-PLMS) and 54 patients without PLMS (iRBD w/o PLMS), and 31 patients with idiopathic PLMS (iPLMS)-were enrolled in this study. Descriptive variables including Epworth Sleepiness Scale (ESS) scores and polysomnography measures were compared among the three patient groups.

Pilot screening for prodromal Parkinson's disease in post-9/11 Veterans with probable rapid eye movement sleep behavior disorder.

This pilot examined prodromal markers in post-9/11 Veterans with TBI and probable REM sleep behavior disorder (N = 12). RBD was confirmed in 5 of 10 (50 %) participants with polysomnography. Veterans with subthreshold parkinsonism had lower olfactory scores. Future studies should quantify REM sleep without atonia and include broader markers.

Literature Cases Summarized Based on Their Polysomnographic Findings in Rett Syndrome.

Rett syndrome (RTT) is a severe and rare neurodevelopmental disorder affecting mostly girls. In RTT, an impaired sleep pattern is a supportive criterion for the diagnosis, yet little is known regarding the sleep structure and sleep respiratory events. Aiming to delineate sleep by aggregating RTT case (series) data from published polysomnographic studies, seventy-four RTT cases were collected from eleven studies up until 6 February 2022 (PROSPERO: CRD 42020198099). We compared the polysomnographic data within RTT stratifications and to a typically developing population. MECP2 cases demonstrated

[Physiology of normal sleep].

Normal sleep of a young adult is composed of around 4 to 6 cycles of 90 minutes each. Two states of sleep have been established, REM sleep and Non REM sleep. REM sleep is mainly characterized by a complete atonia which creates a physiological paralysis. Slow wave sleep is characterized by an increase of more than 20% of EEG delta waves. Sleep could be represented by a two process model. The first one, the homeostatic component, represents the propensity to sleep, the second one, the circadian component, symbolizes 24th light-dark cycle of the rotation of the earth. The homeostatic component is

Reversal of the sleep/wake cycle disorder of sleeping sickness after trypanosomicide treatment.

To determine whether the circadian disruption of the sleep/wake cycle observed in sleeping sickness, human African trypanosomiasis (HAT), can be reversed after trypanosomicide treatment, 10 Congolese patients infected by Trypanosoma brucei gambiense underwent 24-h polysomnographic recordings before treatment with melarsoprol and after each of three weekly treatment sessions. Polysomnography consisted of a continuous recording of the electroencephalogram, electromyogram and electro-oculogram on a Minidix Alvar polygraph. Sleep traces were analysed in 20-sec epochs for wakefulness, REM sleep, an

[Sleep-disordered breathing in long-term opioid therapy].

Chronic opioid use has been known to cause disturbed sleep as well as excessive daytime sleepiness. During induction and maintenance of opioid use there is a reduction of REM- and slow wave sleep. Central sleep apnea (CSA) has been reported in about 30 % of patients with chronic opioid use. Ataxic breathing and CSA are more prominent in NREM- than REM-sleep. CSA does not seem the sole cause of excessive daytime sleepiness in these patients. Further studies are necessary regarding the effects and consequences of chronic opioid use during sleep.

Melanin-concentrating hormone during lactation: a compensatory role for sleep deprivation?

We propose a novel hypothesis regarding the role of melanin-concentrating hormone (MCH) in the medial preoptic area (MPOA) during lactation and its implications for mothers' sleep patterns. We propose that MCH, with its transient expression in the MPOA during lactation, may function as a compensatory mechanism to mitigate the adverse effects of chronic sleep deprivation commonly experienced by lactating mothers. This compensatory role is hypothesized to involve regulating sleep architecture, particularly by promoting rapid eye movement (REM) sleep and modulating the balance between REM and non
